Polysaccharides and virulence of Burkholderia pseudomallei.

Burkholderia pseudomallei is the causative agent of melioidosis, an infectious disease of humans and animals. Gene clusters which encode capsular polysaccharide (type I O-PS) and LPS (type II O-PS), both of which play roles in virulence, have previously been identified. Here, the identification of two further putative clusters, type III O-PS and type IV O-PS, is reported. Mice challenged with type III O-PS or type IV O-PS mutants showed increased mean times to death (7.8 and 11.6 days) compared to those challenged with wild-type B. pseudomallei (3 days). To investigate the possible roles of po

MVA-LACK as a safe and efficient vector for vaccination against leishmaniasis.

An optimal vaccine against leishmaniasis should elicit parasite specific CD4+ and cytotoxic CD8+ T cells. In this investigation, we described a prime/boost immunization approach based on DNA and on poxvirus vectors (Western Reserve, WR, and the highly attenuated modified vaccinia virus Ankara, MVA), both expressing the LACK antigen of Leishmania infantum, that triggers different levels of specific CD8+ T cell responses and protection (reduction in lesion size and parasitemia) against L. major infection in mice. A prime/boost vaccination with DNA-LACK/MVA-LACK elicits higher CD8+ T cell respons

Adult tetanus, diphtheria and pertussis immunization: knowledge, beliefs, behavior and anticipated uptake.

BACKGROUND: Lifetime protection against pertussis has been adopted as a goal of immunization programs in Canada. To anticipate adult coverage with a combined product containing tetanus (T) and diphtheria (d) toxoids and acellular pertussis (aP) vaccine as a booster dose, we conducted a survey of households in British Columbia, Canada. METHODS: In a random telephone survey involving 800 adults, 25 years of age and older, we assessed current behaviors related to adult Td immunization and beliefs regarding pertussis vaccine under various scenarios relevant to adult decision-making. RESULTS: For

Immunogenicity of a recombinant MVA and a DNA vaccine for Japanese encephalitis virus in swine.

We previously reported that mice immunized with recombinant modified vaccinia virus Ankara (MVA) encoding Japanese encephalitis virus (JEV) prM and E genes were completely protected against JEV challenge (Nam, J.H., Wyatt, L.S., Chae, S.L., Cho, H.W., Park, Y.K., Moss, B. Vaccine 1999,17: 261-268). In this study, we examined the immunogenicity in swine of this recombinant MVA (vJH9) or a DNA vaccine (pcJH-1) expressing the same JEV genes. Although the booster effect in mice with a combination of vJH9, pcJH-1 and inactivated JEV commercial vaccine was not apparent by measuring JEV antibodies, t

The effect of Staphylococcus aureus phage lysate vaccine on a rabbit model of staphylococcal blepharitis, phlyctenulosis, and catarrhal infiltrates.

PURPOSE: To evaluate the effect of Staphylococcus aureus phage lysate (SPL) vaccination on the development of blepharitis, corneal phlyctenules, and catarrhal infiltrates and on the development of antibodies and the delayed-type hypersensitivity response to S. aureus. METHODS: Eighty rabbits received an intradermal immunization of cell wall-complete Freund's adjuvant followed by a booster immunization. Rabbits were given topical applications of viable S. aureus in both eyes and 40 rabbits received subcutaneous SPL vaccinations. Clinical observations were made weekly. An enzyme-linked immunoso

[An evaluation of the epidemiological and immunological efficacy of the vaccinal prophylaxis of diphtheria and tetanus].

The extensive use of adsorbed diphtheria-tetanus toxoid with reduced antigen content was found to be ineffective in the prophylaxis of diphtheria: 86.3% of diphtheria patients among those who had been immunized with this preparation fell only a year after the first booster immunization, and the proportion of those who proved to be unprotected against diphtheria on years 3, 4 and 5 after immunization reached, respectively, 21.0%, 35.5% and 49.4%. The number of children immunized with this preparation at common preschool institutions may reach 50% and more, and with an increase in the coverage o

[Kinetics of vaccine antibodies to tetanus toxoid, diphtheria toxoid, measles virus, poliomyelitis virus and pneumococci after allogenic and autologous bone marrow transplantation and booster immunization. 1: The kinetics of vaccine antibodies to tetanus toxoid after allogenic and autologous bone marrow transplantation].

Today BMT belongs to the established methods of treatment in haematology and oncology. Because of the constant increase of healthy long-term survivors after BMT the problem of immunological reconstitution and eventual possible late effects gets more and more importance. One problem, which til now has been few attention paid to, is that of the protection by vaccination after BMT. We report on the kinetics of the tetanus-antitoxin in 20 patients after allogeneic or autologous BMT and demonstrate the influence of a graft-versus-host disease and its therapy on the antibody kinetics. In the group o

Protective effect of inactivated bovine herpesvirus-1 in calves experimentally infected with bovine herpesvirus-1 and Pasteurella haemolytica.

The protective effect of an inactivated whole-virion bovine herpesvirus-1 (BHV-1) immunising inoculum, without adjuvant, against viral-bacterial respiratory disease was studied in three experimental treatment groups of five calves each. One group was boosted 14 days after the first vaccination and at this time the second group received their initial inoculation. Seven days later, calves were challenged with BHV-1 in aerosol and four days after this challenge all calves were exposed to Pasteurella haemolytica A1 in aerosol. Among the three groups, differences in rectal temperature responses fou

Genetic predisposition to porto-sinusoidal vascular disorder.

Porto-sinusoidal vascular disorder (PSVD) is a rare liver disease. The pathophysiological mechanisms underlying the development of PSVD are unknown. Isolated cases of PSVD associated with gene mutations have been reported, but no overview is available. Therefore, we performed an extensive literature search to provide a comprehensive overview of gene mutations associated with PSVD. We identified 34 genes and 1 chromosomal abnormality associated with PSVD in the literature, and we describe here 1 additional gene mutation ( TBL1XR1 mutation, leading to Pierpont syndrome). These gene mutations are

Immune cell clustering identifies a CD163⁺/CSF1R⁺ macrophage and neutrophil-enriched phenotype with distinct biological signatures and poor prognosis in angiosarcoma.

The tumor immune microenvironment (TIME) influences tumor biology and therapeutic response, yet its composition and clinical relevance in angiosarcoma, an ultrarare and aggressive sarcoma with a dismal prognosis, remain poorly defined. To address this gap, we comprehensively profiled the TIME of 63 angiosarcomas (AS), including primary cutaneous, secondary, and visceral subtypes, with a focus on colony-stimulating factor 1 receptor (CSF1R)-expressing tumor-associated macrophages (TAMs). Using conventional and multiplex immunohistochemistry with quantitative analysis, we found that CD68⁺/CD163⁺

Coordination-Stabilized Manganese(IV) Peptide Hydrogels Reveal Valence-Dependent Structure and Immunomodulation.

High-valence manganese is typically confined to solid oxide precursors in manganese-based biomaterials, leaving valence-dependent coordination-structure-function relationships largely unresolved. Here, we established a peptide-coordinated hydrogel platform that stabilized Mn(IV) in an aqueous/gel environment and enabled direct comparison between Mn(II)- and Mn(IV)-dominated supramolecular networks. By tuning FE tandem repeats, two representative systems, FE3-Mn(II) and FE4-Mn(IV), were constructed with distinct manganese valence distributions and coordination environments. These valence-define

Gut-liver axis molecular mechanisms in alcohol-associated liver disease.

Alcohol-associated liver disease (ALD) represents one of the most prevalent and devastating chronic liver diseases worldwide. Yet, its pathogenesis and effective therapeutic strategies remain elusive. The gut-liver axis, the bidirectional interaction between intestinal microbiota, microbial products, metabolites, bile acids, immune mediators, and the liver itself, has become a key focus in the study of ALD. Chronic alcohol consumption disrupts intestinal homeostasis, leading to microbial dysbiosis, compromised barrier integrity, and translocation of pathogen-associated molecular patterns (PAMP

Narcolepsy as an immune-associated hypothalamic encephalopathy: orexin dysfunction and implications for precision sleep medicine.

Narcolepsy can no longer be adequately conceptualized by excessive sleepiness and cataplexy. It is increasingly recognized as a multisystem hypothalamic encephalopathy, rooted in the selective loss or dysfunction of orexin neurons, yet extending across motor, psychiatric, metabolic, and autonomic domains. Over the past two decades, convergent genetic, neuropathological, and immunological evidence has positioned narcolepsy type 1 as increasingly consistent with the spectrum of immune-mediated neurological diseases while challenging the validity of current classifications that hinge on cataplexy

The leaky gut and microbiome in critical illness: emerging insights into microbial "translocation".

PURPOSE OF REVIEW: Microbial translocation has long been viewed as a nonspecific consequence of gut barrier failure in critical illness, with downstream effects attributed primarily to the host immune response. This review examines emerging evidence that the gut microbiome plays a more active and specific role in this process than previously appreciated. RECENT FINDINGS: Dysbiosis during critical illness directly contributes to barrier dysfunction through depletion of metabolites that sustain epithelial integrity. Culture-independent approaches have revealed that gut-derived organisms are a m

Multidisciplinary Surgical Treatment of Hepatic Abscess in a Geriatric Dog with Congenital Extrahepatic Portosystemic Shunt.

Hepatic abscesses are uncommon in dogs and typically develop secondary to biliary tract disease or ascending bacterial infections. Although congenital extrahepatic portosystemic shunt (EHPSS) is known to impair hepatic perfusion and immune clearance, its potential role in predisposing geriatric dogs to hepatic abscess formation has not been previously reported. This case report describes the diagnostic approach, therapeutic decision-making, and clinical outcome of a geriatric dog in which a multidrug-resistant hepatic abscess occurred in association with congenital EHPSS, and to propose a path

Sinomenine ameliorates experimental autoimmune neuritis by suppressing pro-inflammatory immune components and restoring immune balance.

Sinomenine (SIN), a bioactive alkaloid derived from Sinomenium acutum, exhibits well-documented anti-inflammatory and analgesic properties with a favorable safety profile. Here, we pharmacologically characterized the preventive and therapeutic efficacy of SIN in a rat model of experimental autoimmune neuritis (EAN), an established model of immune-mediated peripheral neuropathy. Although SIN has been evaluated in several neuroinflammatory disorders, its mode of action in EAN has not been systematically addressed. Disease progression, body-weight changes, and neurological scores were monitored f

Risk factors for EBV reactivation and impact of graft composition following GVHD prophylaxis with rabbit ATG or PTCy.

BACKGROUND: EBV reactivation is frequent after allogeneic HCT and can lead to post-transplant lymphoproliferative disorder (PTLD). While ATG and PTCy are established strategies to prevent GVHD, their comparative impact on EBV reactivation remains insufficiently characterized. METHODS: We conducted a retrospective study in patients who received either rabbit ATG or PTCy. Primary endpoints were incidences of positive and high-level EBV DNAemia (≥10⁴ copies/mL). Secondary endpoints were incidence of PTLD, predictive risk factors for EBV reactivation, GVHD, survival, immune reconstitution and res

Antigen-specific B cell responses in treatment-naive and chlorambucil-treated patients with chronic lymphocytic leukemia.

Chronic lymphocytic leukemia (CLL) is characterized by secondary immunodeficiency, yet the extent to which global disease-associated immune dysregulation impairs the ability to generate antigen-specific B cell immunity—independent of targeted therapy—remains unclear. To address this, we assessed the non-malignant B cell compartment and responses to SARS-CoV-2 vaccination in treatment-naive (CLL-TN) and chlorambucil-treated (CLL-CTx) patients. Despite marked alterations in peripheral B cell subsets, both CLL-TN and CLL-CTx patients exhibited higher absolute numbers of SARS-CoV-2 spike-specific

The calcium-sensing receptor in chronic kidney disease-mineral and bone disorder: Structural biology, biased signaling, osteoimmune crosstalk, and therapeutic implications.

The calcium-sensing receptor (CaSR) is a Family C G protein-coupled receptor (GPCR) that functions as a principal sensor of extracellular calcium ions ([Ca2+]o) and a broad-spectrum integrator of polyanionic, amino acid, and ionic stimuli. Its structural architecture - encompassing a bilobed Venus flytrap domain (VFT), cysteine-rich domain (CRD), heptahelical transmembrane domain (TMD), and an intrinsically disordered intracellular C-terminal tail - enables a remarkable allosteric landscape that goes far beyond simple on/off switching. CaSR operates as an obligate homodimer stabilized by inter

Cryptosporidiosis in inborn errors of immunity: a single center cohort and review of the literature.

BACKGROUND: Severe intestinal or hepatobiliary cryptosporidiosis affects patients with inborn errors of immunity (IEI), particularly those with CD40 ligand or CD40 deficiency, major histocompatibility complex class II deficiency, interleukin-21 receptor deficiency, and DOCK8 deficiency. Susceptibility to Cryptosporidium reflects defects in lymphocyte development, activation, and effector function. METHODS: We conducted a retrospective chart review of IEI patients with cryptosporidiosis, focusing on 17 individuals with chronic infection who underwent hematopoietic cell transplant (HCT), to cha
