Participation of noradrenergic neurotransmission in angiotensin III-induced dipsogenic behavior in the rat.

Conscious, adult, male Sprague-Dawley rats, instrumented with in-dwelling cannula for drug application into the lateral cerebral ventricle, were used to evaluate the participation of noradrenergic neurotransmission in angiotensin III (AIII)-induced dipsogenic behavior. Intracerebroventricular (i.c.v.) administration of AIII (20, 40 or 80 pmol) elicited a robust and dose-related drinking response. Chemical lesion produced by i.c.v. injection of the catecholaminergic neurotoxin, 6-hydroxydopamine (25 micrograms x 3), or the selective noradrenergic neurotoxin, N-(2-chloroethyl)-N-ethyl-2-bromoben

Changes in choline acetyltransferase activity and high-affinity choline uptake, but not in acetylcholinesterase activity and muscarinic cholinergic receptors, in rat somatosensory cortex after sciatic nerve injury.

Selected cholinergic markers (choline acetyltransferase, acetylcholinesterase, muscarinic acetylcholine receptor, high-affinity choline uptake) were studied in the hindlimb representation areas of the rat somatosensory cortex and within the visual cortex 1 to 63 days after unilateral transection of the sciatic nerve. In the contralateral somatosensory cortex, peripheral deafferentation resulted in a significant reduction of choline acetyltransferase activity (by 15%) 3 days after sciatic nerve injury, and in a significant reduction of high-affinity choline uptake (by 30%) 1 day after nerve tra

The endothelium modulates adrenergic neurotransmission to canine pulmonary arteries and veins.

We tested the postulate that endothelium-derived relaxing factor (EDRF) modulates adrenergic neuroeffector transmission in isolated canine pulmonary arteries and veins, using the technique of superfusion and measurement of the efflux of [2-14C]NE during transmural nerve stimulation at 1, 2, 4, 8, 16 and 32 Hz for 10 and 30 min. In endothelium-rubbed artery and vein the contractile responses to low frequency nerve stimulation were enhanced, when compared to those from endothelium rubbed blood vessels. Transmural nerve stimulation of endothelium competent arteries and veins for 10 min released l

GTP-binding proteins mediate the M2-muscarinic effect on the action potential in isolated sympathetic neurons of rabbits.

Activation of M2-muscarinic receptors alters the configuration of the action potential due to depression of the calcium-dependent components, the shoulder in the falling phase and the afterhyperpolarization, in isolated superior cervical ganglionic neurons of rabbits. This effect was inhibited by preincubation of the cells with pertussis toxin, or by the intracellular administration of guanosine 5'-O-(2-thiodiphosphate) (GDP-beta-S). The muscarinic effect persisted in the cells loaded with guanosine 5'-O-(3-thiotriphosphate) (GTP-gamma-S). Intracellular application of cAMP and 3-isobutyl-l-met

Mechanisms of substance P-induced contraction of rabbit airway smooth muscle.

The contractile effects of substance P (SP) were studied in isolated rabbit tracheal smooth muscle (TSM) segments in vitro. Noncumulative administration of SP produced dose-dependent increases in TSM tension. The mean (+/- SE) peak isometric tension (Tmax) with SP was 35.7 (+/- 6.2%) of the corresponding Tmax response to methacholine. The dose of agonist producing 50% of Tmax (ED50) was significantly lower for SP, averaging 1.8 (+/- 0.4) X 10(-7) M, vs. 1.7 (+/- 0.32) X 10(-6) M for methacholine. Blockade of both parasympathetic ganglia with hexamethonium (10(-4) M) and neural transmission wit

Baclofen reduces tone-evoked activity of cochlear nucleus neurons.

Recent evidence suggests that an excitant amino acid may be a neurotransmitter at acoustic nerve synapses in cochlear nucleus (CN). Release of excitant amino acids is reportedly reduced by baclofen, a lipophilic GABA-mimetic used to treat the spasticity of multiple sclerosis and spinal injury. Microiontophoresis of (-)baclofen suppressed spontaneous and tone-evoked activity in CN neurons. GABA inhibited the responses of most neurons responsive to (-)baclofen. However, iontophoresis of these two substances onto the same CN neuron resulted in dramatic differences in time course to maximum effect

The role of the medial olfactory pathways in olfaction: behavioral and electrophysiological data.

The role of the olfactory inputs transmitted by the medial olfactory pathways --i.e. anterior limb of the anterior commissure and medial forebrain bundle-was studied in rats submitted either to a bilateral lesion of the anterior commissure (bAC group) or to a bilateral transection of the lateral olfactory tract followed by an anterior commissure bilateral lesion (bCL group). In the bCL group, the characteristic emotional reactions usually observed in sham-operated rats to biologically meaningful odorants (odor of a predator or of conspecifics) were no longer observed; in this group, the modula

Somatosensory cerebral evoked potentials after vascular lesions of the brain-stem and diencephalon.

Clinical and electrophysiological observations are described in 7 patients with clinically well-identified vascular lesions of the brain-stem or diencephalon. In the patients of Group A with a thalamic syndrome, the somatosensory cerebral evoked potentials had a reduced voltage and increased latency on the affected side. No significant anomalies were recorded in the patients of Group B with a Wallenberg or Weber syndrome. In patients of group C with a locked-in syndrome, the cerebral evoked potentials presented marked bilateral anomalies which provided interesting data about the extension of t

T cells contribute to approaching positive but potentially risky stimuli through medial prefrontal cortex immune modulation.

Avoidance behaviour, a core feature of the "Behavioural Immune System," minimizes exposure to pathogens but limits social and environmental engagement. For some individuals, the benefits of approaching attractive but risky stimuli may outweigh the costs of avoiding them. In spontaneously approaching (AP) mice, we identified the neuronal and immune correlates that contribute to individual differences in approach to positive yet potentially risky stimuli. AP mice displayed enhanced excitatory synaptic transmission and excitability in the pyramidal neurons of the medial prefrontal cortex (mPFC), …

The nanobody Nb.C1 potentiates human acid-sensing ion channel 1a and 1b.

Acid-sensing ion channels (ASICs) are proton-gated ion channels involved in synaptic transmission, pain, and ischaemic injury. Nb.C1 is a nanobody which targets human ASIC1a and was first described as a silent binder, but its broader pharmacological profile has not been resolved. Here, we show that Nb.C1 potentiates acid-evoked peak currents of both hASIC1a and hASIC1b by increasing current amplitude relative to control, with EC50 values of ∼55 nM and ∼73 nM, respectively. This activity is consistent with sequence conservation of the thumb domain between these isoforms, which serves as the Nb.…

Structure and Function of Ocular Proteoglycans: Essential Proteins for Vision.

This narrative review outlines the structure and essential functions of ocular proteoglycans (PGs) in visual processing as documented in the extensive literature on this subject matter. The eye, as one of the most complex sensory organs, relies on the coordinated activity of various tissues and cell types, with PGs playing a central role in facilitating communication and maintaining tissue function. These molecules stabilise ocular tissues; for example, SPACRCAN (IMPG2) and hyaluronan aggregates in the interphotoreceptor matrix protect photoreceptors from oxidative stress. Specialised heparan

A Narrative Review of Amyloid-β Monoclonal Antibodies for Alzheimer Disease: How Amyloid Species Engagement May Affect Clinical Outcomes.

BACKGROUND: Alzheimer's disease (AD) is a leading cause of death worldwide, with growing prevalence as life expectancy increases. An important neurological hallmark of AD is the deposition of extracellular neuritic amyloid-β (Aβ) plaques that can disrupt synaptic transmission and cause neuronal death. More recent studies suggest that targeting Aβ species can slow the progression of cognitive decline in AD. REVIEW SUMMARY: This narrative review examines the efficacy of monoclonal antibodies targeting the amyloid-β (Aβ) protein in the treatment of AD. It discusses the mechanisms by which these …

Mitochondrial Ca2+ uniporter b (MCUb) regulates neuronal Ca2+ dynamics and resistance to ischemic stroke.

Mitochondrial Ca2+ transport regulates many neuronal functions including synaptic transmission, ATP production, gene expression and neuronal survival. The mitochondrial Ca2+ uniporter (MCU) is the core molecular component of the mitochondrial Ca2+ uptake complex in the inner mitochondrial membrane. MCUb is a paralog of MCU that negatively regulates mitochondrial Ca2+ uptake in the heart and the cells of the immune system. However, the function of MCUb in the brain is largely unknown. Here, we report that MCUb knockout (KO) led to enhanced mitochondrial Ca2+ uptake in cortical neurons. By simul…

Zinc ion dyshomeostasis in autism spectrum disorder.

Autism spectrum disorder (ASD) is a neurodevelopmental disorder with significant social, communicative, and behavioral challenges, and its prevalence is increasing globally at an alarming rate. Children with ASD often have nutritional imbalances, and multiple micronutrient deficiencies. Among these, zinc (Zn2+) deficiency is prominent and has gained extensive scientific interest over the past few years. Zn2+ supports numerous proteins, including enzymes and transcription factors, and controls neurogenesis and cell differentiation. It modulates synaptic transmission and plasticity by binding to…

BoNT/Action beyond neurons.

Botulinum neurotoxin type A (BoNT/A) has expanded its therapeutic uses beyond neuromuscular disorders to include treatments for various pain syndromes and neurological conditions. Originally recognized for blocking acetylcholine release at neuromuscular junctions, BoNT/A's effects extend to both peripheral and central nervous systems. Its ability to undergo retrograde transport allows BoNT/A to modulate synaptic transmission and reduce pain centrally, influencing neurotransmitter systems beyond muscle control. BoNT/A also interacts with glial cells, such as Schwann cells, satellite glial cells…
