Translational Progress of Inorganic Nanotheranostic Platforms for Gynecologic Malignancies: A Narrative Review of Endometrial, Ovarian, and Cervical Cancer Focusing on Fertility Preservation, Biosafety, and Industrial Translation.

Endometrial, ovarian, and cervical cancers differ substantially in epidemiology, anatomy, and unmet clinical need, yet the same inorganic nanomaterial toolbox is often discussed without disease-specific translational context. This narrative review synthesizes preclinical and clinical literature on gold, magnetic, mesoporous silica, carbon-based, and metal-organic framework nanotheranostic platforms identified through PubMed and Web of Science searches from database inception through August 2026, using a structured comparison across endometrial, ovarian, and cervical cancer. Particular attentio

Core Outcome Sets for Pediatric Perioperative Research: An International Stakeholder Engagement Exercise and Consensus Report from the Pediatric Perioperative Outcomes Group.

BACKGROUND: Identifying the outcomes that matter in clinical research is important, especially those that matter to patients and their parents/guardians. Consistency in outcome reporting enables meaningful assessments of interventions and facilitates comparison of results across trials. The aim of this study was to develop core outcome sets for pediatric perioperative research. METHODS: The authors determined core outcome sets through extensive stakeholder engagement, following a stepwise process as recommended by the Core Outcome Measures in Effectiveness Trials (COMET) initiative. They surv

Chronic social stress induces generalized hyper-sensitivity to aversion: A mouse model with translational validity for understanding and treating negative valence disorders.

The RDoC framework focuses on neurobehavioral processes often dysfunctional in mental disorders and commensurate with translational research. Generalized hyper-sensitivity to aversion/threat is common in various stress-related emotional disorders; increased Pavlovian aversion learning-memory (PAL, PAM) provides a translational paradigm for its study. Here we present the development and application of a mouse model for the study of generalized hyper-sensitivity to aversion/threat. In male adult mice, chronic exposure to social aversion (chronic social stress, CSS) leads, relative to controls (C

Biodegradable Contact Lenses for Targeted Ocular Drug Delivery: Recent Advances, Clinical Applications, and Translational Perspectives.

Ocular drug delivery presents a persistent clinical challenge due to the protective anatomical structure of the eye, physiological barriers such as reflex blinking, and continuous tear fluid turnover. These factors significantly limit the bioavailability of topically applied medications, reducing the therapeutic effectiveness of conventional formulations, such as eye drops, ointments, and suspensions, particularly in the management of chronic ocular disorders, including dry eye syndrome, diabetic retinopathy, and age-related macular degeneration. Drug-eluting contact lenses (DECLs) offer a pro

Advances in nanomedicine-based retinal drug delivery: mechanisms and translational applications.

Retinal diseases like age-related macular degeneration (AMD), diabetic retinopathy (DR), and diabetic macular edema (DME), alongside optic neuropathies such as glaucoma, are primary contributors to irreversible visual impairment and blindness, and not only impact the quality of life but also place considerable socioeconomic pressures on healthcare systems worldwide. Despite the availability of several therapeutic modalities, the clinical management of these conditions remains challenging due to the unique anatomical and physiological barriers of the eye (i.e., tear film, cornea, blood-aqueous

Translating medication effects for alcohol use disorder across preclinical, human laboratory, and clinical trial outcomes using meta-analysis.

Animal models are used for preliminary testing of novel compounds for alcohol use disorder (AUD). However, it is unclear whether early efficacy in preclinical models reliably predicts efficacy in human laboratory and clinical trials. We searched the literature for medications tested for AUD in preclinical models (i.e., two-bottle choice [2-BC] and operant reinstatement), human laboratory cue-reactivity, and randomized clinical trials (RCTs). For preclinical models, we computed medication effects on 2-BC alcohol preference and consumption (k = 77 studies, 14 medications) as well as operant rein

Prospective Harmonization, Common Data Elements, and Sharing Strategies for Multicenter Pre-Clinical Traumatic Brain Injury Research in the Translational Outcomes Project in Neurotrauma Consortium.

Effective team science requires procedural harmonization for rigor and reproducibility. Multicenter studies across experimental modalities (domains) can help accelerate translation. The Translational Outcomes Project in NeuroTrauma (TOP-NT) is a pre-clinical traumatic brain injury (TBI) consortium charged with establishing and validating noninvasive TBI assessment tools through team science. Here, we present practical approaches for harmonization of TBI research across five centers providing needed vocabulary and structure to achieve centralized data organization and use. This includes data sh

Toward a Translational Model of Sex-Associated Pavlovian Phenotypes.

A recent study by Hakus et al. (2025) demonstrated sex-associated differences in Pavlovian phenotypes in rodents, with females more likely to exhibit sign-tracking behaviour and males more likely to exhibit goal-tracking behaviour. In the present work, we provide evidence that similar patterns emerge in humans. Using a validated eye-tracking procedure in a Pavlovian learning paradigm, we show that women are more frequently classified as sign-trackers and quantitatively show greater sign-tracking behaviour than men in a large human sample. These results support the translational value of precli

Towards a minimal core dataset for systemic lupus erythematosus studies.

OBJECTIVE: SLE is a complex, heterogenous autoimmune disease. SLE researchers do not always collect the same data, making comparative studies difficult. We aimed to ascertain what variables SLE clinical researchers commonly collect for SLE research. Our ultimate goal is to generate a minimal core dataset for future SLE studies. METHODS: In 2020, we designed and distributed a questionnaire to members of the Systemic Lupus Erythematosus International Collaborating Clinics (SLICC) as well as additional active research centres in China. Our survey included 26 questions about the types of data tha

Translational insights from EAE models : decoding MOGAD pathogenesis and therapeutic innovation.

Myelin Oligodendrocyte Glycoprotein Antibody-Associated Disease (MOGAD) is a rare acquired demyelinating syndrome manifesting as optic neuritis (ON), transverse myelitis (TM), acute disseminated encephalomyelitis (ADEM), and brainstem encephalitis. The disease is characterized by serum autoantibodies targeting myelin oligodendrocyte glycoprotein (MOG), which is exclusively expressed on central nervous system (CNS) myelin and oligodendrocyte membranes. Experimental autoimmune encephalomyelitis (EAE) models have been instrumental in elucidating how these antibodies trigger complement-dependent c

Role of macrophages in aortic dissection pathogenesis: insights from preclinical studies to translational prospective.

Aortic dissection is a critical vascular disease that is characterized by a high mortality rate and inflammation significantly influences its onset and progression. Recent studies highlight the integral role of macrophages, key players in the immune system, in the pathological landscape of aortic dissection. These cells are involved in crucial processes, such as the remodeling of the extracellular matrix, immunocyte infiltration, and phenotypic switching of smooth muscle cells, which are essential for the structural integrity and functional dynamics of the aortic wall. Despite these insights,

[Our visit in the Czech National Institute of Mental Health - experiences and impressions].

In late April 2024, we visited the National Institute of Mental Health (NIMH) with our psychiatry residency class in Klecany, near Prague, the Czech Republic. There is no similar institution in Hungary, so it was worthwhile to learn not only about the research being conducted there, but also how the successful integration of preclinical and clinical research, patient care, and education is achieved in a psychiatric institute. In this article, we briefly describe the history and professional profile of the institute, its capacities, and the work of the research groups and departments that were

Reflections on the Rega Institute for Medical Research, at the fiftieth anniversary of the Rega Stichting vzw (Rega Instituut vzw, Rega Foundation).

The idea to start the Rega Foundation was conceived in 1971 at an informal meeting organized by Prof. Piet De Somer (where Prof. Alfons Billiau, Prof. André Vlerick and I were also present), before the Foundation was formally created in 1972. From the early years some antiviral compounds, such as BVDU and the aminoacyl esters of acyclovir (from which ultimately valacyclovir evolved) originated. The advent of AIDS in 1981 and the discovery of the etiologic agent (HIV) thereof in 1983 have led to the identification of an avalanche of anti-HIV compounds in which the Rega Institute has played a pr

A Mouse Model of Multiple System Atrophy: Bench to Bedside.

Multiple system atrophy (MSA) is a rare neurodegenerative disorder with unclear etiology, currently difficult and delayed diagnosis, and rapid progression, leading to disability and lethality within 6 to 9 years after symptom onset. The neuropathology of MSA classifies the disease in the group of a-synucleinopathies together with Parkinson's disease and other Lewy body disorders, but features specific oligodendroglial inclusions, which are pathognomonic for MSA. MSA has no efficient therapy to date. Development of experimental models is crucial to elucidate the disease mechanisms in progressio

Mendelian randomisation and experimental medicine approaches to interleukin-6 as a drug target in pulmonary arterial hypertension.

BACKGROUND: Inflammation and dysregulated immunity are important in the development of pulmonary arterial hypertension (PAH). Compelling preclinical data supports the therapeutic blockade of interleukin-6 (IL-6) signalling. METHODS: We conducted a phase 2 open-label study of intravenous tocilizumab (8 mg·kg-1) over 6 months in patients with group 1 PAH. Co-primary end-points were safety, defined by incidence and severity of adverse events, and change in pulmonary vascular resistance. Separately, a mendelian randomisation study was undertaken on 11 744 individuals with European ancestry includ
