Pharmacological inhibition of G protein-coupled receptor kinase 2 (GRK2) by paroxetine attenuates acetaminophen-induced hepatotoxicity.

Acetaminophen (APAP) overdose is a leading cause of acute liver injury and is associated with high mortality. G protein-coupled receptor kinase 2 (GRK2) is a widely expressed serine/threonine kinase involved in the regulation of multiple cellular signaling pathways. Dysregulation of GRK2 expression has been linked to numerous pathological states, highlighting its potential contribution to disease mechanisms. However, its involvement in drug-induced acute liver injury is yet to be elucidated. This study explored the hepatoprotective effects of paroxetine, a GRK2 inhibitor, in the context of APA

Isoquercetin and Zafirlukast Cooperatively Suppress Tumor Growth and Thromboinflammatory Signaling in a Xenograft Model of Ovarian Cancer.

Cancer-associated thrombosis (CAT), encompassing both venous thromboembolism and arterial thrombosis, contributes to up to 14% of cancer-related mortality and remains difficult to treat due to the bleeding risks of conventional anticoagulants. Protein disulfide isomerase (PDI) and its family member ERp57 (PDIA3) are thiol isomerases that regulate both arterial and venous thrombosis and are also upregulated in tumors, where they promote growth, metastasis, and immune evasion. Here, we evaluated the therapeutic potential of two thiol isomerase inhibitors-isoquercetin (ISOQ), a selective PDI inhi

Fe-POM Anchored on mSiO2-Coated Upconversion Nanoparticles for Cascading Catalytic Nano-Synergistic Therapy.

Traditional tumor treatments have limitations, such as poor targeting and systemic toxicity. Therefore, the development of intelligent nanotheranostic systems is necessary. In this work, we developed a multifunctional core-shell nanocomposite named upconversion nanoparticles (UCNPs) coated with mesoporous silica and anchored with Fe-substituted polyoxometalate (UCNPs@mSiO2/Fe-POM). This system uses UCNPs as the core for imaging. The core is encapsulated within a mesoporous silica (mSiO2) shell. This shell contains Fe-substituted polyoxometalate (POM, Fe-POM), in which Fe ions are incorporated

Defects in PDIA4 increase individuals' susceptibility to congenital heart disease.

INTRODUCTION: Congenital heart disease (CHD) comprises structural abnormalities of the heart and major blood vessels arising during fetal development. Protein disulfide isomerase family member 4 (PDIA4) facilitates protein folding processes. However, its potential involvement in CHD has not been investigated. In this study, we identified PDIA4 as a candidate gene potentially involved in cardiac development. METHODS: Whole-exome sequencing and targeted sequencing were performed to identify PDIA4 as a candidate gene of CHD. To investigate the functional role of PDIA4, PDIA4-knockdown human umbi

Unraveling Oral Fate of Poly(Lipoic Acid)s: Mechanistic Insights and Delivery Horizons.

Efficient oral delivery remains fundamentally constrained by harsh gastric milieu and intrinsic challenge of traversing multiple heterogeneous physiological barriers. Here, we elucidate that poly(lipoic acid)s (PLAs) holds great promise for oral delivery. Reversible thiol-disulfide exchange between PLAs and biological thiols at mucosal, epithelial and target cell interfaces drives traversal of each barrier, yielding 46% mucus permeability within 4 h, 26% epithelial transcytosis within 8 h, and high target-cell penetration, all markedly reduced upon thiol blockade. Acid-resistant yet neutral-pH

The Positive Roles for Reactive Oxygen Species in Human Reproduction; Implications for the Therapeutic Application of Antioxidants.

While the pathological impact of reactive oxygen species (ROS) in the aetiology of human infertility has received much attention, this review explores the counterproposal that these highly reactive metabolites play a positive role in mediating reproductive success. The physiological importance of ROS in biological systems can be distilled into three main categories of influence: (1) ROS can oxidize thiols to generate either the corresponding sulfenic acid or disulfide bridges. This oxidizing capacity is critical for several reproductive processes, including the cross linking of sperm chromatin

Crystal structure and functional characterization of an Asp49 phospholipase A2 from the bushmaster (Lachesis muta).

Snake-venom phospholipases A2 (PLA2s) are small, structurally conserved enzymes that contribute significantly to the pathophysiology of envenomation. Here, we report the purification and crystal structure of an Asp49-PLA2 isolated from the venom of Lachesis muta, a pit viper from the Peruvian Amazon. The enzyme was purified using ion-exchange and size-exclusion chromatography and exhibited phospholipase activity in a dose- and time-dependent egg-yolk degradation assay. Pure protein crystals were obtained in space group P6222 and diffracted to 2.36 Å resolution, with two molecules in the asymme

Carvedilol, an antihypertensive drug that reduces morbidity and mortality in heart failure, alleviates ferroptotic damage in cardiomyocytes and hepatocytes.

Oxidative stress-driven ferroptosis plays particularly important role in ischemia/reperfusion-associated myocardial injury and toxicant-induced liver injury. Carvedilol, a widely-prescribed antihypertensive agent with proven survival benefits in chronic heart failure patients, was evaluated in this study for its antiferroptotic activity. Using H9C2 rat cardiomyocytes and BRL-3A rat hepatocytes as in vitro models, we examined whether carvedilol can effectively alleviate chemically-induced ferroptotic injuries. We found that carvedilol afforded robust cytoprotection against ferroptosis triggered

An online SO2 and CS2 detection system combining weighted elliptical dual-spectrum reconstruction (WEDSR) with convolutional neural network (CNN).

Sulfur dioxide (SO2) and carbon disulfide (CS2) are indicator gases that are typically employed to assess the insulation condition of gas-insulated switchgear (GIS). However, there is significant spectral overlap between SO2 and CS2 in the ultraviolet (UV) band, which makes it difficult to quantify them simultaneously using UV differential optical absorption spectroscopy (UV-DOAS). In this paper, we propose a weighted elliptical dual-spectrum reconstruction (WEDSR) method, which enhances the feature signals by performing shunt spectral reconstruction on SO2 and CS2 mixture spectra. Our approac

Chemical Synthesis and Biological Evaluation of the Insecticidal Peptide AaIT5 Identified From the Androctonus australis Venom.

Scorpion venom contains various bioactive peptides. Voltage-gated sodium channel toxins (NaTxs) have been the primary focus of scorpion venom research because of their significant potency and high selectivity. A variety of NaTxs have been identified in the venom of Androctonus australis, six of which exhibit insecticidal activity. Among these, the biological activity of AaIT5 remains poorly characterized because of its low abundance in the venom. In this study, AaIT5 was chemically synthesized via native chemical ligation (NCL), suppressing side reactions associated with aspartimide formation

Ruthenium Compounds Differentially Inhibit Group IA and IIA Snake Venom Phospholipase A2 Anticoagulant Activity.

Neurotoxicity caused by snake venom phospholipase A2 (PLA2) activity derived from coral snakes (e.g., Micrurus tener, Micrurus fulvius, group IA PLA2) and some rattlesnakes (e.g., Crotalus scutulatus, group IIA PLA2) is medically significant. Of interest, the catalytic site of PLA2 also binds to activated clotting factor X, causing anticoagulation. Given that ruthenium (Ru)-containing compounds have been demonstrated to inactivate hemotoxic venoms in a solvent-dependent manner (e.g., 0.9% NaCl, phosphate-buffered saline), we wished to determine if RuCl3 would cause solvent-dependent inhibition

Microbial volatile organic compounds in plant defense: mechanisms, signaling networks, and agricultural applications.

Microbial volatile organic compounds (mVOCs) are low-molecular-weight metabolites released by bacteria and fungi in the rhizosphere and phyllosphere. mVOCs have emerged as important mediators of plant-microbes' communication and systemic defense priming. This review covers current knowledge of the biochemical diversity of the mVOCs, encompassing major classes of mVOCs, and their mechanisms of action in plant defense. We delineate the molecular signaling cascades activated by key mVOCs such as 2,3-butanediol, acetoin, dimethyl disulfide, and geosmin, highlighting their convergence on jasmonic a

Cocaine- and amphetamine-regulated transcripts in two percomorphs: evolutionary conservation and energy-status dependent responses.

Cocaine and amphetamine-regulated transcript (Cart) is a pleiotropic neuropeptide involved in appetite regulation and maintenance of energy homeostasis, among other processes. Although Cart is widely studied in vertebrates, its multigenic nature in fish raises questions about which functions are affected by specific Cart peptides. This study compared two important aquaculture percomorph species with distinct habitats and feeding behaviors: Nile tilapia (Oreochromis niloticus) and gilthead seabream (Sparus aurata). Bioinformatics revealed seven cart genes in tilapia and six in seabream that sho

Enhancement of crispiness in mahua through the incorporation of salt-modified wheat amylose.

Crispness is an important feature of high-quality Mahua. The results of this study demonstrate that crispness of Mahua could be enhanced by incorporating salt-modified wheat amylose in this study. The results showed that the crispness of Mahua increased from 40.63 to 20.60 g as 10% salt-modified wheat amylose (where the ratio of 50% wheat amylose and 20% NaCl solutions is 1:5, and the salt-modified time was 36 h at 4 °C) was added, and the dough was mixed at 45 °C for 12 min. It was hypothesized that the possible mechanism might involve the bound water within the intermolecular β-sheets struct

Thiolated Chitosan-Stearic Acid Thiol-Linked Biomimetic Hydrogels for Axonal Regrowth and Neuronal Tissue Regeneration.

Neuronal disorders like spinal cord injury, stroke, peripheral neuropathy, and neurodegenerative diseases like Alzheimer's disease, Parkinson's disease, etc., need regenerative approaches and therapeutics like tissue engineering and cell therapy for neuronal systems. Significant factors such as neuronal damage, impaired immune response, and glial scar formation create a hostile environment for neural repair and functional recovery. To overcome these challenges, the development of biocompatible and bioactive biomaterials is essential for therapeutic applications. In this work, a thiolated chito

A conserved mechanism for dimerization and activation of superfamily 1A UvrD-family helicases.

DNA helicases are ATP-dependent motor proteins that catalyze duplex DNA unwinding and are involved in DNA repair, recombination, and replication restart. Prominent members of the non-hexameric SF1A UvrD-family helicases are Escherichia coli UvrD, Rep, Bacillus stearothermophilus PcrA, and Mycobacterium tuberculosis (Mtb) UvrD1. SF1A monomers are processive 3'-to-5' single-stranded DNA translocases but need to be activated to become DNA helicases. One mechanism of activation is dimerization. Whereas Rep, UvrD, and PcrA form non-covalent dimers, the Mtb UvrD1 helicase forms a redox-dependent cov

Hirudin as a Therapeutic and Brain-Delivery Platform: From Stroke to Central Nervous System Disorders.

Crossing the blood-brain barrier (BBB) remains a central obstacle in central nervous system (CNS) therapeutics. Hirudin, a 65-amino acid, disulfide-stabilized, direct thrombin inhibitor with a long history in Traditional Chinese Medicine, has shown compelling efficacy in thrombotic disorders and mounting neuroprotective activity in preclinical stroke models, with supportive clinical signals. Although few studies have directly quantified its BBB permeability, available evidence indicates low but measurable brain exposure under pathological conditions, likely via paracellular leakage rather than

NRF2 coordinates ferroptosis and disulfidptosis in dermal papilla cells via redox metabolic reprogramming in androgenetic alopecia.

Dysfunction of dermal papilla cells (DPCs) is a central feature of androgenetic alopecia (AGA), in which oxidative imbalance plays a critical role. Nuclear factor erythroid 2-related factor 2 (NRF2) is a master regulator of cellular redox homeostasis; however, its role in AGA remains unclear. Here, using primary DPCs, hair follicle organoids, and dihydrotestosterone (DHT)-induced mouse models, we found that NRF2 was markedly downregulated under AGA-associated conditions. Mechanistically, our data suggest that reduced NRF2 activity is associated with two distinct regulated cell death-related pr
