Differential patterns of receptor activator of nuclear factor kappa B ligand/osteoprotegerin expression in human periapical granulomas: possible association with progressive or stable nature of the lesions.

Receptor activator of nuclear factor kappa B ligand (RANKL) and osteoprotegerin (OPG) are expressed in apical periodontitis, suggesting a role for these molecules during lesion development. However, the profiles of RANKL/OPG expression in periapical lesions remain unknown. In this study we investigated the patterns of RANKL and OPG mRNA expression by real-time polymerase chain reaction in human periapical granulomas (N = 44) and compared them with sites presenting characteristic bone resorbing activity: healthy (n = 14) and orthodontically stretched and compressed periodontal ligament (n = 26)

Periodontal repair in dogs: healing in experimentally created chronic periodontal defects.

A need exists for well-defined animal models to objectively evaluate surgical principles and a possible role for biochemical wound conditioning and biomaterials in promoting periodontal regeneration. To test an existing model for its usefulness in quantitative evaluation of periodontal wound healing, large supraalveolar periodontal defects were surgically created around the mandibular premolars (P2, P3, P4) in left or right jaw quadrants in 5 beagle dogs. The defects were exposed to the oral environment for 6 months and were then subjected to reconstructive flap surgery (chronic defects). Heal

Oral Health Status and Cardinal Symptoms of Depression in the Old Order Amish: a cross-sectional study.

Background Although results have been mixed, possibly owing to high confounding heterogeneity within and across studies, multiple efforts suggest that tooth loss and periodontitis are associated with depression. Methods We analyzed questionnaires on dental health and depression obtained as a component of a Wellness study in 4,288 Old Order Amish (mean age = 43.5 ± 16.8 years; 57% women), a population with relatively high genetic and lifestyle homogeneity. Specifically, we obtained questionnaires reporting the use of removable partial/complete dentures (false teeth) and cardinal symptoms of dep…

Patterns of multimorbidity and their association with edentulism: the moderating role of health literacy in the Lifelines Cohort.

Multimorbidity may affect oral health, especially in individuals with limited health literacy (HL), or specific multimorbidity patterns; however, evidence remains scarce. Understanding these associations could improve early detection and prevention, both in clinical care and public health strategies. This study aimed to assess the association of multimorbidity and multimorbidity patterns with complete tooth loss, i.e. edentulism, and whether HL moderates this association. A total of 42 357 participants from the prospective multigenerational Dutch Lifelines Cohort Study were included. Multimorb…

Optimizing non-crosslinked type I collagen barrier membranes for guided bone regeneration: a comparative study of a BMP-2 derived peptide adsorption and covalent grafting.

Oral diseases like periodontitis and tooth loss affect billions worldwide, causing alveolar bone resorption and complicating implant placement and bone regeneration. Guided bone regeneration addresses these defects using barrier membranes that block soft tissue infiltration and promote bone growth. CollaTape®, a type I bovine collagen membrane, is widely used for its biocompatibility and resorbability, though its bioactivity and antibacterial properties could be improved. This study compares two functionalization methods for enhancing CollaTape® membranes: adsorption of GBMP1α peptide (a BMP-2…

ACP4 Variants in Hypoplastic Amelogenesis Imperfecta.

Amelogenesis imperfecta (AI) is a group of rare inherited conditions causing tooth enamel defects. Human acid phosphatase 4 (ACP4) is a transmembrane protein involved in maintaining appositional enamel growth. Variants in ACP4 cause recessive hypoplastic AI. Here we identify further families and review published ACP4 variants causing AI. In three Pakistani families, we identified a new ACP4 variant, c.254T > C, p.(Pro85Leu), which long-read sequencing revealed to be a founder variant. Two further families were homozygous for previously reported pathogenic ACP4 variants. Further details are als

Aberrant Molecular Myelin Architecture in Charcot-Marie-Tooth Disease Type 1A and Hereditary Neuropathy With Liability to Pressure Palsies.

Charcot-Marie-Tooth Disease Type 1A (CMT1A) and Hereditary Neuropathy with Liability to Pressure Palsies (HNPP) are the most common inherited peripheral neuropathies and arise from copy number variation of the Peripheral Myelin Protein 22 (PMP22) gene. While secondary axon degeneration has been proposed as the primary driver of disability, our prior work demonstrated pronounced neuromuscular impairment in CMT1A model mice in the absence of overt axonal loss, prompting investigation into primary myelin dysfunction. Here, we reveal that altered PMP22 dosage profoundly disrupts molecular architec

Zinc transporter ZIP13 in mesenchymal tissues: From intracellular metal distribution to systemic homeostasis.

BACKGROUND: Zinc is an essential trace element that plays pivotal roles in development, immunity, inflammation, and aging. Precise regulation of intracellular zinc distribution is mediated by zinc transporters of the ZIP (SLC39) and ZnT (SLC30) families, and their functions are increasingly recognized as tissue- and organelle-specific. Among these transporters, ZIP13 (SLC39A13) is a key regulator of mesenchymal biology. Genetic studies have demonstrated that loss of ZIP13 function causes connective tissue abnormalities, impaired bone and tooth development, and spondylodysplastic Ehlers-Danlos
