A Genetic Test Predicts Providence Brace Success for Adolescent Idiopathic Scoliosis When Failure Is Defined as Progression to >45 Degrees.

STUDY DESIGN: Retrospective cohort. OBJECTIVE: To determine whether a genetic test is associated with successful Providence bracing for adolescent idiopathic scoliosis (AIS). SUMMARY OF BACKGROUND DATA: Genetic factors have been defined that predict the risk of progression of AIS in a polygenic fashion. From these data, a commercially available genetic test, ScoliScore, was developed. It is now used in clinical practice for counseling and to guide clinical management. Bracing is a mainstay of treatment for AIS. Large efforts have been made recently to reduce potential confounding across stud

Microevolution and epidemic spread of serogroup A Neisseria meningitidis--a review.

An extensive and representative strain collection of serogroup A Neisseria meningitidis was established. These bacteria were obtained from different endemic and epidemic/pandemic sources and include strains from diseased patients and healthy carriers. The genetic relationships of the bacteria were defined by multi-locus enzyme electrophoresis and sequence polymorphisms of genetically variable antigens have been analyzed in closely-related groupings. The results are interpreted as reflecting a balance of recombination events, which disrupt clonal relationships, and sequential bottlenecks, which

Histocompatibility antigens in primary biliary cirrhosis.

Primary biliary cirrhosis (PBC) is characterized by marked perturbation of the immune system. In addition, there are familial associations which suggest genetic background to the disease. We have studied HLA-A, B, and D loci in 15 Japanese women with PBC, and compared them to controls. Although HLA-A and B loci in PBC patients showed similar prevalence to the control population, HLA-DHO was significantly more common, occurring in 69% of patients with PBC and 21% of controls. This suggests a genetic predisposition to PBC, and the HLA-DHO locus association may reflect a genetic abnormality of th

Feasibility and pitfalls of combining small subpopulations for genome-wide association analysis of maize (Zea mays L.) kernel traits.

Genome-wide association studies (GWASs) typically require large populations to achieve sufficient statistical power; however, maintaining large populations is labor-intensive and costly. In this study, we evaluated the feasibility of improving GWAS detection power by strategically integrating small maize (Zea mays L.) subpopulations for kernel-related traits. Using high-throughput phenotyping of eight kernel traits, GWAS was conducted across four different population combinations. Expanding population size generally increased single-nucleotide polymorphism (SNP) association strength and enable

Association between HLA-B*46 allele and Graves disease in Asian populations: a meta-analysis.

Graves' disease (GD) is a leading cause of hyperthyroidism, which affects 1.0-1.6% of the general population. Previous studies reported a higher GD prevalence in Asian populations compared to Caucasian populations. The etiology of GD involves complex interactions between predisposing genes and environmental triggers. Genetic studies have shown that the human leukocyte antigen (HLA) is an important candidate genetic region associated with GD in Asian populations. However, the results were inconsistent and inconclusive. Here, we performed a meta-analysis to evaluate the role of the HLA-B*46 alle

Possible transmission of Sarcoptes scabiei between herbivorous Japanese serows and omnivorous Caniformia in Japan: a cryptic transmission and persistence?

BACKGROUND: Two transmission patterns of Sarcoptes scabiei in host mammal communities have been reported based on microsatellite-level genetic studies in the last two decades. While one involves restrictions among different host taxa, the other is associated with predator-prey interactions between different host taxa. In contrast to these observations, the present study reports a possible irregular case of transmission of S. scabiei between herbivorous Japanese serow and omnivorous Caniformia mammals in Japan, though under very weak predator-prey relationships. METHODS: DNA from 93 Sarcoptes

Genetic Risk Score Is Associated with Vertical Cup-to-Disc Ratio and Improves Prediction of Primary Open-Angle Glaucoma in Latinos.

PURPOSE: Genome-wide association studies have identified multiple genetic variants associated with vertical cup-to-disc ratio (VCDR). Genetic risk scores (GRS) examine the aggregate genetic effect of individual variants on a trait by combining these separate genetic variants into a single measure. The purpose of this study was to construct GRS for VCDR and to determine whether the GRS are associated with VCDR and whether the GRS increase the discriminatory ability for primary open-angle glaucoma (POAG) in a Latino population. DESIGN: Population-based genetic association study. PARTICIPANTS:

Spatial analyzes of HLA data in Rio Grande do Sul, south Brazil: genetic structure and possible correlation with autoimmune diseases.

BACKGROUND: HLA genes are the most polymorphic of the human genome and have distinct allelic frequencies in populations of different geographical regions of the world, serving as genetic markers in ancestry studies. In addition, specific HLA alleles may be associated with various autoimmune and infectious diseases. The bone marrow donor registry in Brazil is the third largest in the world, and it counts with genetic typing of HLA-A, -B, and -DRB1. Since 1991 Brazil has maintained the DATASUS database, a system fed with epidemiological and health data from compulsory registration throughout the

Spatial genetic structure reflects extensive clonality, low genotypic diversity and habitat fragmentation in Grevillea renwickiana (Proteaceae), a rare, sterile shrub from south-eastern Australia.

BACKGROUND AND AIMS: The association of clonality, polyploidy and reduced fecundity has been identified as an extinction risk for clonal plants. Compromised sexual reproduction limits both their ability to adapt to new conditions and their capacity to disperse to more favourable environments. Grevillea renwickiana is a prostrate, putatively sterile shrub reliant on asexual reproduction. Dispersal is most likely limited by the rate of clonal expansion via rhizomes. The nine localized populations constituting this species provide an opportunity to examine the extent of clonality and spatial geno

Unique social, identity, and community factors impact prophylactic surgery discussions in young adults who are BRCA1, BRCA2, and PALB2 positive.

The role of a cancer genetic counselor includes communication about emotions, thoughts, and decision-making related to prophylactic surgery. No prior study has fully evaluated the preferences of young adults who are BRCA1, BRCA2 (BRCA1/2), or PALB2 positive and their communication preferences on discussing prophylactic surgery in a genetic counseling session. We addressed this through semi-structured interviews with 13 individuals who were assigned female sex at birth, aged 18-30 during the time of genetic counseling, had seen a genetic counselor in the past 7 years, and tested positive for a

The role of endocrine dysfunction in pregnancy loss and the modulating influence of genetic factors: A structured narrative review.

BACKGROUND: Pregnancy loss, including recurrent cases, is a common problem for couples worldwide, with serious medical and emotional effects. While many factors can cause pregnancy loss, hormonal imbalances stand out as the main, treatable reason. Genetics rarely causes pregnancy loss by itself, but it can affect hormone pathways and explain differences in risk among populations. OBJECTIVE: This review looks at how key hormone-related conditions-like thyroid problems, PCOS with insulin resistance, luteal phase defects, poorly controlled diabetes, and high prolactin-raise the risk of miscarria

Investigating sex-specific associations of Parkinson's disease with sex hormones and sex hormones-related phenotypes using Mendelian randomization.

BACKGROUND: It has been reported that the incidence of Parkinson's disease (PD) in men is 1.5 times that of women, which is easily associated with the protective effect of estrogens on the dopaminergic system. However, the exact direction and magnitude of the effect of sex hormones on PD risk have not been clarified. This study aimed to evaluate the sex-specific association between PD and sex hormones and their related phenotypes using Mendelian randomization (MR). METHOD: We utilized summary statistics from seven sex-specific genome-wide association studies (GWAS), including the UK Biobank,
