Praziquantel Targets M1 Macrophages and Ameliorates Splenomegaly in Chronic Schistosomiasis.

Splenomegaly is a common feature of many infectious diseases, including schistosomiasis japonica. However, the immunopathogenesis and the treatment of splenomegaly due to schistosomiasis have been largely neglected. Praziquantel (PZQ), a classical schistosomicide, has been demonstrated by us and others to have antifibrotic and anti-inflammatory activities against schistosomiasis. In this study, we investigated the effect of PZQ on alleviating the splenomegaly caused by Schistosoma japonicum infection in mice. The results showed that the number of macrophages, especially the number of M1 macrop

Macrophage-derived IL-1α promotes sterile inflammation in a mouse model of acetaminophen hepatotoxicity.

The metabolic intermediate of acetaminophen (APAP) can cause severe hepatocyte necrosis, which triggers aberrant immune activation of liver non-parenchymal cells (NPC). Overzealous hepatic inflammation determines the morbidity and mortality of APAP-induced liver injury (AILI). Interleukin-1 receptor (IL-1R) signaling has been shown to play a critical role in various inflammatory conditions, but its precise role and underlying mechanism in AILI remain debatable. Herein, we show that NLRP3 inflammasome activation of IL-1β is dispensable to AILI, whereas IL-1α, the other ligand of IL-1R1, account

Mevalonate Pathway Blockade, Mitochondrial Dysfunction and Autophagy: A Possible Link.

The mevalonate pathway, crucial for cholesterol synthesis, plays a key role in multiple cellular processes. Deregulation of this pathway is also correlated with diminished protein prenylation, an important post-translational modification necessary to localize certain proteins, such as small GTPases, to membranes. Mevalonate pathway blockade has been linked to mitochondrial dysfunction: especially involving lower mitochondrial membrane potential and increased release of pro-apoptotic factors in cytosol. Furthermore a severe reduction of protein prenylation has also been associated with defectiv

[Cryopyrine-associated periodic syndrome: CAPS seen from adulthood].

Cryopyrin-associated periodic syndrome is a rare hereditary periodic fever syndrome for which, the genetic mechanism, mutation in the NLRP3 gene, has allowed to gather 3 clinical phenotypes (familial cold urticaria [FCAS], Muckle-Wells syndrome [MWS], and chronic infantile neurological cutaneous and articular syndrome [CINCA]) initially described independently, and to discover the NLRP3 inflammasome, a key receptor of the innate immunity, which regulates the interleukine-1β secretion into the mononuclear cells. The clinical manifestation of CAPS : urticaria-like skin rash, eyes redness, myalgi

Subversion of Cell-Autonomous Immunity and Cell Migration by Legionella pneumophila Effectors.

Bacteria trigger host defense and inflammatory processes, such as cytokine production, pyroptosis, and the chemotactic migration of immune cells toward the source of infection. However, a number of pathogens interfere with these immune functions by producing specific so-called "effector" proteins, which are delivered to host cells via dedicated secretion systems. Air-borne Legionella pneumophila bacteria trigger an acute and potential fatal inflammation in the lung termed Legionnaires' disease. The opportunistic pathogen L. pneumophila is a natural parasite of free-living amoebae, but also rep

Autoimmunity and autoinflammation as the yin and yang of idiopathic recurrent acute pericarditis.

Autoimmunity and autoinflammation are generally considered as mutually exclusive mechanisms of diseases but may concur to specific syndromes. Idiopathic recurrent acute pericarditis (IRAP) is defined as the recurrence of pericardial symptoms at any point following the prior cessation of acute pericarditis, and the latency is generally 6 weeks. Manifestations of pericarditis such as pericardial friction rub, electrocardiographic changes, and pericardial effusion are less frequent in the subsequent episodes compared to the index attack, and in some cases the only clinical sign is represented by

Anti-Interleukin-1 Agents in Adult Onset Still's Disease.

Interleukin 1β (IL-1β) is emerging as a master mediator of adult onset Still's disease (AOSD) pathogenesis. This pleiotropic cytokine, whose expression is under the control of the inflammasome pathway, has a wide type of effects. As a key mediator of innate immunity is a potent pyrogen and facilitates neutrophilic proliferation and diapedesis into the inflamed tissues, which are key AOSD manifestations. The study of proinflammatory cytokines profiles in sera and pathological tissues of AOSD patients has shown elevated levels of IL-1β, these levels being highly correlated with disease activity

Uric acid as a danger signal in gout and its comorbidities.

Uric acid is a waste product of purine catabolism. This molecule comes to clinical attention when it nucleates to form crystals of monosodium urate (MSU) in joints or other tissues, and thereby causes the inflammatory disease of gout. Patients with gout frequently suffer from a number of comorbid conditions including hypertension, diabetes mellitus and cardiovascular disease. Why MSU crystals trigger inflammation and are associated with comorbidities of gout has been unclear, but recent studies provide new insights into these issues. Rather than simply being a waste product, uric acid could se

Nano-granulated zoledronate sensitizes innate immune metabolism to enhance vaccine-induced and antitumor immunity.

Metabolic reprogramming targeting the mevalonate pathway represents an emerging innate immune activation target. However, its regulatory mechanisms remain incompletely elucidated. Here, we target the mevalonate pathway and construct a nano-granulated zoledronate (Nano-ZD) modulator. Following subcutaneous injection, Nano-ZD preferentially accumulates in draining lymph nodes rather than in bone tissues, enabling targeted delivery to innate immune cells. Nano-ZD functions as an immune-metabolic adjuvant, sensitizing and amplifying immune responses. By integrating Nano-ZD with the TLR4 agonist mo

Neuroinflammation in Spinal Cord Injury: Cellular and Molecular Mechanisms.

Traumatic spinal cord injury (SCI) elicits a coordinated cascade of innate and adaptive immune responses that evolve from the acute to the chronic phase and strongly influence regeneration and functional outcomes. This review synthesizes phase-specific pathobiology and the roles of microglia and infiltrating macrophages, astrocytes, neutrophils, dendritic cells, and T and B lymphocytes in neuroinflammation. We summarize glial heterogeneity beyond historical binary models, the dual actions of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6), and central signalin

Neuroinflammation in Epilepsy: Biochemical and Molecular Mechanisms and Implications for Natural Product-Driven Drug Discovery.

Epilepsy is a chronic neurological disorder prevalent worldwide, characterized by recurrent episodes of epileptic seizures. The primary current treatment approach is pharmacological, aimed at reducing the intensity and frequency of seizures, though it does not provide a cure. Neuroinflammation plays a central role in epilepsy by activating glial cells and stimulating the release of inflammatory mediators, further disrupting the balance between excitation and inhibition, thereby promoting the onset and recurrence of seizures. Furthermore, persistent inflammatory processes induce synaptic remode

Apolipoprotein E Deficiency Impairs Human Microglial Proliferation Accompanied by Elevated Cellular Oxidative Stress.

The APOE gene, which encodes Apolipoprotein E (ApoE), is the strongest genetic risk locus for Alzheimer's disease (AD). A substantial fraction of AD risk genes converges on pathways controlling lipid metabolism and immune regulation, in which microglia serve as a central integrative hub in the brain. Although microglial phenotypes linked to different APOE genotypes have been extensively characterised, the fundamental question of how ApoE shapes the core functions of human microglia remains unresolved. Here, we generated APOE knockout (KO) microglia from AD patient-derived induced pluripotent s

The SHIP1-inflammatory-lipid axis in Alzheimer's disease: from genetic risk to spatiotemporal mechanistic insights.

The INPP5D gene, encoding the SHIP1 protein, has been identified as a prominent genetic risk factor for Alzheimer’s disease (AD). However, its multifunctional roles remain complex. SHIP1 serves as a pivotal integrative node linking microglial immune activation with lipid metabolism. This review synthesizes current evidence on the SHIP1-inflammatory-lipid axis, elucidating how its phosphatase and scaffold functions modulate microglial homeostasis through the PI3K-Akt signaling pathway and the NLRP3 inflammasome. We propose a novel lipid‒lysosome‒inflammation cycle model in which SHIP1 deficienc

Short-chain fatty acid-producing commensals: Key modulators of AD neuroinflammation via the microbiota-gut-brain axis.

Alzheimer's disease (AD) lacks effective disease-modifying therapies. Neuroinflammation has emerged as a central driver of AD pathogenesis, and the gut-brain axis represents a key modulator of neuroimmune responses. Short-chain fatty acids (SCFAs), such as acetate, propionate, and butyrate, are microbial metabolites that bridge gut homeostasis with brain function. This review synthesizes current evidence on the functional depletion of SCFA-producing gut bacteria in AD, the journey of SCFAs from the gut to the brain, and their pleiotropic actions on neuroinflammatory cell states. SCFAs regulate

Precision nutrition for NAFLD: a mechanistic rationale for targeting the cytokine landscape.

Non-alcoholic fatty liver disease (NAFLD) represents a significant global health burden with limited approved pharmacotherapies. This therapeutic gap necessitates exploration of alternative strategies targeting the underlying inflammatory drivers of disease progression. NAFLD pathogenesis is characterized by chronic dysregulation of the cytokine milieu, promoting the transition from simple steatosis to steatohepatitis and fibrosis. This review critically evaluates mechanistic evidence for specific nutraceuticals-omega-3 fatty acids (EPA/DHA), vitamins D, E, and B, and curcumin-in modulating th

Lipid Droplet-Accumulating Microglia as a Therapeutic Node in Neurodegenerative Disease.

Neurodegenerative disorders increasingly reflect failures of cellular state control rather than the linear accumulation of a single toxic lesion. Microglia become trapped in maladaptive states in which inflammatory activation is decoupled from effective cargo processing. Lipid droplet-accumulating microglia (LDAM) represent a recurrent convergence state across aging and neurodegeneration, characterized by persistent neutral lipid sequestration, reduced phagocytosis-to-degradation capacity, oxidative amplification, and chronic but functionally inefficient inflammation. LDAM emerges when lipid s

Icariin Modulates the Reproductive-Immune Axis: Molecular Insights and Therapeutic Potential.

The immune system is a pivotal regulator of reproductive physiology, maintaining tissue homeostasis essential for successful pregnancy while contributing to infertility and reproductive disorders when dysregulated. Natural products represent a valuable source of novel immunomodulatory agents. Icariin (ICA), a prenylated flavonoid glycoside isolated from Epimedium species (Horny Goat Weed), has a long-standing traditional use for "invigorating yang," which modern research attributes to its reproductive function-enhancing properties. This review synthesizes emerging evidence that the beneficial

AMBRA1 allosterically activates NLRP3 by releasing its autoinhibition.

Nod-like receptor family pyrin domain-containing 3 (NLRP3) is activated by many stimuli, and its dysfunction is involved in various inflammatory diseases. Activation of NLRP3 is thought to happen via a multistep process involving phase separation, conformational opening and oligomerization. However, how NLRP3 is released from its autorepressed conformation remains elusive. Here we report that activating molecule in Beclin1-regulated autophagy protein 1 (AMBRA1), previously known for its role in autophagy, bound NLRP3 to scaffold and allosterically activate NLRP3. AMBRA1 engaged the leucine-ric

The SGK1/ENaC axis in renal injury during salt-sensitive hypertension: from sodium sensing to inflammation.

Salt-sensitive hypertension (SSHT) is commonly framed as a disorder of renal sodium transport and vascular dysfunction. Yet increasing evidence indicates that immune cells also detect sodium excess and convert it into inflammatory signaling. In this article, we argue that the serum and glucocorticoid-regulated kinase 1 (SGK1)/epithelial sodium channel (ENaC) immune axis provides a useful framework for linking salt exposure to renal injury. In antigen-presenting cells (APCs), high salt activates SGK1, increases ENaC expression and sodium entry, and promotes oxidative stress, isolevuglandin form

Macrophage RSAD2 Couples mtDNA Synthesis With a Self-Amplifying Inflammatory Circuit to Orchestrate Tissue Repair.

It is well established that mitochondrial DNA (mtDNA) synthesis governs macrophage function, yet its role in tissue repair and inflammation remains poorly understood. Using a mouse skin injury model combined with spatial transcriptomics and functional assays, we identify radical s-adenosyl methionine domain containing 2 (RSAD2) as a critical regulator of mtDNA-driven inflammation in macrophages. Mechanistically, RSAD2 directly binds the N-terminal domain of cytidylate monophosphate kinase 2 (CMPK2), a rate-limiting enzyme for mtDNA synthesis, and dually modulates its activity: it inhibits K63-
