Acetyl-l-carnitine: a pathogenesis based treatment for HIV-associated antiretroviral toxic neuropathy.

BACKGROUND: Nucleoside analogue reverse transcriptase inhibitors (NRTI) disrupt neuronal mitochondrial DNA synthesis, impairing energy metabolism and resulting in a distal symmetrical polyneuropathy (DSP), an antiretroviral toxic neuropathy (ATN) that causes significant morbidity in HIV disease. Serum acetyl-l-carnitine (ALCAR) levels are decreased in neuropathy associated with NRTI therapy. ALCAR enhances neurotrophic support of sensory neurons and promotes energy metabolism, potentially causing nerve regeneration and symptom relief. OBJECTIVE: To assess the efficacy of oral ALCAR (1500 mg t

alpha-Sarcoglycan (adhalin) deficiency: complete deficiency patients are 5% of childhood-onset dystrophin-normal muscular dystrophy and most partial deficiency patients do not have gene mutations.

alpha-Sarcoglycan (adhalin), a 50-kDa component of the dystrophin-associated complex of proteins, participates in the stabilization of the myofiber plasma membrane in the membrane cytoskeleton. Deficiencies of alpha-sarcoglycan cause a subset of childhood-onset muscular dystrophy (SCARMD) cases. However, secondary deficiencies of alpha-sarcoglycan are common. To begin to establish the rates of false positives (secondary deficiencies), we used immunofluorescence to screen 30 Italian dystrophin-normal muscular dystrophy patient biopsies and identified 4 patients with partial alpha-sarcoglycan de

Metagenomic analysis of rhizosphere soil microbiota in wild and cultivated Notopterygium incisum, an umbelliferae medicinal herb.

Notopterygium incisum is not only a traditional Chinese medicine but also an endemic herb. Artificial domestication and large-scale cultivation are crucial for resolving the crisis of wild resources and the supply-demand imbalance of N. incisum, yet current techniques have failed to stably provide the herb medicine in good quality. Metagenomic analyses revealed significant differences in the rhizomicrobiota between wild and cultivated N. incisum, particularly in microbial composition, gene functions, and community assembly. The rhizomicrobiota of the wild N. incisum from 3 different sites with

Cyperotundone Regulates Polo-like Kinase 1-mediated Glycolysis to Inhibit the Proliferation of Breast Cancer Cells.

Introduction Cyperotundone (CYT), a vital component of Cyperus rotundus L., is known to suppress tumor growth effectively. This investigation was performed to reveal the antibreast cancer(BC) effect of CYT and its mechanism. Methods CCK-8 assay, colony formation, cell cycle/apoptosis assays, and subcutaneous tumor model were used to investigate the anticancer effects of CYT. RNA sequencing was used to predict the targets of CYT, which were then verified by drug affinity responsive target stability (DARTS), cellular thermal shift assay (CETSA), and molecular docking. The pan-cancer analysis on

Identification of METTL3 and PRMT5 as an oncogenic pair for the prognosis and therapeutic targets of colorectal cancer subtype.

BACKGROUND: METTL3 is a pivotal N6-methyladenosine (m⁶A) regulator that is linked to a worse prognosis of colorectal cancer (CRC). However, METTL3's oncogenic molecular networks in CRC remain unclear. This study aims to establish a co-expression network of METTL3 and identify novel diagnostic and therapeutic targets for CRC. METHODS: We integrated transcriptomic datasets with clinical cohorts and patient-derived organoids (PDOs) to evaluate the expression profiles of METTL3 and PRMT5. Mechanistic insights were gained through co-immunoprecipitation (Co-IP), m⁶A-methylated RNA immunoprecipitati

Comparative evaluation of reference-free transcriptomic deconvolution highlights the importance of biological validation in astrocytes across Alzheimer's disease.

INTRODUCTION: Astrocytes are central regulators of neuronal energy metabolism and redox homeostasis-processes that become progressively disrupted across the Alzheimer's disease (AD) continuum. However, bulk transcriptomic data obscure cell-type-specific signals, and existing reference-free deconvolution methods often prioritize either statistical robustness or quantitative accuracy without fully integrating both dimensions. METHODS: Here, we present a comparative framework evaluating two complementary unsupervised approaches, CDSeq and DECODER, to reconstruct astrocyte-associated transcriptom

HBO1 suppresses anti-tumor immunity of CD8⁺ T Cells in thyroid cancer by recruiting BRD4 to regulate VTCN1 expression.

Thyroid cancer is the most common endocrine malignancy. Although surgery remains the primary treatment, advanced disease frequently recurs or metastasizes, and subsequent therapies yield suboptimal outcomes. Immunotherapy holds promise, yet the underlying immune-evasion mechanisms remain incompletely understood. HBO1, a multifunctional acyltransferase, plays a significant role in tumor immune regulation. Our previous research revealed that HBO1 is highly expressed in thyroid cancer tissues, promotes malignant progression, and inhibits CD8+ T cell function. To further elucidate the role and mec

Transcriptomic changes in embryonic blood of subsequent generations following in ovo stimulation with nutriepigenetic factors.

BACKGROUND: Epigenetic modifications regulate gene expression and are influenced by environmental factors, shaping phenotypic and clinical outcomes. These changes have the potential to persist across generations, although their stability may vary between tissues. This study aims to observe, using an in ovo model, the effects of single (F1) and repeated (F1-F3) prenatal stimulation with potential epigenetic factors on the transcriptome of embryonic blood in subsequent generations. METHOD: Using an in ovo model across three generations of Green-legged Partridgelike chickens (with additional F4

CIRP Promotes Redox-Inflammatory Endothelial Injury in High-Fat Diet-Fed ApoE-/- Mice and HUVECs: Involvement of TLR4/SIRT6 Signaling.

Cold-inducible RNA-binding protein (CIRP/CIRBP) is recognized as an extracellular damage-associated molecular pattern. However, its relationship to endothelial redox-inflammatory injury in atherosclerosis remains poorly characterized. Here, we explored the associations among CIRP, endothelial dysfunction, and TLR4-/SIRT6-related changes. In high-fat diet (HFD)-fed ApoE-/- mice, circulating CIRP was elevated and positively correlated with atherosclerotic plaque burden. This increase coincided with systemic redox imbalance, impaired NO/eNOS activity, and vascular inflammation. In HUVECs, CIRP ex

[Construction and characterization of a stable Cas9-expressing monoclonal WSL cell line].

The molecular mechanism underlying the cellular invasion of African swine fever virus (ASFV) remains incompletely understood, particularly with respect to its key cellular receptors, which constitutes a major bottleneck in the development of effective vaccines and targeted antiviral therapies. To establish a robust experimental platform that supports efficient ASFV replication and enables genome-wide CRISPR screening for systematic identification of host factors involved in viral entry, we employed the CRISPR/Cas9 system to generate stable monoclonal cell lines expressing Cas9 protein based on

A 3D-bioprinted osteogenic niche model reveals bone microenvironment-driven malignant phenotypes in prostate cancer.

INTRODUCTION: Prostate cancer (PCa) is one of the most prevalent malignancies in men and frequently progresses to bone metastasis. Understanding the interactions between PCa cells and the bone microenvironment, as well as their impact on therapeutic response, is therefore of critical clinical relevance. OBJECTIVES: This study aimed to investigate the effects of the osteogenic microenvironment on prostate cancer cells by constructing three-dimensional (3D) bioprinted in vitro co-culture models, and to evaluate how the osteogenic niche influences tumor malignant phenotypes. METHODS: This study

ELAVL1 modulates periodontal ligament fibroblast pyroptosis in periodontitis through the JAK2/STAT3/NLRP3 axis.

Periodontitis is an inflammatory disorder that leads to the destruction of periodontal tissues. Pyroptosis, related to the Nod-like receptor protein 3 (NLRP3) inflammasome, is implicated in the pathogenesis of periodontitis. ELAVL1 (embryonic lethal, abnormal vision, Drosophila-like 1), an N6-methyladenine (m6A) reader, is associated with inflammatory responses; however, its role in periodontitis requires further clarification. Our research elucidated the regulatory functions of ELAVL1 in periodontitis and explored the underlying mechanisms. Expression of ELAVL1 in periodontitis mice and Porph

Spatially localized immune metaprograms reveal micro-niche organization in the human Dorsal Root Ganglion.

Dorsal root ganglion (DRG)-targeted local interventions are widely used in pain management; however, the cellular and spatial organization of immune transcriptional states within human DRG tissue remains poorly defined. In particular, it is unclear whether DRG immune activity reflects a single dominant inflammatory axis or a spatially heterogeneous microenvironment composed of parallel transcriptional programs. To address this question, we analyzed publicly available human DRG single-nucleus RNA sequencing data (GSE189501) to identify immune transcriptional metaprograms using rank-stable non-n

Clerodendrum japonicum Extract Suppresses the Pathogenicity of Rheumatoid Arthritis Fibroblast-Like Synoviocytes by Inhibiting METTL3-Mediated ICAM2 mRNA Methylation.

Clerodendrum japonicum (Thunb.) Sweet (Clerodendrum japonicum) is a traditional medicinal plant with reported anti-inflammatory use, but its anti-rheumatoid arthritis (RA) activity and molecular basis remain insufficiently characterized. Activated fibroblast-like synoviocytes (FLSs) contribute to persistent inflammation and joint destruction in rheumatoid arthritis (RA). This study investigated whether extracts of Clerodendrum japonicum attenuate pathogenic RA-FLS phenotypes and examined the possible involvement of METTL3-dependent N6-methyladenosine (m6A) regulation of ICAM2 mRNA. Tumor necro

Transposable elements as a possible missing link between genetic predisposition and environmental triggers of autoimmune disorders: insights from type 1 diabetes, systemic lupus erythematosus and rheumatoid arthritis.

Transposable elements (TEs) make up almost half of the human genome and are among its most densely methylated regions. Their epigenetic silencing is crucial for genomic stability and immune homeostasis, and accumulating evidence indicates that dysregulated TE methylation and expression contribute to autoimmune disease pathogenesis. Hypomethylation of selected TE families can permit transcriptional reactivation, production of immunostimulatory nucleic acids and peptides, and engagement of pattern-recognition receptors, thereby driving type I interferon (IFN-I) signaling through "viral mimicry"-
