Hybrid chain reaction and selective recognition-based homogeneous dual-fluorescence analysis of circulating tumor cells in clinical ovarian cancer samples.

BACKGROUND: Oncological analysis is important in tumor diagnosis. We constructed a dual-fluorescence and binary visual analysis system for c … clinical ovarian cancer samples; the results agreed with those of both the clinical folate receptor-polymerase chain reaction kit and radiological and pathological results. SIGNIFICANCE: This dual-fluorescence and binary visual CTCs detection method provides multiple options for clinical tumor liquid biopsy.

A Multiplex Nanopore Sequencing Approach for the Detection of Multiple Arboviral Species.

The emergence and continued geographic expansion of arboviruses and the growing number of infected people have highlighted the need to develop and improve multiplex methods for rapid and specific detection of pathogens. Sequencing technologies are promising tools that can help in the laboratory diagnosis of conditions that share common symptoms, such as pathologies caused by emerging arboviruses. In this study, we integrated nanopore sequencing and the advantages of reverse transcription polymerase chain reaction (RT-PCR) to develop a multiplex RT-PCR protocol for the detection of Chikungunya …

Point-of-Care Chlamydia trachomatis Detection Using Loop-Mediated Isothermal Amplification and Hydroxynaphthol Blue.

Loop-mediated isothermal amplification (LAMP) is a new nucleic acid amplification technique that provides high sensitivity and specificity compatible to conventional polymerase chain reaction (PCR) and simplicity, without the requirement of an expensive thermal cycler. Additionally, LAMP coupled hydroxynaphthol blue (HNB) allowed for visual color detection by the naked eye. Here we describe the methodology of Chlamydia trachomatis detection of vaginal swab from crude DNA lysis to LAMP-HNB reaction. The result can be read by the naked eye through color change from violet (negative) to sky blue …

An atypical case of disseminated cutaneous leishmaniasis due to Leishmania peruviana in the valleys of Ancash-Peru.

We present an atypical case of disseminated cutaneous leishmaniasis in the Sihuas district, located in the Andean valleys of Ancash-Peru. A 62-year-old man with no particular medical history presented multiple lesions located on the inferior abdomen, lumbar region and the right anterior thigh. Histological analysis found leishmanial amastigotes in the lesion sample, the Montenegro reaction was positive for Leishmania spp, and the polymerase chain reaction was positive for Leishmania peruviana. In conclusion, the atypical presentation of this disease may be related to the presence of an uncommo…

Development of a single-round and multiplex PCR method for the simultaneous detection of Babesia caballi and Babesia equi in horse blood.

With the aim of developing more simple diagnostic alternatives, a differential single-round and multiplex polymerase chain reaction (PCR) method was designed for the simultaneous detection of Babesia caballi and Babesia equi, by targeting 18S ribosomal RNA genes. The multiplex PCR amplified DNA fragments of 540 and 392 bp from B. caballi and B. equi, respectively, in one reaction. The PCR method evaluated on 39 blood samples collected from domestic horses in Mongolia yielded similar results to those obtained from confirmative PCR methods that had been established earlier. Thus, the single-roun…

Bartonella henselae associated with Parinaud's oculoglandular syndrome.

Bartonella henselae was recovered from the conjunctival scraping of a 38-year-old woman who presented with a 2-week history of tender preauricular lymphadenopathy and a 1-day history of a red left eye. Dry adherent colonies were observed on agar plates at 21 days of incubation, and the isolate was identified through conventional and molecular tests. Polymerase chain reaction (PCR) amplification of a specific region of the 16S rRNA gene and confirmation by a separate PCR reaction with hybridization of the product with a B. henselae-specific probe confirmed the isolate as B. henselae. This is th…

Mucosa-associated lymphoid tissue lymphoma in the conjunctiva of a child.

Marginal zone lymphoma of the mucosa-associated lymphoid tissue (MALT) occurring in the conjunctiva has yet not been described in pediatric patients. We present a case of a 10-year-old girl with a MALT lymphoma involving the conjunctiva. The tumor consisted of plasma cells and marginal zone cells with discrete epitheliotropism. Immunohistochemical studies revealed positivity for CD20 and cytoplasmic immunoglobulin lambda light chain restriction. Polymerase chain reaction-based molecular analysis of the infiltrate showed a monoclonal rearrangement for the hypervariable complimentary determining…

Clonality assessment in blood of patients with mantle cell lymphoma.

The presence of circulating neoplastic cells at diagnosis was assessed in the blood of patients presenting with mantle cell lymphoma (MCL) to determine the feasibility of a diagnostic molecular assay. Blood samples from 16 patients with pathologically reviewed MCL were analysed for the t(11;14)(q13;q32) translocation by the polymerase chain reaction (PCR): 7 (44%) were found positive. The remaining cases were examined by PCR for the presence of circulating neoplastic B-cells by amplifying the third complementarity region (CDR3) of immunoglobulin heavy chain genes and the immunoglobulin light k…

Sequential malt lymphomas of the stomach, small intestine, and gall bladder.

Low grade lymphomas of mucosa associated lymphoid tissue (MALT) are indolent neoplasms that, although tending to remain localised for many y … gle clone by the demonstration of an identical immunoglobulin heavy chain gene rearrangement by the polymerase chain reaction. The case illustrates the propensity of MALT lymphomas to "home" to mucosal sites and gives an insight into their behavior over an extended follow up.

Laboratory detection of donors implicated in transfusion-transmitted malaria.

BACKGROUND: Transfusion-transmitted malaria (TTM) is rare in non-endemic areas (non-EAs) but can potentially be fatal. This review analyzes the laboratory results of donors causing TTM in non-EAs, to assess the detectability of their Plasmodium infection by molecular or antibody tests. STUDY DESIGN AND METHODS: TTM cases in the United States, Canada, and Europe since 2010 were identified through a literature review. Authors and laboratories were contacted for missing details about sample types and laboratory methods. Results of Plasmodium polymerase chain reaction (PCR) and antibody tests wer…

The development of a novel diagnostic PCR for Madurella mycetomatis using a comparative genome approach.

BACKGROUND: Eumycetoma is a neglected tropical disease most commonly caused by the fungus Madurella mycetomatis. Identification of eumycetoma causative agents can only be reliably performed by molecular identification, most commonly by species-specific PCR. The current M. mycetomatis specific PCR primers were recently discovered to cross-react with Madurella pseudomycetomatis. Here, we used a comparative genome approach to develop a new M. mycetomatis specific PCR for species identification. METHODOLOGY: Predicted-protein coding sequences unique to M. mycetomatis were first identified in BLAS

Detection of North American orthopoxviruses by real time-PCR.

The prevalence of North American orthopoxviruses in nature is unknown and may be more difficult to ascertain due to wide spread use of vaccinia virus recombinant vaccines in the wild. A real time PCR assay was developed to allow for highly sensitive and specific detection of North American orthopoxvirus DNA in animal tissues and bodily fluids. This method is based on the amplification of a 156 bp sequence within a myristylated protein, highly conserved within the North American orthopoxviruses but distinct from orthologous genes present in other orthopoxviruses. The analytical sensitivity was

Cost implications of introducing the BIOFIRE FILMARRAY meningitis/encephalitis panel vs. real-time PCR in adult and pediatric populations in the UK.

AIMS/BACKGROUND: Meningitis is a life-threatening infection of the protective membranes surrounding the brain and spinal cord, requiring early and accurate diagnosis to inform clinical management. Standard diagnostic methods, a mix of cerebrospinal fluid (CSF) culture and polymerase chain reaction (PCR) testing, can be constrained by delayed processing or reduced sensitivity following antibiotic administration. The BIOFIRE FILMARRAY Meningitis/Encephalitis (ME) Panel is a rapid multiplex PCR test that detects 14 specific pathogens from a CSF sample in approximately 1 h, thereby avoiding multip…

A novel real-time PCR assay for the simultaneous detection of the four main causes of bacterial meningitis.

OBJECTIVES: Current multiplex assays cannot detect all four World Health Organization priority pathogens for meningitis diagnosis (Neisseria meningitidis, Haemophilus influenzae, Streptococcus pneumoniae, and Streptococcus agalactiae). This work developed a novel real-time polymerase chain reaction (PCR) assay capable of simultaneously detecting these pathogens. METHODS: Forty-four DNA samples, including type cultures (NCTC and ATCC), were tested. Specific primers and probes targeting sodC, to detect N. meningitidis, dmsA for H. influenzae, SP2020 for S. pneumoniae, and cfb for S. agalactiae …

Jejunum FABP6 expression and potential mechanism response to Salmonella Pullorum inoculation revealed by proteomic profiling and RT-PCR in Chinese Taihe black-bone silky fowls.

Salmonella Pullorum (S. Pullorum) incurs high mortality in chicks and disrupts intestinal health, and poses a severe threat to poultry industry and human health. However, in Chinese Taihe Black-Bone Silky Fowls (TBSF), the jejunum biomarker and molecular mechanism responding to S. Pullorum inoculation remain elusive. This study aims to characterize the jejunum proteome changes of TBSF affected by S. Pullorum. Using a data-independent acquisition (DIA) proteomics method, jejunum samples were collected from chicks and analyzed. These samples corresponded to the following treatment groups: the bl

Design and performance of a real-time RT-PCR assay for detection of influenza C viruses.

Influenza C virus (ICV) usually causes a mild upper respiratory tract infection in children and those infected are frequently co-infected with other respiratory viruses. However, there have only been a few hundred documented cases of ICV infection in humans as of the end of 2024. To better understand the epidemiology and clinical impact of ICVs, we developed an influenza C real-time RT-PCR (InfC rRT-PCR) assay that targets a highly conserved region of the matrix gene segment of ICVs. The analytical sensitivity evaluation demonstrated that the InfC rRT-PCR assay was highly sensitive, as it was

Development and evaluation of a duplex RT-qPCR assay for the detection and identification of Mayaro and chikungunya viruses.

Mayaro virus (MAYV) is a mosquito-borne alphavirus that is widespread in the Amazon basin, where it co-circulates with the closely related chikungunya virus (CHIKV). Due to the limited surveillance and technical limitations of diagnostic assays (scarcity of commercial assays, serology cross-reactivity), the true burden of MAYV is uncertain. We designed a new RT-qPCR assay targeting the nsp1 gene for MAYV detection, which can be used in monoplex or duplex format. In the duplex format, the new MAYV assay is combined with a CHIKV assay and a second MAYV assay, both previously published. The lower

A new quantitative reverse transcription PCR assay to improve the routine diagnosis of paracoccidioidomycosis.

Paracoccidioides are dimorphic fungal pathogens and the etiological agents of paracoccidioidomycosis (PCM). This severe systemic mycosis is restricted to Latin America, where it has been historically endemic. Currently, PCM presents the fewest diagnostic tools available when compared to other endemic mycoses. The main PCM diagnostic methods also have limitations. Molecular methods using different protocols have been proposed, but are restricted to a few regions. An analytical transversal study was conducted to evaluate a new molecular tool using specimens from patients diagnosed with PCM at a
