Inhibitory Circuits Can Restore OFF Pathway Responses in Retinal Prostheses.

One of the first steps in processing visual information is to split the light signal captured by photoreceptors into complementary ON and OFF pathways, which separately encode increases and decreases in luminance. In blind patients with retinal degeneration, optoelectronic prostheses can successfully activate the ON pathway and evoke bright percepts; however, patients do not perceive dark features. This indicates that the OFF pathway is not being activated by existing prosthetics. To quantify OFF pathway deficits, we stimulated retinal ganglion cells (RGCs) in a mouse model of retinitis pigmen

Characterizing insula functional connectivity alterations in obsessive-compulsive disorder: from parcellation and gradient models.

The insula serves as an integrative hub across sensory, affective, and cognitive domains, yet its heterogeneous connectivity architecture has been largely overlooked in obsessive-compulsive disorder (OCD). Using both discrete parcellation and continuous gradient approaches, we comprehensively characterized insula functional connectivity (FC) alterations in 92 medication-free, noncomorbid OCD patients and 90 healthy controls. Data-driven clustering divided the insula into anterior and posterior subregions, followed by a diagnosis-by-subregion factorial analysis of variance (ANOVA) to assess gro

Electroacupuncture Ameliorates Learning and Memory Deficits in Vascular Cognitive Impairment Rats Through Activation of the Supramammillary Nucleus-Dentate Gyrus Circuit.

AIMS: This study aimed to elucidate the potential mechanism, especially neural circuit mechanisms, through which electroacupuncture (EA) mitigates cognitive decline in a rat model of vascular cognitive impairment (VCI). METHODS: Using a rat two-vessel occlusion (2VO) model, we assessed the therapeutic impact of EA on spatial and non-spatial memory. We integrated in vivo fiber photometry and immunofluorescence to monitor neuronal activities in the dentate gyrus (DG) and supramammillary nucleus (SuM), while Golgi staining was employed to evaluate hippocampal synaptic plasticity. Improving Emotion Control in Social Anxiety by Targeting Rhythmic Brain Circuits.

Social avoidance is a hallmark of social anxiety disorder. Difficulties in controlling avoidance behavior are the core maintaining factor of this impairing condition, hampering the efficacy of existing therapies. This preregistered study tested a physiologically grounded noninvasive enhancement of control over social approach and avoidance behavior in socially anxious individuals. Participants received dual-site phase-coupled electrical stimulation aimed at enhancing endogenous interregional theta-gamma phase-amplitude coupling between prefrontal and sensorimotor cortex, a mechanism known to s

Beyond atrophy: Network-level functional disintegration pattern along the cognitive decline in Parkinson's disease.

OBJECTIVE: This study aimed to elucidate the network-level intrinsic functional connectivity (iFC) disruptions associated with Parkinson's disease (PD). By focusing on the iFC changes among the areas of the neurodegeneration pattern in the dementia stage of PD (PD-D) and by adopting a mass-univariate iFC analysis approach, the network-based statistics (NBS), we aimed to characterize the network-level dynamics along the steps of cognitive decline in PD with a high statistical sensitivity. METHODS: We conducted surface-based measurements of cortical thickness in PD-D to identify the extent of n

Neuroinflammation-associated thalamocortical functional connectivity and cognition in older adults.

Neuroinflammation has detrimental neurocognitive effects in patients with systemic and neuropsychiatric diseases. Yet, outside of these conditions, it is unclear to what extent neuroinflammation modulates brain network-level function in older adults in the general population and, what, if any cognitive performance issues arise as a result. To evaluate these questions, we compared cerebrospinal fluid (CSF) inflammatory cytokine levels, resting-state functional magnetic resonance imaging, and cognitive performance data in 96 community-dwelling older adults (age ≥ 60 yrs). Principal component ana

A multi-metric profiling of OFC subregion connectivity identifies system-level alterations in major depressive disorder.

BACKGROUND: Orbitofrontal cortex (OFC) dysfunction contributes to major depressive disorder (MDD), particularly through disrupted network interactions. Current studies lack whole-brain coverage and fine-grained OFC mapping. We employed OFC parcellation and multi-metric connectivity to comprehensively profile OFC-related network disturbances in MDD. METHODS: OFC was parcellated into medial (MOFC), anterior middle (aMidOFC), posterior middle (pMidOFC), and lateral (LOFC) subregions via community analysis of resting-state fMRI from 586 healthy individuals. Using these subregions and whole OFC as

Atogepant modulates brain connectivity in episodic migraine: a longitudinal exploratory fMRI study.

AIM: Episodic Migraine (EM) is a highly prevalent and disabling neurological disorder. Recent therapeutic advances have targeted the calcitonin gene-related peptide (CGRP), a neuropeptide implicated in the pathophysiology of migraine. Promising outcomes have been obtained with monoclonal antibodies and, more recently, with a new class of drugs consisting of small-molecule CGRP receptor antagonists, known as gepants. Atogepant, an oral agent of the gepant class, has demonstrated efficacy in migraine prevention, yet its effects on central brain activity and neurotransmitter circuits remain large

Pathway-specific regulation of the paraventricular thalamic nucleus in depressive-like behavior.

Bipolar disorder is one of major psychiatric disorders afflicting about 1% of the world population. While genetic factors have been increasingly recognized as contributing to the disorder, the underlying etiology of bipolar disorder, such as associated brain regions and cellular pathology, are yet to be elucidated. Our previous studies using a genetic bipolar disorder animal model indicated potential role of the paraventricular thalamic nucleus (PVT) in recurrent depressive-like episodes that exhibit phenotypic similarity to the depression phase of bipolar disorder. Notably, chronic activation

An extensive dataset of spiking activity to reveal the syntax of the ventral stream.

Visual neuroscience benefits from high-quality datasets with neuronal responses to many images. Several neuroimaging datasets have been published in recent years, but no comparable dataset with spiking activity exists. Here, we introduce the THINGS ventral stream spiking dataset (TVSD). We extensively sampled neuronal activity in response to >25,000 natural images from the THINGS database in macaques, using high-channel-count implants in three key cortical regions: primary visual cortex (V1), V4, and the inferotemporal cortex. We showcase the utility of TVSD by using an artificial neural netwo

Default mode network and reward network dysconnectivity among young individuals with major depressive disorder with and without systemic low-grade inflammation.

BACKGROUND: Evidence indicates that up to 30 % of patients with major depressive disorder (MDD) exhibit low-grade inflammation (LGI). However, the effect of LGI on functional connectivity (FC) involving the default mode network (DMN) and reward network remains unclear among young patients with MDD. METHODS: A total of 161 young people with MDD and 112 age-matched healthy individuals were included in this study. Among patients with MDD, 23 met the LGI criteria (C-reactive protein levels ≥3 mg/L). All participants underwent resting-state FC magnetic resonance imaging. RESULTS: Seed-based conne

Age-Related Increase in Locus Ceruleus Activity and Connectivity with the Prefrontal Cortex during Ambiguity Processing.

Interpreting ambiguous environmental cues, like facial expressions, becomes increasingly challenging with age, especially as cognitive resources decline. Managing these challenges requires adaptive neural mechanisms that are essential for maintaining mental well-being. The locus ceruleus (LC), the brain's main norepinephrine source, regulates attention, arousal, and stress response. With extensive cortical connections, the LC supports adapting to cognitive demands and resolving conflicting cues from environment, particularly in later life. Previous research suggests that LC interacts with the

Alterations of white matter functional networks in pediatric drug-resistant temporal lobe epilepsy: A graph theory analysis study.

Neurological disorder can cause functional network changes in white matter (WM). However, changes in the WM functional network in children with drug-resistant temporal lobe epilepsy (DRTLE) require further clarification. Therefore, we combine graph theory with resting-state functional magnetic resonance imaging (rs-fMRI) and T1-weighted imaging (T1WI) to investigate the topological features of the WM network in children with DRTLE, discover potential biomarkers, and understand the underlying neurological mechanisms. We included 91 children (43 with DRTLE and 48 healthy controls), acquiring str

Mapping the Cross-species Brain Connectivity Atlas and Hemispheric Asymmetry of the Temporal Pole in Humans and Macaques.

The temporal pole (TP), one of the most expanded cortical regions in humans relative to other primates, plays a crucial role in human language processing. It is also one of the most structurally and functionally asymmetric regions. However, whether the functional architecture of the TP is shared by humans and macaques is an open question. We used spectral clustering algorithms to define a cross-species fine-grained TP atlas with different anatomical connectivity patterns. We identified three similar subregions, two ventral and one dorsal, within the TP in both humans and macaques. The parcella

Neural basis of the association between future time perspective and ADHD characteristics: functional connectivity between Left inferior parietal lobule and mPFC.

ADHD is a neurodevelopmental disorder characterized by inattention, hyperactivity, and impulsivity, leading to ongoing challenges in academic, career, and social adaptation. Previous research indicated that individuals with higher levels of future time perspective (FTP) tend to exhibit lower ADHD traits. However, little is known about the neural substrates between the relationship of FTP and ADHD traits. To address this question, we adopted voxel-based morphometry (VBM) and resting-state functional connectivity (RSFC) analyses in a sample of 240 participants to investigate the neural substrate

Dual-site beta transcranial alternating current stimulation during a bimanual coordination task modulates functional connectivity between motor areas.

BACKGROUND: Communication within brain networks depends on functional connectivity. One promising approach to modulate such connectivity between cortical areas is dual-site transcranial alternating current stimulation (tACS), which non-invasively applies weak alternating currents to two brain areas. OBJECTIVES: In the current study, we aimed to modulate inter-regional functional connectivity with dual-site tACS to bilateral primary motor cortices (M1s) during bimanual coordination and, in turn, alter behaviour. METHODS: Using functional magnetic resonance imaging (fMRI), we recorded particip
