Targeted mutagenesis of the ICP4 transactivation domain generates a protective DIVA vaccine against duck plague.

Duck plague, caused by duck plague virus (DPV), causes significant economic losses to the global waterfowl industry. The control of this disease is hampered by the limitations of conventional vaccines, which include the risk of virulence reversion, short-lived immunity, and the inability to differentiate infected from vaccinated animals (DIVA). To address these issues, we explored the rational design of a novel live-attenuated DIVA vaccine by targeting the essential immediate-early gene ICP4. A quadruple mutation was introduced into the N-terminal transactivation domain of ICP4. The resulting

Genotypic characterization of bovine rotavirus A from diagnostic submissions from 9 U.S. states, 2020-2025.

Bovine rotavirus A is a major cause of neonatal calf diarrhea and can lead to significant economic losses for cattle producers. Studying genotypic diversity among circulating RVA strains is critical to understanding virus epidemiology and to enhancing vaccine development. We identified 1,515 fecal samples from calves with enteric disease. Samples originated from 9 U.S. states and were submitted to the Nebraska Veterinary Diagnostic Center between 2020-2025 for bovine rotavirus A (RVA) testing. Of these 1,515 samples, 540 tested positive by reverse-transcription quantitative real-time PCR (RT-q

[Revolutionizing physiology: advances in organ-on-a-chip technology].

To address the low predictability (92% clinical failure rate) and high costs associated with species differences in traditional animal testing, this research advocates for New Approach Methodologies (NAMs) in alignment with the US FDA Modernization Act 2.0. The central achievement presented is the development of a "human Heart-on-a-chip." This microfluidic device co-cultures human iPS-derived cardiomyocytes, fibroblasts, and vascular endothelial cells to mimic living 3D tissue structures and fluid environments. The system demonstrated superior physiological reproducibility compared to conventi

Diagnostic and therapeutic strategies in combating implanted medical device-associated bacterial biofilm infections.

Bacterial biofilms exhibit remarkable resistance against conventional antibiotics and are capable of evading the humoral immune response. They account for nearly 80% of chronic infections in humans. Development of bacterial biofilms on medical implants results in their malfunctioning and subsequently leads to high mortality rates worldwide. Therefore, early and precise diagnosis of bacterial biofilms on implanted medical devices is essential to prevent their failure and associated complications. Culture-based methods are time consuming, more prone to contamination and often exhibit low sensiti

Gaining Biological Insights Into Deltamethrin-Induced Toxicity and Resistance at the Molecular Level.

Deltamethrin (DM), a type II pyrethroid, is one of the most widely used insecticides around the world. The link between diseases including obesity, neurodevelopmental disorders, liver fibrosis and DM treatment has been elucidated in organisms that exposed DM. Also, in aquatic organisms, hepatopancreatic necrosis disease, impaired cellular development, and decreased mobility could be related to DM treatment for pond cleaning and fending off arthropods causing diseases such as Caligidosis. Apart from that, DM resistance in target organisms has been a staggering issue due to the prevalence of DM-
