Genome-wide identification, phylogenetic analysis, and expression profiles of ATP-binding cassette transporter genes in the oriental fruit fly, Bactrocera dorsalis (Hendel) (Diptera: Tephritidae).

The ATP-binding cassette (ABC) is the largest transporter gene family and the genes play key roles in xenobiotic resistance, metabolism, and development of all phyla. However, the specific functions of ABC gene families in insects is unclear. We report a genome-wide identification, phylogenetic, and transcriptional analysis of the ABC genes in the oriental fruit fly, Bactrocera dorsalis (Hendel). We identified a total of 47 ABC genes (BdABCs) from the transcriptomic and genomic databases of B. dorsalis and classified these genes into eight subfamilies (A-H), including 7 ABCAs, 7 ABCBs, 9 ABCCs

A Review of the Control of Aedes aegypti (Diptera: Culicidae) in the Continental United States.

Aedes aegypti (L) is an anthropophilic mosquito involved in the transmission of a variety of viral pathogens worldwide including dengue, chikungunya, yellow fever, and Zika viruses. This species, native to Africa, is well established in the continental U.S. (CONUS) and occasionally contributes to localized outbreaks of viral diseases. In the last seven decades, mosquito control programs in the CONUS have been focused on vectors of eastern equine encephalitis, St. Louis encephalitis, and West Nile viruses, as well as nuisance species. Aedes aegypti receives little control focus except during ou

Abamectin resistance in Drosophila is related to increased expression of P-glycoprotein via the dEGFR and dAkt pathways.

Many insects have evolved resistance to abamectin but the mechanisms involved in this resistance have not been well characterized. P-glycoprotein (P-gp), an ATP-dependent drug-efflux pump transmembrane protein, may be involved in abamectin resistance. We investigated the role of P-gp in abamectin (ABM) resistance in Drosophila using an ABM-resistant strain developed in the laboratory. A toxicity assay, Western blotting analysis and a vanadate-sensitive ATPase activity assay all demonstrated the existence of a direct relationship between P-gp expression and ABM resistance in these flies. Our ob

A research agenda for malaria eradication: vector control.

Different challenges are presented by the variety of malaria transmission environments present in the world today. In each setting, improved control for reduction of morbidity is a necessary first step towards the long-range goal of malaria eradication and a priority for regions where the disease burden is high. For many geographic areas where transmission rates are low to moderate, sustained and well-managed application of currently available tools may be sufficient to achieve local elimination. The research needs for these areas will be to sustain and perhaps improve the effectiveness of cur

Fluorescent molecularly imprinted hydrogel sensing strip based on nitrogen-doped carbon dots and inverse opal photonic crystals applying for effective detection for imidacloprid in fruits and vegetables.

This study integrated nitrogen-doped carbon dots (nitrogen-doped CDs) with remarkable fluorescence into a high-porosity inverse opal photonic crystal (IOPC) structure. A portable fluorescent hydrogel strip was developed by incorporating molecular imprinted biomimetic recognition, enabling the rapid identification and accurate detection of the insecticide imidacloprid (IMI). The ordered and hierarchical architecture of the IOPCs was advantageous to the uniform dispersion of nitrogen-doped CDs while providing efficient mass transfer channels for IMI. Additionally, the sensing strips achieved ads

Protective effects of Lavandula stoechas and Thymus numidicus essential oils against deltamethrin-induced hematological and biochemical toxicity in female rabbits.

Recent studies have shown that essential oils (EOs) extracted from medicinal and aromatic plants have herbicidal and/or insecticidal properties, helping to mitigate the toxicity experienced by living organisms exposed to pesticides. Moreover, the primary compounds isolated from these EOs also have the potential to reduce pesticide-induced damage. The present work aimed to evaluate the protective effects of Thymus numidicus (TNEO) and Lavandula stoechas (LSEO) against Deltamethrin-induced toxicity in female rabbits. The results obtained by GC/MS analysis showed that monoterpenes and oxygenated

A progress report in advancements of heterocyclic compounds as novel antimalarial agents over the last 5 years.

Malaria, caused by Plasmodium parasites and transmitted by Anopheles mosquitoes, remains a significant global health challenge, especially in tropical and subtropical regions where the disease is endemic. The complex Plasmodium lifecycle, involving stages in both the liver and bloodstream, leads to symptoms such as high fever, anemia, and, in severe cases, life-threatening complications, particularly P. falciparum infections. While historical treatments such as quinine and modern therapies such as artemisinin-based combination therapies (ACTs) have been effective, the growing issue of drug and

A male-drive female-sterile system for the self-limited control of the malaria mosquito Anopheles gambiae.

Despite great leaps forward in preventing and treating malaria, several challenges, including insecticide resistance, have hindered progress in fighting the disease. Thus, there is a pressing need for new tools to control malaria, including the use of genetically modified mosquitoes (GMMs) in the field. Various genetic strategies for vector control are currently explored, ranging from self-sustaining GMMs with unrestricted geographic and temporal spread to self-limiting alternatives. Here, we describe a self-limiting gene drive strategy called Male Drive Female Sterile (MDFS) targeting Anophel

Glutathione mitigates meiotic defects in porcine oocytes exposed to beta-cypermethrin by regulating ROS levels.

Beta-cypermethrin (β-CYP) is a commonly used insecticide that is potentially toxic and has adverse effects on the health of both animals and humans. Studies have indicated that β-CYP damages organs like the liver, thyroid, intestinal tract, and uterus. However, the underlying mechanisms that β-CYP affects oocyte quality are poorly understood. According to our research, β-CYP exposure led to the aberrant assembly of spindles and alignment of chromosomes, resulting in porcine oocytes' defective nuclear maturation. Concurrently, β-CYP exposure perturbed the cytoplasmic maturation by disturbing th

CRISPR-based gene drives generate super-Mendelian inheritance in the disease vector Culex quinquefasciatus.

Culex mosquitoes pose a significant public health threat as vectors for a variety of diseases including West Nile virus and lymphatic filariasis, and transmit pathogens threatening livestock, companion animals, and endangered birds. Rampant insecticide resistance makes controlling these mosquitoes challenging and necessitates the development of new control strategies. Gene drive technologies have made significant progress in other mosquito species, although similar advances have been lagging in Culex. Here we test the first CRISPR-based homing gene drive for Culex quinquefasciatus, demonstrati

Insight into Hyalomma anatolicum biology by comparative genomics analyses.

Hyalomma anatolicum is an obligatory blood-sucking ectoparasite and contributes to the transmission of Crimean-Congo haemorrhagic fever (CCHF) virus, Theileria spp. and Babesia spp. Progress in exploring the adaptive strategy of this ectoparasite and developing tools to fight it has been hindered by the lack of a complete genome. Herein, we assembled the genome using diverse sources of data from multiple sequencing platforms and annotated the 1.96 Gb genome of Hy. anatolicum. Comparative genome analyses and the predicted protein encoding genes reveal unique facets of this genome, including gen

Activation of cryptic milbemycin A4 production in Streptomyces sp. BB47 by the introduction of a functional bldA gene.

Streptomycetes are characterized by their ability to produce structurally diverse compounds as secondary metabolites and by their complex developmental life cycle, which includes aerial mycelium formation and sporulation. The production of secondary metabolites is growth-stage dependent, and generally coincides with morphological development on a solid culture. Streptomyces sp. BB47 produces several types of bioactive compounds and displays a bald phenotype that is devoid of an aerial mycelium and spores. Here, we demonstrated by genome analysis and gene complementation experiments that the ba

Evaluation of toxicity and environmental safety in use of spinosad to rationalize control strategies against Aedes aegypti.

Spinosad is a naturally-occurring insecticide used for the management of Ae. aegypti larvae. The assessment of ecotoxicological parameters of spinosad is required for verifying the environmentally-friendly behavior of the compound and for evaluating toxicity values on non-target species. Thus, the aim of the study was to conduct toxicity tests using Daphnia magna as model organism after exposure to different concentrations of spinosad. Immobility effects were observed in both acute and chronic toxicity tests at the concentration of 2.5 μg/L, and D. magna exhibited an EC50-48 h of 4.1 μg/L and

Hydrogen Cyanide Produced by Pseudomonas chlororaphis O6 Exhibits Nematicidal Activity against Meloidogyne hapla.

Root-knot nematodes (Meloidogyne spp.) are parasites that attack many field crops and orchard trees, and affect both the quantity and quality of the products. A root-colonizing bacterium, Pseudomonas chlororaphis O6, possesses beneficial traits including strong nematicidal activity. To determine the molecular mechanisms involved in the nematicidal activity of P. chlororaphis O6, we constructed two mutants; one lacking hydrogen cyanide production, and a second lacking an insecticidal toxin, FitD. Root drenching with wild-type P. chlororaphis O6 cells caused juvenile mortality in vitro and in pl
