The airway epithelium, a primary target for viral infection, plays a critical role in disease response-particularly in individuals with preexisting airway conditions such as cystic fibrosis (CF). At the onset of the SARS-CoV-2 pandemic, individuals with CF were expected to have severe outcomes based on earlier viral outbreaks; however, those on effective CF transmembrane conductance regulator (CFTR) modulators showed milder disease. Patients with CF on the CFTR modulator combination elexacaftor/tezacaftor/ivacaftor (ETI) combination therapy showed attenuated viral infection and reduced airway

BACKGROUND: Osteosarcoma is the most common malignant bone tumor in adolescents with a poor prognosis. So far, there is still a lack of effective molecular biomarkers for early diagnosis and effective therapeutic targets for osteosarcoma. Bruton tyrosine kinase (BTK) is reported to be overexpressed in various tumors and may be present as an independent prognostic biomarker. The functions of BTK in osteosarcoma are still unclear. Thereby, our study is devoted to investigating the status of BTK in the progression of osteosarcoma.
METHODS: An immunohistochemistry (IHC) assay was used to detect t

Chronic lymphocytic leukemia (CLL) is a B cell malignancy driven by aberrant signaling and microenvironmental support. Cytoskeletal remodeling contributes to these mechanisms, where the hematopoietic protein HS1 appears to play a key role. Thus, we generated C57BL/6 HS1 knockout (HS1KO) mice to assess its impact on lymphocyte development. Young HS1KO mice showed some alterations in B cell maturation and T cell differentiation, which appeared to normalize with age. In double-transgenic Eμ-TCL1-HS1KO mice, combining HS1 loss with the standard Eμ-TCL1 CLL mouse model, we observed a trend of reduc

BACKGROUND: Current U.S. and European guidelines recommend oral anticoagulation as a class IIa indication in patients with atrial fibrillation at intermediate risk for stroke; however, evidence from randomized trials is needed.
METHODS: We conducted a multicenter, open-label, adjudicator-masked superiority trial in South Korea involving patients with atrial fibrillation and an intermediate risk of stroke (a score of 1 in men and 2 in women on the CHA2DS2-VASc scale; range, 0 to 9, with higher scores indicating a greater risk of stroke). Patients were randomly assigned in a 1:1 ratio to receiv

Targeted therapies have transformed modern management of chronic lymphocytic leukaemia (CLL). Still, CLL remains an incurable disease, and key unresolved challenges include development of treatment resistance and intolerance. To address these challenges, it is necessary to define clinically actionable biomarkers that can predict individual treatment responses. Here, we aimed to map the treatment sensitivity and resistance landscapes of CLL cells from treatment-naïve and treatment-exposed patients to understand the evolution of treatment vulnerabilities. We performed ex vivo drug screens with 9

Immune thrombocytopenia (ITP) is associated with both bleeding and thrombotic complications. Thrombopoietin receptor agonists (TPO-RAs), particularly romiplostim and eltrombopag, are widely used as second-line therapy, yet comparative data on thromboembolic risk between these agents remain limited. We conducted a retrospective cohort study using the TriNetX federated research network, including adults with ITP who initiated romiplostim or eltrombopag between January 2010 and December 2025. ITP was defined using ICD-10 code D69.3. Patients with prior TPO-RA exposure, alternative thrombocytopeni

Epstein-Barr virus (EBV) is a ubiquitous pathogen that infects most adults worldwide. Although frequently associated with acute hepatitis, no specific treatment for EBV-induced hepatitis currently exists. Using murine γ-herpesvirus 68 (MHV68) infection in mice, which is a well-established murine model for EBV infection, we previously demonstrated that MHV68-induced hepatitis is mediated via the Toll-like receptor 4 (TLR4) signaling pathway. In this study, we investigated the anti-hepatitis effects of two anti-inflammatory agents, ibudilast and dipyridamole, which are known to suppress LPS/TLR4

Macrocyclization can enhance the selectivity of acyclic compounds toward structurally similar biological targets such as kinases. WEE1 regulates cellular homeostasis and is a promising target in oncology. The clinical candidate AZD1775 (1) failed to progress past Phase II trials because of patient tolerability issues, likely due to off-target inhibition of polo-like kinase 1 (PLK1). Herein, a computer-aided drug design approach was conducted to develop a macrocycle based on the 1-WEE1 X-ray cocrystal structure. Significantly enhanced WEE1 inhibitory selectivity over PLK1 was determined for lea

Bruton’s Tyrosine Kinase (BTK) is an anchor in B-cell receptor signaling and plays an important role in chronic lymphocytic leukemia (CLL). The use of covalent inhibitors of BTK, such as ibrutinib, enhances the survival of patients with CLL. However, mutations at the C481 residue cause resistance to ibrutinib and diminish its clinical efficacy. Recently, super-resistant mutants, i.e., T474M-C481S and T474I-C481S, were reported to cause manifold resistance to the BTK-targeting drug, ibrutinib; however, the mechanism of this resistance is still elusive. Structure-based approaches proved to be ef

BACKGROUND/OBJECTIVES: Colorectal cancer (CRC) remains a significant global health concern, with limited treatment options for metastatic stage 4 CRC. Fibroblast Growth Factor Receptor (FGFR) is a promising therapeutic target in CRC, while cannabidiol (CBD) has shown potential for inducing cell death and overcoming drug resistance. This study evaluates the efficacy of FGFR inhibitors and explores the synergistic effects of combining FGFR inhibitors with CBD in inducing apoptosis in CRC cells.
METHODS: Cannabidiol and FGFR inhibitors were applied, and protein expression was analyzed via Wester

Cutaneous melanoma is an aggressive skin cancer with high metastatic potential and frequent resistance to apoptosis-inducing therapies. Ibrutinib (IBR), a Bruton's tyrosine kinase inhibitor, has demonstrated antitumor potential, yet its topical application is not well established. This study evaluated the cytotoxic, antiproliferative, and skin permeation effects of free IBR and IBR-loaded nanostructured lipid carriers (IBR + NLC) using human melanoma cells (SK-MEL-28) and ex vivo human skin models. NLC + IBR presented enhanced toxicity towards melanoma cells by promoting necroptosis, as eviden

The use of elexacaftor/tezacaftor/ivacaftor (ETI) has been associated with increased fertility in women with cystic fibrosis (CF) and is increasingly used during pregnancy to support both maternal and fetal health. However, little is known about the pharmacokinetics (PK) of ETI during pregnancy, which is crucial for optimizing its efficacy and safety. This study aimed to predict the PK of ETI during pregnancy and to determine the maternal dose required to achieve therapeutic concentrations in both the maternal and fetus. The pregnancy physiological-based pharmacokinetic (PBPK) model within the

Alzheimer's disease (AD) is characterized by amyloid-β (Aβ) deposition, tau hyperphosphorylation, neuroinflammation, oxidative stress, and progressive neuronal loss. The endocannabinoid system regulates synaptic function, inflammation, and redox homeostasis through cannabinoid receptor type 1 (CB1). This study aimed to determine whether pharmacological activation or inhibition of the CB1 receptor differentially modulates Alzheimer's disease-related pathology. 3xTg-AD mice received weekly intraperitoneal injections of the CB1 agonist ACEA (1 mg/kg) or the inverse agonist AM251 (1 mg/kg) from 6

BACKGROUND: The approval of cystic fibrosis transmembrane conductance regulator modulators elexacaftor/tezacaftor/ivacaftor (ETI), has significantly improved pulmonary function for people with cystic fibrosis (pwCF). However, the effects on CF-related bone disease and body composition remain unclear.
METHODS: This retrospective real-world study examined adults with CF who received ETI treatment. Bone density and body composition were measured via dual-energy X-ray absorptiometry (DXA) 1.8 (SD 0.7) years before (preDXA) and 1.5 (SD 0.4) years after ETI initiation (postDXA1). In a subgroup, mea

Diffuse Large B-cell Lymphoma (DLBCL) is a genomically-heterogenous disease affecting over 70,000 patients per year that presents a clinical challenge despite the success of frontline regimens and second-line Chimeric Antigen receptor T-cell (CAR-T) therapy. Recently, genomic alterations and tumor microenvironment features associated with poor CAR-T response have been identified, with MYC amplification emerging in new analyses. This retrospective analysis aimed to integrate various data to identify genomic partnerships capable of providing added clarity and actionable treatment targets within

Disease tolerance is a key defense mechanism that limits damage to the host without directly reducing pathogen levels. In malaria, these mechanisms are essential for preventing severe disease and death but remain poorly understood. In this study, we show that glucocorticoid receptor (GR)-mediated processes play a vital role in disease tolerance during Plasmodium chabaudi AS infection. GR deletion in infected mice resulted in lethal hypoglycemia and a cytokine storm. Hypoglycemia was driven by severe metabolic dysfunction in the liver and spleen, characterized by increased glucose uptake, glyco

Some patients with chronic lymphocytic leukemia who develop progressive disease (PD) during covalent Bruton tyrosine kinase (BTK) inhibitor treatment acquire resistance mutations in BTK or PLCG2. Here, we report gene mutation data from paired baseline and PD peripheral blood samples from 52 patients (zanubrutinib, n = 24; ibrutinib, n = 28) who, at an early median follow-up of 25.7 months, progressed on zanubrutinib or ibrutinib treatment in ALPINE. No BTK mutations were observed at baseline; at PD, 8 patients (zanubrutinib, n = 5; ibrutinib, n = 3) acquired 17 BTK mutations, 82.4% (zanubrutin
