Effects of telomerase modulation in human hematopoietic progenitor cells.

Loss of telomeric repeats has been causally linked to replicative senescence and aging in human cells. In contrast to normal somatic cells, which are telomerase-negative, hematopoietic stem cells have low levels of telomerase, which can be transiently upregulated upon cytokine stimulation. To examine whether ectopic expression of telomerase can overcome telomere erosion in hematopoietic progenitor cells, we overexpressed telomerase in CD34+ and AC133+ cord blood (CB) cells using retroviral vectors containing hTERT, the catalytic component of telomerase. Although the hTERT-transduced CB cells e

SMYD3-mediated H3K4 trimethylation aggravates hypertension-induced renal injury via TXNIP transcriptional activation.

Hypertensive renal disease (HRD) is the second leading cause of end-stage renal disease (ESRD) following diabetes mellitus, with oxidative s … levated creatinine, BUN, urinary albumin), pathological damage, oxidative stress, inflammation, and cellular senescence, accompanied by increased SMYD3 and H3K4me3. Treatment with MTA/BCI-121 alleviated these changes, and SMYD3 knockdown/inhibition reversed Ang II-induced injuries in HK-2 cells. Mechanistically, SMYD3 was associated with enhanced TXNIP transcription …

The first 25 years of the Northwestern University SuperAging Program.

During late life, "average" does not mean "intact." For example, cross-sectional data from a common word list learning test show that average delayed word recall raw score at age 80 (5/15) is approximately half that at age 56 to 66 (9/15). Cognitive and neurobiological dissolution is therefore implicitly incorporated into concepts of the aging brain. This position is being challenged through investigations on "superaging," a term that was coined at the Northwestern Alzheimer's Disease Research Center (ADRC) to define persons ≥ 80 years with delayed word recall raw scores at least equal to thos

JUN activation by disrupted calcium homeostasis orchestrates a Nox4-inflammasome-PUMA apoptotic triad in obesity-exacerbated renal injury following lower limb ischaemia-reperfusion.

Obesity exacerbates renal injury following lower limb ischaemia-reperfusion (LLIRI) through mechanisms involving calcium dysregulation and oxidative-inflammatory-apoptotic cascades. However, the underlying pathways remain to be identified. Thus, this study aimed to elucidate the underlying pathways by using transcriptomics, multi-omics, and pharmacological validation. Utilizing transcriptomics, multi-omics analysis, and targeted biochemical/molecular assays in a high-fat diet-induced obese mouse model of LLIRI, combined with pharmacological validation using nicardipine in human renal tubular c

Intracellular Eimeria bovis macromeront formation induces bystander cell accumulation and TNT formation.

INTRODUCTION: Eimeria bovis first merogony is an intracellular process (~ 3 weeks) resulting in the formation of large macromeronts (≤ 400 μm) containing up to 140,000 merozoites I, each. The production of merozoites I poses critical metabolic stress on bovine endothelial host cells, leading to mitochondrial dysregulation and premature senescence. In this context, an accumulation of non-infected bystander cells (BCs) around E. bovis macromeront-carrying host cells (MCHCs), eventually supporting MCHCs, was observed. METHODS: BC accumulation was quantified by 3D confocal microscopy. A meront-tr

IL-1β Inhibition Partially Negates the Beneficial Effects of Diet-Induced Atherosclerosis Regression in Mice.

BACKGROUND: Thromboembolic events secondary to rupture or erosion of advanced atherosclerotic lesions is the global leading cause of death. The most common and effective means to reduce these major adverse cardiovascular events, including myocardial infarction and stroke, is aggressive lipid lowering via a combination of drugs and dietary modifications. However, we know little regarding the effects of reducing dietary lipids on the composition and stability of advanced atherosclerotic lesions, the mechanisms that regulate these processes, and what therapeutic approaches might augment the benef

Endothelial Dysfunction in Atherosclerotic Cardiovascular Diseases and Beyond: From Mechanism to Pharmacotherapies.

The endothelium, a cellular monolayer lining the blood vessel wall, plays a critical role in maintaining multiorgan health and homeostasis. Endothelial functions in health include dynamic maintenance of vascular tone, angiogenesis, hemostasis, and the provision of an antioxidant, anti-inflammatory, and antithrombotic interface. Dysfunction of the vascular endothelium presents with impaired endothelium-dependent vasodilation, heightened oxidative stress, chronic inflammation, leukocyte adhesion and hyperpermeability, and endothelial cell senescence. Recent studies have implicated altered endoth

Functional p53 is required for effective execution of telomerase inhibition in BCR-ABL-positive CML cells.

OBJECTIVE: In chronic myeloid leukemia (CML), increased cellular turnover of hematopoietic cells driven by the oncogene BCR-ABL leads to accelerated telomere shortening despite increased telomerase activity. It has been postulated that shortened telomeres, particularly in the context of increased telomerase activity, might facilitate accumulation of genetic aberrations and, consequently, disease progression from chronic phase to accelerated phase and blast crisis. Therefore, inhibition of telomerase might be a promising approach in CML therapy. MATERIAL AND METHODS: To investigate the therape

A mitochondrial paradigm for degenerative diseases and ageing.

A variety of degenerative diseases have now been shown to be caused by mutations in mitochondrial genes encoded by the mitochondrial DNA (mtDNA) or the nuclear DNA (nDNA). The mitochondria generate most of the cellular energy by oxidative phosphorylation (OXPHOS), and produce most of the toxic reactive oxygen species (ROS) as a by-product. Genetic defects that inhibit OXPHOS also cause the redirection of OXPHOS electrons into ROS production, thus increasing oxidative stress. A decline in mitochondrial energy production and an increase in oxidative stress can impinge on the mitochondrial permea

Peripheral immune cell subsets as potential predictors of benefit from immune checkpoint blockade therapy in small cell lung cancer.

BACKGROUND: Small cell lung cancer (SCLC) is a lethal neoplasia. Chemotherapy (Ct) plus immune checkpoint blockade therapy (ICBt) is now the standard of care although limited survival benefit. Lack of biomarkers of response leads to suboptimal patient selection, distorting results of clinical trials. Deciphering the ICBt-driven peripheral immune events may recognize those patients who benefit the most. Here, we characterize early peripheral immune kinetics in SCLC patients treated with Ct + ICBt with improved survival. METHODS: SCLC patients from 8 centers were prospectively recruited into 3
