Resveratrol ameliorates carbon tetrachloride-induced acute liver injury in mice.

Present investigation aimed to evaluate the hepatoprotective potential of resveratrol (30mg/kg, po) in mice following two different routes (po and sc) of exposure to carbon tetrachloride (CCl(4), 1.0ml/kg). Administration of CCl(4) caused significant increase in the release of transaminases, alkaline phosphatase, lactate dehydrogenase, γ-glutamyl transpeptidase, creatinine kinase, total bilirubin, urea and uric acid in serum. Significantly enhanced hepatic lipid peroxidation and oxidized glutathione with marked depletion in reduced glutathione were observed after CCl(4) intoxication. It was al

Chronic treatment with fluphenazine alters parameters of oxidative stress in liver and kidney of rats.

The aim of this study was to assess the toxic effects of chronic exposure to fluphenazine in liver and kidney of rats, as well as the possible protective effect of diphenyl diselenide on the fluphenazine-induced damage. Long-term treatment with fluphenazine caused an increase in lipid peroxidation levels in liver and kidney homogenates. Diphenyl diselenide treatment did not affect delta-aminolevulinate dehydratase (delta-ALA-D) activity, but fluphenazine alone or in combination with diphenyl diselenide showed an inhibitory effect on delta-ALA-D activity in liver. Diphenyl diselenide plus fluph

Relationship between proline and Hg2+-induced oxidative stress in a tolerant rice mutant.

There has been little agreement regarding the mechanism by which proline reduces heavy metal stress. The present work examines the relationship between Hg(2+)-induced oxidative stress and proline accumulation in rice and explores the possible mechanisms through which proline protects against Hg(2+) stress. The effect of proline on alleviation of Hg(2+) toxicity was studied by spectrophotography and enzymatic methods. Hg(2+) induced oxidative stress in rice by increasing lipid peroxidation. Pretreatment of the rice with 2 mM proline for 12 h profoundly alleviated Hg(2+)-induced lipid peroxidati

Dinitrosyl iron complexes bind with hemoglobin as markers of oxidative stress.

Prooxidant and antioxidant properties of nitric oxide (NO) during oxidative stress are mostly dependent on its interaction with reactive oxygen species, Fe ions, and hemoproteins. One form of NO storage and transportation in cells and tissues is dinitrosyl iron complexes (DNIC), which can bind with both low-molecular-weight thiols and proteins, including hemoglobin. It was shown that dinitrosyl iron complexes bound with hemoglobin (Hb-DNIC) were formed in rabbit erythrocytes after bringing low-molecular-weight DNIC with thiosulfate into blood. It was ascertained that Hb-DNIC intercepted free r

The LEC rat: a useful model for studying liver carcinogenesis related to oxidative stress and inflammation.

Growing evidence indicates oxidative stress as a mechanism of several diseases including cancer. Oxidative stress can be defined as the imbalance between cellular oxidant species production and antioxidant capability shifted towards the former. Lipid peroxidation is one of the processes that takes place during oxidative stress. Lipid peroxidation products, such as malondialdehyde (MDA) and 4-hydroxy-2-nonenal (HNE), are closely related to carcinogenesis as they are potent mutagens and they have been suggested as modulators of signal pathways related to proliferation and apoptosis, two processe

Prion-derived copper-binding peptide fragments catalyze the generation of superoxide anion in the presence of aromatic monoamines.

OBJECTIVES: Studies have proposed two opposing roles for copper-bound forms of prion protein (PrP) as an anti-oxidant supporting the neuronal functions and as a pro-oxidant leading to neurodegenerative process involving the generation of reactive oxygen species. The aim of this study is to test the hypothesis in which putative copper-binding peptides derived from PrP function as possible catalysts for monoamine-dependent conversion of hydrogen peroxide to superoxide in vitro. MATERIALS AND METHODS: Four peptides corresponding to the copper (II)-binding motifs in PrP were synthesized and used

The OxyR regulon in nontypeable Haemophilus influenzae.

Nontypeable Haemophilus influenzae (NTHi) is a gram-negative bacterium and a common commensal organism of the upper respiratory tract in humans. NTHi causes a number of diseases, including otitis media, sinusitis, conjunctivitis, exacerbations of chronic obstructive pulmonary disease, and bronchitis. During the course of colonization and infection, NTHi must withstand oxidative stress generated by insult due to multiple reactive oxygen species produced endogenously by other copathogens and by host cells. Using an NTHi-specific microarray containing oligonucleotides representing the 1821 open r

Serum antioxidant activities, malondialdehyde and nitric oxide levels in human cutaneous leishmaniasis.

Leishmania sp. are obligate intracellular protozoa that infect and replicate within mammalian macrophages. Macrophages, neutrophils and other phagocytic cells are key components of the antimicrobial and tumoricidal immune responses. These cells are capable of generating large amounts of reactive oxygen species (ROS) and reactive nitrogen species (RNS). To examine antioxidant status and lipid peroxidation in cutaneous leishmaniasis (CL) patients, activities of two ROS scavenging enzymes superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px), and the levels of malondialdehyde (MDA) and n

[Aging and magnesium].

The aim of this article is to clarify the relationships between aging or age-related diseases and magnesium (Mg). The mutation of mitochondrial DNA can occur in both aging and Mg deficiency, resulting in peroxidation, intracellular Ca(2+) accumulation and apoptosis. The capability against peroxidation decreases in aging and Mg deficiency. DNA polymerase I, RNA polimerase and DNA helicase require Mg for their activities. Under Mg deficiency the replication, transcription and translation of DNA become erroneous. The dysfunctions of vascular endothelial cell occur in aging and Mg deficiency. In c

Evidence that free radical generation occurs during scorpion envenomation.

Although it is well established that symptomatology, morbidity and death following scorpion envenomation are due to increases in neurotransmitter release secondary to toxins binding to voltage-sensitive sodium channels, the mechanism by which venom action is involved in damaging heart, liver, lungs and kidneys remains unclear. We hypothesized that scorpion toxins could induce the generation of high levels of free radicals responsible for membrane damage in organs targeted by venom action. We have investigated lipid peroxidation in different organs, through the evaluation of thiobarbituric acid

A review of recent studies on malondialdehyde as toxic molecule and biological marker of oxidative stress.

AIM: Of the many biological targets of oxidative stress, lipids are the most involved class of biomolecules. Lipid oxidation gives rise to a number of secondary products. Malondialdehyde (MDA) is the principal and most studied product of polyunsaturated fatty acid peroxidation. This aldehyde is a highly toxic molecule and should be considered as more than just a marker of lipid peroxidation. Its interaction with DNA and proteins has often been referred to as potentially mutagenic and atherogenic. This review is intended to briefly describe the physiological origin of MDA, to highlight its toxi

Nitrosative stress in early Type 1 diabetes. David H. P. Streeten Memorial Lecture.

Источник пометил публикацию как отозванную

Although hyperglycemia has been shown to cause peripheral nerve dysfunction in patients with diabetes, the biochemical mechanisms for this effect are poorly understood. The excessive production of reactive oxygen species and reactive nitrogen species has been proven to be detrimental in experimental diabetes, but there is little evidence that these metabolic events take place clinically and are physiologically important in man. To assess this we measured nitrite and nitrate (indices of nitric oxide production), nitrotyrosine (an index of peroxynitrite), 8-isoprostaglandin F-2 alpha, an isopros

Melatonin prevents cyclosporine-induced nephrotoxicity in isolated and perfused rat kidney.

Cyclosporine A (CsA) is a potent and effective immunosuppressive agent, but its action is frequently accompanied by severe renal toxicity. The precise mechanism by which CsA causes renal injury is not known. Reactive oxygen species (ROS) have been shown to play a role, since CsA-induced renal lipid peroxidation is attenuated in vivo and in vitro by the concomitant administration of antioxidants such as vitamin E. We show here the effect of the antioxidant melatonin (MLT), a hormone produced by the pineal gland during the dark phase of the circadian cycle, in a model of CsA nephrotoxicity in th
