A pilot study of noradrenergic and HPA axis functioning in PTSD vs. panic disorder.

The biological literature in the anxiety disorders has focused on comparisons between patient groups and normal volunteers, with relatively little comparative study of the anxiety disorders. We therefore conducted this pilot study to compare a group of patients with post-traumatic stress disorder (PTSD) (n = 7) to a contiguously studied panic disorder group (n = 17) and healthy control subjects (n = 16) on baseline levels of cortisol and 3-methoxy-4-hydroxyphenylglycol (MHPG), and response to clonidine challenge. Despite the small sample size, highly significant differences were found on the f

Acute exposure to a novel stressor further reduces the habituated corticosterone response to restraint in rats.

The present study sought to identify dishabituation of the hypothalamic-pituitary-adrenal(HPA) axis response to different psychological stressors. Young adult male Sprague Dawley rats were exposed to five, 1 h sessions of restraint stress on five consecutive days. On the sixth day, and 2 h before additional exposure to restraint, animals were subjected to 30 min of a small (27cm square), elevated open field stressor (pedestal), which served as the dishabituating stimulus. We predicted HPA axis response dishabituation in chronically restrained rats exposed to the novel pedestal. Rats which unde

The neurobiology of social phobia.

Although Social Phobia has been recognised for centuries in comparison with other anxiety disorders, relatively little work has been done to understand its neural basis. The present review attempts to redress this balance by giving an overview of the current state of knowledge in this disorder. By putting together data from the treatment responses to specific agents, the effects of chemical challenges which have been used in other anxiety disorders and by reviewing data on central and peripheral neurotransmitter and endochrine abnormalities, it is possible to begin to generate some potentially

Effect of fetal ethanol exposure on the in vitro release of growth hormone, somatostatin and growth hormone-releasing factor induced by clonidine and growth hormone feedback in male and female rats.

This study was designed to examine the effect of fetal ethanol (ETOH) exposure on the sensitivity of the hypothalamic-growth hormone (GH) axis to clonidine (an alpha 2-adrenoreceptor agonist) stimulation and GH feedback. During gestation, dams were fed either a liquid diet in which 36% of the calories were derived from ETOH, or pair-fed an isocaloric control liquid diet without ETOH. A second set of controls were fed lab chow ad libitum. After birth, offspring of ETOH-fed dams were cross-fostered to a separate group of ad libitum control dams. The hypothalami and pituitaries of 10-, 20-, 30-,

cAMP regulation of vasopressin mRNA content in hypothalamo-neurohypophysial explants.

Stimulation of vasopressin (VP) gene expression by adenosine 3',5'-cyclic monophosphate (cAMP) has been observed in dispersed hypothalamic cultures, in VP-expressing cell lines, and in cells transfected with reporter genes regulated by the VP gene promoter. However, treatment of hypothalamo-neurohypophysial system (HNS) explants with forskolin (25 microM), an activator of adenyl cyclase, and 3-isobutyl-1-methylxanthine (IBMX; 500 microM), a phosphodiesterase inhibitor, resulted in a decrease in VP mRNA. Time course analysis revealed that IBMX and forskolin reduced the VP mRNA content to 50% of

Stimulation of the hypothalamo-pituitary-adrenal axis in the rat by the type 4 phosphodiesterase (PDE-4) inhibitor, denbufylline.

1. Preliminary studies in our laboratories showed that the synthetic xanthine analogue denbufylline, a selective type 4 phosphodiesterase (PDE-4) inhibitor, is a potent activator of the hypothalamo-pituitary-adrenal (HPA) axis when given orally to adult male rats. This paper describes the results of experiments in which well established in vivo and in vitro models were used to (a) examine further the effects of denbufylline on HPA function and (b) identify the site and mode of action of the drug within the axis. 2. In vivo, administration of denbufylline (0.1-2.5 mg kg-1, i.p.) produced a sign

Human corticotropin-releasing hormone test in patients with hypothalamo-pituitary-adrenocortical disorders.

To assess the diagnostic usefulness of a synthetic human corticotropin-releasing hormone (CRH) formulation (Code No. MCI-028), we administered 100 micrograms of the peptide intravenously to 183 patients with hypothalamo-pituitary-adrenocortical (HPA) disorders, and obtained the following findings. Among the 183 patients, data from 125 patients were suitable for analyzing the effects of the test. In patients with Cushing's disease, high plasma adrenocorticotropic hormone (ACTH) and cortisol levels increased further in response to MCI-028, while in patients with adrenal Cushing's syndrome, low A

The role of the adrenoceptors in the activation of the hypothalamic-pituitary-testicular complex of mice induced by the presence of a female.

A study was made of the role of the adrenergic processes in the activation of the testes endocrine function in male mice of CBA/Lac and A/He strains after their sexual arousal induced by the presence of a sexually receptive female without any tactile contact with a male. The alpha-adrenoceptor blocker phentolamine and the alpha 1-adrenoceptor antagonist prazosin inhibited an increase of testosterone level in the peripheral blood plasma caused by the presence of a female. In contrast, antagonist of the beta-adrenoceptor propranolol and beta 2-adrenoceptor blocker ICI-118.551 significantly inten

Loneliness, positive social interactions, and diurnal cortisol among mid-to-later life adults in everyday life.

Loneliness, the subjective feeling of social isolation, is a major public health concern linked to poor health, including stress-related diseases such as cardiovascular disease. However, less is known about how loneliness relates to biological functioning in everyday life and whether age and social interactions shape these associations. We investigated whether loneliness self-reported in everyday life related to the dynamic range of diurnal cortisol (CDR) compared to other indicators of diurnal cortisol (including the cortisol awakening response [CAR], diurnal cortisol slope [DCS], and area un

Corticosteroid-induced gene expression is attenuated in the hippocampus of mice with model of post-traumatic stress disorder.

One of the distinctive features of post-traumatic stress disorder (PTSD) is an impairment of the hypothalamic-pituitary-adrenal (HPA) axis. Increased inhibition of cortisol synthesis after dexamethasone administration in patients indicates hyperactivation of the HPA feedback loop. This phenomenon may be explained by increased sensitivity of hypothalamic CRH-producing neurons to corticosteroids. Since corticosteroids signaling influences memory processing mechanisms, the issue of hippocampal glucocorticoid (GR) and mineralocorticoid (MR) receptors sensitivity is important for the pathogenesis o

Stress-Free Voluntary Exercise Promotes Prophylactic Enhancement of Stress Resilience via the Nucleus Reuniens Affecting the Medial Prefrontal Cortex-Hippocampal Pathway.

PURPOSE: Stress is a risk factor for psychiatric illnesses. However, not all individuals exposed to stress will develop affective disorders. We examined whether pretreatment with stress-free voluntary wheel running (VWR) exercise prophylactically enhances stress resilience in rodents and how it can effectively prevent the development of depressive- and anxiety-like behaviors. METHODS: Eight-week-old C57BL6/J mice were housed in cages with VWR and subjected to chronic restraint stress (CRST) for 2 h daily for 14 d. The mice were assessed for depressive- and anxiety-like behaviors, and a behavi

The Investigation of Vitamin D and Menstrual Cycles Trial (the inVitD Trial): A clinical trial of vitamin D supplementation on the hypothalamic-pituitary-ovarian axis.

BACKGROUND: While there is evidence that vitamin D deficiency is associated with long menstrual cycles, delayed ovulation, and reduced fertility, it is yet unknown if increasing vitamin D levels can regulate menstrual cycles, and thus possibly improve fertility. The primary objective of this randomized clinical trial (RCT) is to test the hypothesis that vitamin D supplementation influences the hypothalamic-pituitary-ovarian axis. METHODS: This two-site RCT (NCT05050916) required participants to be aged 19-40 years old, having spontaneous menstrual cycles, and without certain chronic diseases

Perinatal running training reversed postnatal anxiety and depressive-like behavior and cognitive impairment in mice following prenatal subchronic variable stress.

BACKGROUND: Pregnancy is a very complex and highly stressful time in women. Despite the high prevalence of postpartum depression, more than 50 % of mothers are undiagnosed or untreated, showing an urgent need to explore an effective preventive strategy. Regular physical activity has been suggested to be associated with an increased quality of life in pregnant and postpartum women. The purpose of this study was to determine whether perinatal running training can affect maternal care stress-related anxiety, depressive-like behavior, and cognitive changes in postpartum dams and to explore the pos

The upregulation of glutamate decarboxylase 67 against hippocampal excitability damage in male fetal rats by prenatal caffeine exposure.

As a kind of xanthine alkaloid, caffeine is widely present in beverages, food, and analgesic drugs. Our previous studies have shown that prenatal caffeine exposure (PCE) can induce programmed hypersensitivity of the hypothalamic-pituitary-adrenal (HPA) axis in offspring rats, which is involved in developing many chronic adult diseases. The present study further examined the potential molecular mechanism and toxicity targets of hippocampal dysfunction, which might mediate the programmed hypersensitivity of the HPA axis in offspring. Pregnant rats were intragastrically administered with 0, 30, a

Altered endocannabinoid-dynamics in craniopharyngioma patients and their association with HPA-axis disturbances.

OBJECTIVE: Patients with craniopharyngioma (CP) frequently suffer from morbid obesity. Endocannabinoids (ECs) are involved in weight gain and rewarding behavior but have not been investigated in this context. DESIGN: Cross-sectional single-center study. METHODS: Eighteen patients with CP and 16 age- and sex-matched controls were included. Differences in endocannabinoids (2-arachidonoylglycerol (2-AG) and N-arachidonoylethanolamine (AEA)) and endocannabinoid-like molecules (oleoyl ethanolamide (OEA), palmitoylethanolamide (PEA), and arachidonic acid (AA) were measured at baseline and followin

Reduced expression of the glucocorticoid receptor in the hippocampus of patients with drug-resistant temporal lobe epilepsy and comorbid depression.

OBJECTIVE: Depressive disorders are common among about 50% of the patients with drug-resistant temporal lobe epilepsy (TLE). The underlying etiology remains elusive, but hypothalamus-pituitary-adrenal (HPA) axis activation due to changes in glucocorticoid receptor (GR) protein expression could play an important role. Therefore, we set out to investigate expression of the GR in the hippocampus, an important brain region for HPA axis feedback, of patients with drug-resistant TLE, with and without comorbid depression. METHODS: GR expression was studied using immunohistochemistry on hippocampal s

Dammarane sapogenins alleviates depression-like behaviours induced by chronic social defeat stress in mice through the promotion of the BDNF signalling pathway and neurogenesis in the hippocampus.

Chronic social defeat stress (CSDS) is a widely used behavioural paradigm of psychosocial stress that can be used to research the pathogenesis of depression and seek antidepressant drugs. Dammarane sapogenins (DS), the deglycosylated product of ginsenosides, has a wide range of biological activities, including immunomodulatory, antifatigue, antitumour and antidepressant activities. However, whether DS has antidepressant-like effects in a CSDS mouse model remains unknown. Therefore, the present study was conducted to evaluate the antidepressant properties of DS in CSDS mice and its underlying m

GnRH in the Human Female Reproductive Axis.

Gonadotropin-releasing hormone (GnRH) is recognized as the central regulator of the functions of the pituitary-gonadal axis. The increasing knowledge on the mechanisms controlling the development and the function of GnRH-producing neurons is leading to a better diagnostic and therapeutic approach for hypogonadotropic hypogonadisms and for alterations of the puberty onset. During female life span, the function of the GnRH pulse generator may be affected by a number of inputs from other neuronal systems, offering alternative strategies for diagnostic and therapeutic interventions. Moreover, the
