Tryptophan-producing bacteria to mitigate osteoporosis and intestinal dysfunction.

The relationship between gut microbiota and host health and disease is intricate, with microbiota-derived metabolites playing a crucial role in the gut-organ axis. In this study, we observe significantly decreased levels of microbial metabolites, particularly tryptophan derivatives in osteoporosis mice. Loss of tryptophan induced intestinal epithelial barrier dysfunction which compromised intestinal barrier integrity, leading to bone inflammatory responses and pathological osteoporosis. Through supplementation of tryptophan-producing bacteria, we effectively repair damaged intestinal barriers

Beneficial and detrimental consequences of AHR activation in intestinal infection.

The ligand dependent transcription factor aryl hydrocarbon receptor (AHR) is an environmental sensor whose activation can have physiologically beneficial or detrimental consequences for host immune responses depending on the ligand. Here we investigated the hypothesis that prolonged AHR activation either due to inefficient ligand metabolism or due to genetic manipulation may underlie the distinction between beneficial and detrimental effects. Our data indicate that prolonged AHR activation caused toxic endpoints for liver and thymus but was not per se interfering with the host response to infe

A Translational Model for In Vivo Intestinal Permeability Assessment in Pigs.

BACKGROUND: Several noncommunicable diseases in humans are associated with increases in intestinal permeability, which can be modulated by dietary factors. To investigate diet-induced changes in intestinal permeability, relevant translational models are required. Pigs are relevant models for nutrition research, gastrointestinal tract health, and intestinal permeability, but intestinal permeability assessment in pigs needs optimization. OBJECTIVES: The aim of this study was to develop and optimize protocols to induce and quantify intestinal permeability in pigs as a translational model. METHO

Intestinal Perforation Secondary to Bortezomib-Induced Autonomic Neuropathy.

It is essential to evaluate both the patient's prior conditions and the severity of the current clinical presentation when deciding on BTZ toxicity management. It seems prudent to consider the permanent discontinuation of the drug in patients who have experienced at least grade 3 intestinal neuropathy and have structural abnormalities or other risk factors for intestinal perforation. Trial Registration: NCT03710603.

Surgical Insight Into Paraduodenal Hernia Causing Intestinal Obstruction.

Paraduodenal hernia (PDH), also known as mesocolic hernia, is a rare internal hernia resulting from a congenital anomaly caused by improper retroperitoneal fixation of the mesentery due to abnormal midgut rotation. Although uncommon, internal hernias can cause acute intestinal obstruction and pose a life-threatening risk if not promptly diagnosed and managed. This case report presents a 25-year-old male with a two-month history of progressively worsening abdominal pain, exacerbated by food intake and accompanied by vomiting. Imaging via contrast-enhanced CT revealed a left PDH, with clustering

Live-Cell Calcium Imaging in 3D Intestinal Organoids.

Live-cell Ca2+ imaging is an important tool to detect activation of receptors by a putative ligand/drug and complements studies on transport processes, as intracellular Ca2+ changes provide direct evidence for substrate fluxes. Organoid-based systems offer numerous advantages over other in vitro systems such as cell lines, primary cells, or tissue explants, and in particular, intestinal organoid culture has revolutionized research on functional gastrointestinal processes. Calcium imaging using the fluorescent Ca2+ indicator Fura-2-AM can be applied to 3D intestinal organoids, which show an exc

Jejunal leiomyosarcoma leading to intestinal intussusception: a case report.

BACKGROUND: Leiomyosarcoma (LMS) is a rare form of malignant tumor affecting the small intestine, specifically the jejunum. It can cause intussusception, a condition wherein one segment of the intestine telescopes into another, leading to obstruction, ischemia, and necrosis. Small intestinal intussusception in adults is a rare clinical condition. The majority of affected individuals have pathological factors, such as tumors, polyps, diverticula, inflammation, or intestinal adhesions. Abdominal pain and bowel obstruction are the most common symptoms, but small bowel tumors are often not visible

Ileus Caused by Compression of the Small Intestine in 35 Cows.

Ileus attributable to compression of the small intestine describes extensive constriction of the intestine by nearby space-occupying lesions or other abdominal organs. This retrospective study describes the findings in 35 cows with ileus caused by CSI (compression of the small intestine) by adhesions, abscesses, and the gravid uterus. Colic occurred in 34.3% of the cows. Rumen motility was reduced or absent in all cows, and intestinal motility was decreased in 85.7%. Ballottement and/or percussion and simultaneous auscultation were positive on the right side in 73.5% of the cows. The rectum co

Atypical findings of small intestinal adenocarcinoma with mesenteric extension.

Primary small intestinal cancer is a rare malignancy, accounting for 0.1%-0.3% of all gastrointestinal cancers. Its nonspecific presentation often delays diagnosis and worsens prognosis. This case report presents a patient with small intestinal cancer exhibiting mesenteric invasion and atypical features. We emphasize that mesenteric invasion is a rare finding, and the achieved 5-year recurrence-free survival in a Stage ⅡB small intestinal adenocarcinoma represents an exceptionally favorable outcome. The patient was a 78-year-old female who was referred to our hospital with a chief complaint of

Small intestinal bacterial overgrowth: from malabsorption to misinterpretation.

Small intestinal bacterial overgrowth (SIBO) was originally described as a cause of maldigestion and malabsorption in situations where disruptions of intestinal anatomy or physiology favored the proliferation of bacteria normally confined to the colon. In this context, the pathogenesis of symptoms resulting from SIBO was well described. More recently, the concept of SIBO was extended to explain symptoms such as bloating, altered bowel habit and discomfort among individuals with irritable bowel syndrome and since then a whole host of gastrointestinal and extragastrointestinal disorders have bee

Intestinal flow and digestive parameters of Lutzomyia longipalpis larvae.

Lutzomyia longipalpis Lutz & Neiva, 1912 (Diptera, Psychodidae), is the primary vector of Leishmania infantum Nicole, 1908, the etiological agent of American visceral leishmaniasis. During their development, sandfly larvae pass through four instars, consuming soil particles enriched with microorganisms and decomposing organic material. In numerous insect species, the intestinal epithelium not only secretes digestive enzymes and absorbs digested nutrients but also carries out additional functions, such as regulating luminal pH and facilitating the absorption or secretion of ions and water. The interplay of gut microbiota and intestinal motility in gastrointestinal function.

The relationship between gut microbiota and intestinal motility is crucial for maintaining gastrointestinal health. Intestinal motility refers to the coordinated movements of the digestive tract, essential for effective digestion, nutrient absorption, and timely waste elimination. Recent studies have demonstrated that microbiota play a crucial role not only in the maturation of intestinal motility but also in the ongoing maintenance of established motility patterns. Disruptions in motility can lead to various disorders, such as chronic constipation, irritable bowel syndrome, and chronic idiopa

Mechanism of reduced intestinal phosphate absorption by tenapanor: a hypothesis.

Tenapanor, a selective inhibitor of the sodium/hydrogen exchanger isoform 3 (NHE3), was initially developed for the treatment of irritable bowel syndrome with constipation. Subsequent preclinical and clinical studies revealed its ability to reduce gastrointestinal phosphate absorption, leading to effective serum phosphate control with minimal pill burden in patients with kidney failure undergoing dialysis therapy. However, the precise mechanisms underlying NHE3 inhibition, its impact on phosphate handling and the primary site of action within the gastrointestinal tract remain incompletely unde

RELMβ in intestinal helminth infection: Mechanisms and research advances.

Intestinal helminth infections remain a major global public health challenge, affecting both human and animal health. The immune mechanisms by which hosts resist and expel helminths are multifaceted, with intestinal epithelial cells and their secretory products playing pivotal roles. Resistin-like molecule β (RELMβ), also known as Found in Inflammatory Zone 2 (FIZZ2), is a secreted protein predominantly produced by intestinal goblet cells. Recent studies have revealed that RELMβ exerts important functions in intestinal inflammation and immune regulation, particularly in type 2 immunity-driven

Intestinal tuberculosis as a diagnostic challenge in a high prevalence center.

The differential diagnosis between Crohn's disease (CD) and intestinal tuberculosis (ITB) continues to pose a diagnostic challenge, as clinical presentation can be indistinguishable, as well as findings in imaging studies and endoscopy. It is essential to make a definitive diagnosis early on, screening for possible latent infectious diseases, especially in patients from endemic areas, as inmmunosuppressive treatment can result in a fatal outcome during the course of TBI.
