[Nifedipine in primary pulmonary hypertension while waiting for heart-lung transplantation].

Primary pulmonary hypertension is a disease of unknown etiology. No effective treatment is known. Heart-lung transplantation is a new possibility for these otherwise healthy young persons. A woman aged 27 years was treated successfully with the calcium-channel-blocker, nifedipine, with a good hemodynamic result after the initial treatment and three weeks thereafter.

The role of alkalinization-induced Ca2+ influx in sperm motility activation of a viviparous fish Redtail Splitfin (Xenotoca eiseni).

Mechanisms regulating sperm motility activation are generally known in oviparous fishes, but are poorly understood in viviparous species. The mechanism of osmotic-shock induced signaling for oviparous fishes is not suitable for viviparous fishes which activate sperm motility within an isotonic environment. In addition, the presence of sperm bundles in viviparous fishes further complicates study of sperm activation mechanisms. The goal of this study was to establish methodologies to detect intracellular Ca2+ signals from sperm cells within bundles, and to investigate the signaling mechanism of

Involvement of calcium ion in the stimulated shoot elongation of arrowhead tubers under anaerobic conditions.

Shoot elongation of arrowhead (Sagittaria pygmaea Miq.) tubers was stimulated in anaerobic conditions. The anaerobic elongation was attributed to stimulation of cell elongation in the middle of the shoots. The anaerobic elongation of the shoots was severely inhibited by ethylene glycol bis(beta-aminoethylether)-N,N,N',N'-tetraacetic acid (EGTA). The EGTA inhibition was completely nullified by exogenous CaCl2, which acts as an enhancer of anaerobic elongation. Moreover, calcium channel blockers, verapamil, diltiazem and LaCl3, inhibited the anaerobic elongation enhanced by CaCl2. These results

Synergistic antiarrhythmic mechanism of INa-L and RyR2 blockade: normalization of pathological calcium cycling and CaMKII signaling.

BACKGROUND: Arrhythmias represent a leading cause of mortality among individuals with cardiovascular diseases. Considering the failure of antiarrhythmic drugs targeting single-ion channels in clinical trials, this study aims to evaluate the pharmacodynamics of various combinations of single-channel blockers to identify safer and more effective therapeutic regimens. METHODS: The antiarrhythmic effects of inhibitors alone and pairwise combination strategies targeting late sodium current (INa-L), hERG potassium current (IKr), ryanodine receptor 2 (RyR2), and L-type calcium current (ICa-L) inhibit

The secretory pathway calcium ATPase PMR-1/SPCA1 has essential roles in cell migration during Caenorhabditis elegans embryonic development.

Maintaining levels of calcium in the cytosol is important for many cellular events, including cell migration, where localized regions of hig … c-68/RyR modulates levels of embryonic lethality in Pmr-1 strains, indicating pmr-1 acts with these calcium channels to regulate cell migration. This analysis reveals novel genes involved in C. elegans cell migration, as well as a new role in cell migration for the highly conserved SPCA gene family.

Protease secretions by the invading blastocyst induce calcium oscillations in endometrial epithelial cells via the protease-activated receptor 2.

BACKGROUND: Early embryo implantation is a complex phenomenon characterized by the presence of an implantation-competent blastocyst and a receptive endometrium. Embryo development and endometrial receptivity must be synchronized and an adequate two-way dialogue between them is necessary for maternal recognition and implantation. Proteases have been described as blastocyst-secreted proteins involved in the hatching process and early implantation events. These enzymes stimulate intracellular calcium signaling pathways in endometrial epithelial cells (EEC). However, the exact molecular players un

Spontaneous, synchronous zinc spikes oscillate with neural excitability and calcium spikes in primary hippocampal neuron culture.

Zinc has been suggested to act as an intracellular signaling molecule due to its regulatory effects on numerous protein targets including enzymes, transcription factors, ion channels, neurotrophic factors, and postsynaptic scaffolding proteins. However, intracellular zinc concentration is tightly maintained at steady levels under natural physiological conditions. Dynamic changes in intracellular zinc concentration have only been detected in certain types of cells that are exposed to pathologic stimuli or upon receptor ligand binding. Unlike calcium, the ubiquitous signaling metal ion that can

Non-motile primary cilia as fluid shear stress mechanosensors.

Primary cilia are sensory organelles that transmit extracellular signals into intracellular biochemical responses. Structural and functional defects in primary cilia are associated with a group of human diseases, known as ciliopathies, with phenotypes ranging from cystic kidney and obesity to blindness and mental retardation. Primary cilia mediate mechano- and chemosensation in many cell types. The mechanosensory function of the primary cilia requires the atypical G-protein-coupled receptor polycystin-1 and the calcium-permeable nonselective cation channel polycystin-2. Mechanical stimulations

[Function and role of the sarcoplasmic reticulum in heart disease].

The sarcoplasmic reticulum (SR) constitutes the main intracellular calcium store in striated muscle and plays an important role in the regulation of excitation-contraction-coupling (ECC) and of intracellular calcium concentrations during contraction and relaxation. The regulation of ECC occurs due to the interaction among the main proteins of the SR that are the calcium release channel or ryanodine receptor, the Ca2+-ATPase, phospholamban and calsequestrin. Due to the importance of ECC in the physiopathology of a number of cardiac diseases, the role of the SR and its components has been widely

Malignant hyperthermia.

Malignant hyperthermia is an autosomal-dominant inherited disorder of the skeletal muscle cell characterized by a hypermetabolic response to all commonly used inhalational anaesthetics and depolarizing muscle relaxants. The clinical syndrome includes muscle rigidity, hypercapnia, tachycardia and myoglobinuria as result of increased carbon dioxide production, oxygen consumption and muscle membrane breakdown. In human beings and animals susceptible to malignant hyperthermia, it is generally accepted that an increase in the level of myoplasmic free calcium is the cause of the syndrome. Various hy

Experiment study on the effect of Sheng-Mai injection on the contractivity of diaphragm and its mechanism.

In this study the effect of Sheng-Mai Injection i.e. Red Ginseng-Ophiopogon Root Injection (one kind of traditional Chinese medicines) on the contractivity of diaphragm was observed. The results confirmed that Sheng-Mai Injection increased Pdi of the fatigued diaphragm in rabbits and reduced the time needed for the recovery of Pdi of fatigued diaphragm to the normal value. These results suggest that Sheng-Mai Injection can increase the contractive force and promote the recovery of the fatigued diaphragm. The effect of Sheng-Mai Injection on the contractivity of the isolated diaphragmatic bundl

Hyperactive deoxy-PIEZO1 shapes the circulatory life cycle of irreversibly sickled cells.

Sickle cell disease (SCD), affecting millions worldwide, is caused by the homozygous inheritance of the abnormal hemoglobin HbS. Deoxygenation of HbS in the venous circulation permeabilizes sickle cells to calcium via PIEZO1 channels, triggering a dehydration cascade driven by the outward electrochemical potassium gradient. This mechanism operates with particular intensity in a subpopulation of sickle red blood cells (RBCs), the irreversibly sickled cells (ISCs). The lifespan of ISCs is extremely short, about 4-7 days. Most of this time is spent in a profoundly dehydrated condition, the irreve

Fluorescence-coupled micropipette aspiration assay to examine calcium mobilization caused by red blood cell mechanosensing.

Mechanical stimuli such as tension, compression, and shear stress play critical roles in the physiological functions of red blood cells (RBCs) and their homeostasis, ATP release, and rheological properties. Intracellular calcium (Ca2+) mobilization reflects RBC mechanosensing as they transverse the complex vasculature. Emerging studies have demonstrated the presence of mechanosensitive Ca2+ permeable ion channels and their function has been implicated in the regulation of RBC volume and deformability. However, how these mechanoreceptors trigger Ca2+ influx and subsequent cellular responses are
