High yields of K-ras mutations in intraductal papillary mucinous tumors and invasive adenocarcinomas induced by N-nitroso(2-hydroxypropyl)(2-oxopropyl)amine in the pancreas of female Syrian hamsters.

Ductal adenocarcinoma, the most common form of pancreatic cancer in humans, is associated with activation of the K-ras oncogene in approximately 90% of cases. In contrast, K-ras mutations are found in < 50% of the relatively rare intraductal papillary mucinous tumor (IPMT), which arises in the main pancreatic ducts. Since both adenocarcinomas and IPMTs are believed to arise from ductal cells and progress through similar sequences of morphological changes (i.e. flat hyperplasia, papillary hyperplasia, atypia and carcinoma in situ), it is clear that such progression may not always necessitate ac

Mechanistic prediction of apixaban and rivaroxaban secretion into human milk and infant systemic exposure through an integrated physiologically based pharmacokinetic framework.

Accurately characterizing the transfer of apixaban and rivaroxaban into human milk is essential for evaluating their safety during lactation and for guiding anticoagulant therapy in breastfeeding patients. This study aimed to establish a mechanistic physiologically based pharmacokinetic (PBPK) modeling approach linking maternal exposure, mammary drug transport, and infant systemic levels for both anticoagulants. Passive permeability and efflux transport across the mammary epithelium were measured using an in vitro human mammary epithelial cell model, providing information on diffusion and tran

A model to simulate cardio-respiratory responses to fentanyl analgesia after traumatic injury.

Prehospital pain management is challenging and is expected to become even more complex in future large-scale combat operations, where mass-casualty events and evacuation delay may be inevitable. An improved understanding of how analgesic drugs affect the physiological response of trauma patients can enhance treatment efficacy of combat casualties. We previously developed and validated a cardio-respiratory (CR) model for humans that accounts for vital-sign responses to hemorrhagic injuries, resuscitation with six fluid types, airway obstruction, and ketamine analgesia. Here, we extended the mod

Dietary Caffeine Synergizes Adverse Peripheral and Central Responses to Anesthesia in Malignant Hyperthermia Susceptible Mice.

Ryanodine receptor (RYR) mutations confer stress-triggered malignant hyperthermia (MH) susceptibility. Dietary caffeine (CAF) is the most commonly consumed psychoactive compound by humans. CAF-triggered Ca2+ release and its influences on skeletal muscle contractility are widely used as experimental tools to study RYR function/dysfunction and diagnose MH susceptibility. We hypothesize that dietary CAF achieving blood levels measured in human plasma exacerbates the penetrance of RYR1 MH susceptibility mutations triggered by gaseous anesthetic, affecting both central and peripheral adverse respon

Prenatal exposure to pesticides and risk for holoprosencephaly: a case-control study.

BACKGROUND: Pesticide exposure during susceptible windows and at certain doses are linked to numerous birth defects. Early experimental evidence suggests an association between active ingredients in pesticides and holoprosencephaly (HPE), the most common malformation of the forebrain in humans (1 in 250 embryos). No human studies to date have examined the association. This study investigated pesticides during multiple windows of exposure and fetal risk for HPE. It is hypothesized that pre-conception and early pregnancy, the time of brain development in utero, are the most critical windows of e

The development of an extended normothermic ex vivo reperfusion model of the sheep uterus to evaluate organ quality after cold ischemia in relation to uterus transplantation.

INTRODUCTION: Uterus transplantation has recently proved that infertility in women with uterine factor infertility can be cured. It is still an experimental procedure with numerous critical details remaining to be established, including tolerance to warm and cold ischemic insults. In preparation for human uterus transplantation trials, most teams use the sheep as a model system for research and team training, since the vasculature and the uterus is of similar size as in the human. We, therefore, aimed to develop an ex vivo sheep uterus reperfusion platform that mimics the reperfusion situation

APOE ε3 attenuates experimental autoimmune neuritis by modulating T cell, macrophage and Schwann cell functions.

Human apolipoprotein E (apoE) is a 34.2kDa glycosylated protein with three isoforms (apoE2, apoE3 and apoE4). Experimental autoimmune neuritis (EAN), an animal model for human Guillain-Barré syndrome, is an immune-mediated experimental disorder of the peripheral nervous system (PNS). Increased susceptibility to EAN in apoE deficient mice has been previously found. To elucidate the isoform-dependent effects of apoE on EAN, we used human apoE2, E3 and E4 transgenic mice (Tg) immunized with P0 peptide 180-199, as well as T cell proliferation test, macrophage and Schwann cell (SC) cultures to inve

[Invasive and non-invasive studies of the protective action of a silicon-containing cream and its excipient in skin irritation induced by sodium laurylsulfate].

The purpose of our study was to evaluate the protective effect of a new silicone-containing barrier-cream (Anthydro) and its vehicle (Anthydro without silicone) in the prevention of cutaneous irritation by detergents. We therefore planned a study in several stages, using an anionic surfactant well known for its irritant properties: sodium laurylsulphate (SLS) in aqueous solution. In a first series of experiments, the protective effect of the Anthydro cream against SLS was studied by invasive methods on guinea-pigs in order to determine histologically the protective effect of the cream when a 1

The rat as a novel model for chronic rotator cuff injuries.

Chronic rotator cuff injuries (CRCIs) still present a great challenge for orthopaedics surgeons. Many new therapeutic strategies are developed to facilitate repair and improve the healing process. However, there is no reliable animal model for chronic rotator cuff injury research. To present a new valuable rat model for future chronic rotator cuff injuries (CRCIs) repair studies, and describe the changes of CRCIs on the perspectives of histology, behavior and MRI. Sixty male Wistar rats were enrolled and underwent surgery of the left shoulder joint for persistent subacromial impingement. They

Safety and efficacy of pegylated recombinant human granulocyte colony-stimulating factor during concurrent chemoradiotherapy for small-cell lung cancer: a retrospective, cohort-controlled trial.

OBJECTIVE: To investigate pegylated recombinant human granulocyte colony-stimulating factor (PEG-rhG-CSF) safety and efficacy in preventing hematological toxicity during concurrent chemoradiotherapy (CCRT) for small-cell lung cancer (SCLC). METHODS: We retrospectively assessed 80 SCLC patients treated with CCRT from January 2013 to December 2018 who received PEG-rhG-CSF within 48 hours after the end of chemotherapy, defined as prophylactic use, as the experimental group. An additional 80 patients who were not treated with PEG-rhG-CSF were matched 1:1 by the propensity score matching method an

Guidelines for the experimental design of pharmacokinetic studies with nanomaterials in preclinical animal models.

A shared feature in the value proposition of every nanomaterial-based drug delivery systems is the desirable improvement in the disposition (or ADME) and pharmacokinetic profiles of the encapsulated drug being delivered. Remarkable progress has been made towards understanding the complex and multifactorial relationships between pharmacokinetic profiles and nanomaterial physicochemical properties, biological interactions, species physiology, etc. These advances have fuelled the rational design of numerous nanomaterials with long-circulation times and improved tissue accumulation (e.g., in tumou

Determination of bleb capsule porosity with an experimental glaucoma drainage device and measurement system.

IMPORTANCE: Control of intraocular pressure after implantation of a glaucoma drainage device (GDD) depends on the porosity of the capsule that forms around the plate of the GDD. OBJECTIVE: To compare capsular porosity after insertion of 2 different GDDs using a novel implant and measurement system. DESIGN, SETTING, AND SUBJECTS: We performed an experimental interventional study at an eye research facility in a tertiary eye care center. Testing was performed on 22 adult New Zealand white rabbits that received the experimental GDD or an existing GDD. INTERVENTIONS: A new experimental GDD, the

Fine Structural Features of Complex InDels and NHEJ Repair at Naturally Occurring Damage Sites in Normal Human Colon Crypts.

DNA repair in biochemical and genetic experimental systems permits a precise definition of enzyme requirements and mechanistic steps. Comparing these findings to repair events at naturally occurring damage sites in multicellular organisms is essential for confirming and expanding these insights into a physiologic context. However, heterogeneity in any normal cell population increases with each cell division, and the reliable detection of replication-independent DNA damage sites and their repair has been a major barrier. Here, we examine single human colon crypts, which harbor natural cell clon

Research on Carotid Blood Flow Detection Based on Interferometric Diffuse Speckle Contrast Analysis and Its Application in Preventing Ischemic Stroke.

This study applied the Interferometric Diffusion Speckle Contrast Analysis (iDSCA) system developed by our research group to detect changes in common carotid artery blood flow. Through animal experiments combining color Doppler ultrasound and human in vivo experiments integrating Transcranial Doppler, the iDSCA system was validated as a technical modality for early warning of ischemic stroke. The experimental results show that in animal experiments, the average relative blood flow index change rate detected by iDSCA after stenosis is -56.66%, which is consistent with the detection result of co

Molecular mechanism of binding selectivity in four stereoisomeric europium complexes with human serum albumin.

A series of stereoisomeric europium complexes displays stereoselectivity in binding to the major human transport protein human serum albumin, (HSA). Details of the nature of the binding interaction between two pairs of chiral Eu(III) complexes and drug site 2 in HSA have been investigated using molecular modelling calculations. Ensemble docking and refinement were carried out in all-atom molecular dynamics simulations, allowing an assessment of the salient binding poses. This approach allowed a comparison with experimental data and was able to rationalise relative binding free energies, and tr

A Tissue-Engineered In Vitro Intestinal Wall Model for Assessing the Immunomodulatory Activity of the Human Gut Microbiota.

The development of in vitro models for screening the immunological activity of commensal bacteria in the human microbiota is a promising and highly relevant field of tissue engineering and experimental microbiology. This article describes a technology for fabricating a standardized, accessible, and easy-to-implement tissue-engineered in vitro model of the human intestinal wall. This model reproduces the physiologically relevant ratio of absorptive and mucin-producing enterocytes, while ensuring the spatial distribution and interaction of enterocytes and immune cells without the need for xenoge

The Mouse Model of Tuberculosis: Uses and Abuses.

The mouse provides a flexible tool to probe the interactions between the immune system and the slow growing and persistent bacterium, Mycobacterium tuberculosis (Mtb). Removal of specific cells or molecules from mice demonstrates the causal connection between these elements and control of Mtb; these connections are also seen in humans. Systems genetics, with single-cell and spatial technologies, have transformed the working model of immunity to tuberculosis (TB) in mice and humans; determining the spatiotemporal role of immune elements in immunity will allow development of targeted and precise

Whole-body in vivo MPI cytometry reveals injection route-, dose-, cell size-, and disease-dependent differences in organ distribution.

Robust quantification of the whole-body biodistribution of injected therapeutic cells remains a major challenge. We performed in vivo cytometry using magnetic particle imaging (MPI) to track magnetically labeled cells on a timescale of minutes with high sensitivity, zero background signal, and simple linear quantification. Human mesenchymal stem cells (hMSCs; ~25 micrometers) and human neural precursor cells (~10 micrometers) were labeled with superparamagnetic iron oxide nanoparticles and mapped with MPI after intravenous or intra-arterial injection in normal mice. The tissue site and numbers

Computing aesthetic taste.

Recent breakthroughs in computational and experimental techniques have produced major discoveries in the science of aesthetic taste. This paper reviews key findings from two complementary approaches: machine learning and large-scale human experiments. We argue that together these approaches represent a fundamental shift in computing aesthetic taste, from measuring preferences and choices to reconstructing the complex, high-dimensional representations that generate them. Using these representations, which we refer to as 'taste spaces', researchers can move beyond asking what people like to expl

Single-trial characterization of frontal theta and parietal alpha oscillatory episodes during spatial navigation in humans.

Theoretical proposals and empirical findings both highlight the relevance of theta brain oscillations in human spatial navigation. However, whilst the general assumption is that the relevant theta band activity is purely oscillatory, most empirical studies fail to disentangle oscillatory episodes from wide band activity. In addition, experimental approaches often rely on averaged activity across trials and subjects, disregarding moment-to-moment fluctuations in theta activity, contingent on key aspects of the task. Here, we used novel oscillation detection approaches to investigate the dynamic
