Competitive PCR assays were developed for the enumeration of the rumen cellulolytic bacterial species: Fibrobacter succinogenes, Ruminococcus albus and Ruminococcus flavefaciens. The assays, targeting species-specific regions of 16S rDNA, were evaluated using DNA from pure culture and rumen digesta spiked with the relevant cellulolytic species. Minimum detection levels for F. succinogenes, R. albus and R. flavefaciens were 1-10 cells in pure culture and 10(3-4) cells per ml in mixed culture. Human renal calculi surgically removed from kidney stone patients were obtained and chemically analysed. Stones with CaOx (calcium oxalate) as the major component were washed in 0.15 M NaCl with gentle stirring for 48 h and then pulverised to a fine powder. Five patients presenting Hunter's syndrome were biochemically studied. Quantification of urinary glycosaminoglycans (GAGs), electrophoretic characterization and correlation with enzymatic activity in leucocytes were carried out. In all cases, urinary GAGs/creatinine ratio was increased. Electrophoresis revealed the presence of heparan sulfate (HS) and dermatan sulfate (DS) in four cases (80%), but in the remaining patient, only DS was present. In all patients, deficient enzymatic activity was demonstrated. These results show evidences of biochemical differences in this syndrome.

Striped bass (Morone saxatilis) were implanted with beta-estradiol to induce the production of vitellogenin, the egg yolk precursor produced by the liver. In order to clarify the pathogenesis of carbon disulfide retinopathy, fluorescein fundus angiograms were analyzed for 143 workers exposed to CS2 and 40 unexposed workers (reference) from viscose rayon factories. Retinal abnormalities, characterized by saccular microaneurysms, ellipsoidal microaneurysms and loop formation of small vessels, and atrophic/degenerative changes of pigmentary epithelium, were found in workers exposed to CS2. When diabetes and cardiovascular disease were first classed as possible fibre-deficiency diseases, laboratory and clinical evidence was lacking. Fundic mucosal pepsinogens reveal six major bands of protease activity on gel electrophoresis and four minor bands. Antral mucosa shows two major bands of activity; the four rapidly moving bands found in fundic mucosa are found in lesser activity in the antral mucosa. Duodenal mucosa shows only one slowly moving band of protease activity.
