Post-stroke gastrodin treatment ameliorates ischemic injury and increases neurogenesis and restores the Wnt/β-Catenin signaling in focal cerebral ischemia in mice.

Cerebral ischemic stroke is one of the leading causes of death and disability worldwide, and the only available drug treatment is limited to a short window following the ischemic event. Gastrodin is the major bioactive constituent extracted from thetuberGastrodia elata, and is currently used to treat dizziness in the clinic. "Early" application of gastrodin (before modeling or immediately after ischemic injury) has shown antioxidative and neuroprotective effects in a transient focal brain ischemia model in rodents; however, it is not known whether the delayed administration of gastrodin after

Spina bifida-predisposing heterozygous mutations in Planar Cell Polarity genes and Zic2 reduce bone mass in young mice.

Fractures are a common comorbidity in children with the neural tube defect (NTD) spina bifida. Mutations in the Wnt/planar cell polarity (PCP) pathway contribute to NTDs in humans and mice, but whether this pathway independently determines bone mass is poorly understood. Here, we first confirmed that core Wnt/PCP components are expressed in osteoblasts and osteoclasts in vitro. In vivo, we performed detailed µCT comparisons of bone structure in tibiae from young male mice heterozygous for NTD-associated mutations versus WT littermates. PCP signalling disruption caused by Vangl2 (Vangl2Lp/+) or

Dorsomedial hypothalamic NPY affects cholecystokinin-induced satiety via modulation of brain stem catecholamine neuronal signaling.

Increased neuropeptide Y (NPY) gene expression in the dorsomedial hypothalamus (DMH) has been shown to cause hyperphagia, but the pathway underlying this effect remains less clear. Hypothalamic neural systems play a key role in the control of food intake, in part, by modulating the effects of meal-related signals, such as cholecystokinin (CCK). An increase in DMH NPY gene expression decreases CCK-induced satiety. Since activation of catecholaminergic neurons within the nucleus of solitary tract (NTS) contributes to the feeding effects of CCK, we hypothesized that DMH NPY modulates NTS neural c

Transcriptomics Profiling of Alzheimer's Disease Reveal Neurovascular Defects, Altered Amyloid-β Homeostasis, and Deregulated Expression of Long Noncoding RNAs.

The underlying genetic variations of late-onset Alzheimer's disease (LOAD) cases remain largely unknown. A combination of genetic variations with variable penetrance and lifetime epigenetic factors may converge on transcriptomic alterations that drive LOAD pathological process. Transcriptome profiling using deep sequencing technology offers insight into common altered pathways regardless of underpinning genetic or epigenetic factors and thus represents an ideal tool to investigate molecular mechanisms related to the pathophysiology of LOAD. We performed directional RNA sequencing on high quali

Wip1 knockout inhibits neurogenesis by affecting the Wnt/β-catenin signaling pathway in focal cerebral ischemia in mice.

Neurogenesis correlates closely with the recovery of neural function after brain ischemia but the critical proteins and signaling pathways involved remain unclear. The phosphatase WIP1 has been shown to regulate neurogenesis in models of aging. However, it is not known if WIP1 affects neurogenesis and functional recovery after brain ischemia. To explore these questions, we performed permanent middle cerebral artery occlusion (MCAO) in mice and performed BrdU labeling, neurobehavioral testing, western blotting, and immunofluorescence staining. We found that ischemia induced WIP1 expression in t

Migraine pathophysiology: anatomy of the trigeminovascular pathway and associated neurological symptoms, cortical spreading depression, sensitization, and modulation of pain.

Scientific evidence supports the notion that migraine pathophysiology involves inherited alteration of brain excitability, intracranial arterial dilatation, recurrent activation, and sensitization of the trigeminovascular pathway, and consequential structural and functional changes in genetically susceptible individuals. Evidence of altered brain excitability emerged from clinical and preclinical investigation of sensory auras, ictal and interictal hypersensitivity to visual, auditory, and olfactory stimulation, and reduced activation of descending inhibitory pain pathways. Data supporting the

Single-cell transcriptional profiling reveals heterogeneity and developmental trajectories of Ewing sarcoma.

PURPOSE: Ewing sarcoma (EwS) is an aggressive malignant neoplasm composed of small round cells. The heterogeneity and developmental trajectories of EwS are uncertain. METHODS: Single-cell RNA sequencing was performed on 4 EwS tumor tissue samples, and 3 transcriptional atlases were generated. K-nearest neighbor algorithm was used to predict the origin of tumor cells at single-cell resolution. Monocle2 package was used to perform pseudotime trajectory analysis in tumor cells. Differentially expressed genes were compared against those in all other clusters via the FindMarkers function, and then

Optimized in vivo two-photon imaging reveals the essential role of the contralateral eye in functional optic nerve regeneration in zebrafish larvae.

BACKGROUND: The visual pathway, consisting of the eye, optic nerve, and brain, serves as a valuable model for studying neural regeneration. The exceptional regenerative capacity of the zebrafish visual system enables detailed investigation of neural repair mechanisms in vivo. Although the transparency of zebrafish larvae permits real-time imaging of axonal regeneration following transection, previous methodological limitations such as pigment interference and suboptimal imaging protocols have hindered high-resolution analyses of structural recovery and cellular interaction throughout the entir

Father engagement in child and adolescent mental health care: From neurobiology to clinical practice.

Fathers influence child development through biological, relational, and family-system pathways, yet they remain comparatively underrepresented in research and child mental health care. Emerging neurobiological evidence suggests that paternal caregiving is associated with experience-dependent adaptations in neural and neuroendocrine systems involved in caregiving and social responsiveness. Developmental research further identifies both direct pathways, including father-child interaction, play, caregiving, and emotional responsiveness, and indirect pathways involving coparenting, family function

Genetic Architecture of Cognitive Resilience in Alzheimer's Disease: Mechanisms, Pathways, and Therapeutic Implications.

Background/Objectives: Alzheimer's disease (AD) is defined by amyloid-β plaques and tau neurofibrillary tangles and is typically associated with progressive cognitive decline. However, a substantial subset of individuals remains cognitively intact despite intermediate-to-high AD pathology, a phenomenon termed cognitive resilience. This review aims to synthesize genetic variants and biological pathways associated with preserved cognition in the presence of AD neuropathology. Methods: We performed a narrative thematic synthesis of human genetic studies (GWAS, sequencing, biomarker-informed cohor

Role of Anti-GD2 Targeted PEG-b-PLGA Nanoparticles in the Treatment of MYCN Driven Neuroblastoma.

Neuroblastoma (NB) is an embryonic tumor originating from the neural crest. The most well-defined genetic alteration in NB is the overexpression of the MYCN protein. PI3K/AKT/mTOR and AURORA signaling pathways play a role in MYCN stabilization, and abnormal activation of these pathways has been identified in NB. Nanoparticle (NP) drug delivery systems targeting tumor cells directly assist in the combined delivery of agents and reducing toxicity. In this study, the aim was to develop NPs that reduce the activity of mTOR and AURORA pathways, potentially decreasing MYCN protein enhancement and st

Sympathetic regulation of bone homeostasis and remodeling: molecular mechanisms, pathophysiological roles, and therapeutic implications.

Bone metabolic disorders, such as osteoporosis, osteoarthritis, and rheumatoid arthritis, are characterized by disrupted skeletal homeostasis and represent major challenges in musculoskeletal health. Bone remodeling has long been interpreted as a coordinated process between osteoblast-mediated bone formation and osteoclast-mediated bone resorption. However, emerging evidence indicates that bone is not merely a structural tissue, but a dynamic regulatory organ influenced by neural, endocrine, immune, metabolic, and vascular signals. Among these regulatory systems, the sympathetic nervous system

Hypoxia impedes differentiation of cranial neural crest cells into derivatives relevant for craniofacial development.

Orofacial clefts are the second-most prevalent congenital malformation. Risk factors are multifactorial and include genetic components, but also environmental factors. One environmental factor is hypoxia during pregnancy, caused for instance by tobacco smoking, medication or living at high altitudes. We here show that hypoxia has only modest effects on proliferating cranial neural crest cells (CNCC), but dramatically influences their differentiation potential. We detected massive perturbations in their differentiation to chondrocytes, osteoblasts and smooth muscle cells. The transcriptional in

Morphogenesis and molecular considerations on congenital cardiac septal defects.

The primary unseptated heart tube undergoes extensive remodeling including septation at the atrial, atrioventricular, ventricular, and ventriculo-arterial level. Alignment and fusion of the septal components is required to ensure full septation of the heart. Deficiencies lead to septal defects at various levels. Addition of myocardium and mesenchymal tissues from the second heart field (SHF) to the primary heart tube, as well as a population of neural crest cells, provides the necessary cellular players. Surprisingly, the study of the molecular background of these defects does not show a great

Tissue-specific ablation of Prkar1a causes schwannomas by suppressing neurofibromatosis protein production.

Signaling events leading to Schwann cell tumor initiation have been extensively characterized in the context of neurofibromatosis (NF). Similar tumors are also observed in patients with the endocrine neoplasia syndrome Carney complex, which results from inactivating mutations in PRKAR1A. Loss of PRKAR1A causes enhanced protein kinase A activity, although the pathways leading to tumorigenesis are not well characterized. Tissue-specific ablation of Prkar1a in neural crest precursor cells (TEC3KO mice) causes schwannomas with nearly 80% penetrance by 10 months. These heterogeneous neoplasms were

Slow-wave sleep engages brainstem circuitry to prevent stress-induced anxiety.

The beneficial effects of sleep on anxiety are established, but the mechanisms remain unclear. We identify a GABAergic circuit from the parafacial zone (PZ) to the lateral parabrachial nucleus (LPB) neurons that project to the oval bed nucleus of the stria terminalis (ovBNST) as a node for slow-wave sleep (SWS)-mediated anxiolysis. Optogenetic activation of PZ GABAergic neurons following social defeat stress induces time-locked SWS and prevents anxiety. Multi-region Ca2+ recording reveals suppressed activity in LPB and ovBNST during natural and PZ-initiated SWS. The LPB-ovBNST pathway is requi

Structural connectome changes as biomarkers of stroke recovery: a longitudinal 7T MRI study.

BACKGROUND: Stroke leads to widespread brain connectivity changes, impacting areas both close and remote to the lesion. Post-stroke recovery dynamics are not fully understood. Investigating structural network reorganization over time can thus provide valuable information on adaptive and maladaptive neural plasticity changes on a subject-specific level. METHODS: Four first-time ischemic stroke patients (3 M, aged 50-69 years) with upper-extremity motor impairment were examined using ultra-high field 7T MRI structural imaging protocols. For each patient, we performed longitudinal lesion quantif
