Heterogeneity in the trypanosomosis incidence in Zebu cattle of different ages and sex on the plateau of eastern Zambia.

On the plateau of eastern Zambia, trypanosomosis is endemic. Glossina morsitans morsitans Westwood (Diptera: Glossinidae), the only tsetse species present, is almost entirely dependent on livestock as its source of food with cattle being the most preferred host. To determine if tsetse challenge is distributed equally over the various age categories and sexes within a cattle herd, a longitudinal study of trypanosomosis incidence was conducted during the rainy season. A total of 354 head of cattle consisting of 40% oxen, 30% cows, 15% young stock, 13% calves and 2% bulls were sampled for three c

African trypanosomes: intracellular trafficking of host defense molecules.

Trypanosoma brucei brucei is the causative agent of Nagana in cattle and can infect a wide range of mammals but is unable to infect humans because it is susceptible to the innate cytotoxic activity of normal human serum. A minor subfraction of human high-density lipoprotein (HDL), containing apolipoprotein A-I (APOA1), apolipoprotein L-I (APOL1) and haptoglobin-related protein (HPR) provides this innate protection against T. b. brucei infection. Both HPR and APOL1 are cytotoxic to T. b. brucei but their specific activities for killing increase several hundred-fold when assembled in the same HD

Detection of mutant P2 adenosine transporter (TbAT1) gene in Trypanosoma brucei gambiense isolates from northwest Uganda using allele-specific polymerase chain reaction.

OBJECTIVE: To assess the application of allele-specific PCR (AS-PCR) as a fast, cheap and reliable method for detecting mutant TbAT1 associated with melarsoprol relapse in Trypanosoma brucei gambiense isolates from northwest Uganda. METHODS: A total of 105 trypanosome isolates were analysed using SfaN1 restriction fragment length polymorphism (RFLP) and AS-PCR, the former used as the gold standard. Sensitivity, specificity, positive and negative predictive values of AS-PCR as well as agreement between the tests were determined. RESULTS: Eleven trypanosome isolates had mutant TbAT1 while 94 e

Adenosine kinase mediates high affinity adenosine salvage in Trypanosoma brucei.

African sleeping sickness is caused by Trypanosoma brucei. This extracellular parasite lacks de novo purine biosynthesis, and it is therefore dependent on exogenous purines such as adenosine that is taken up from the blood and other body fluids by high affinity transporters. The general belief is that adenosine needs to be cleaved to adenine inside the parasites in order to be used for purine nucleotide synthesis. We have found that T. brucei also can salvage this nucleoside by adenosine kinase (AK), which has a higher affinity to adenosine than the cleavage-dependent pathway. The recombinant

Tropical parasitic diseases and women.

Tropical parasitic diseases constitute the greatest threat to the health and socio-economic status of women as a gender and social group. There are some gender specific ways in which parasitic diseases affect women in contrast to men due to differences in exposure, occupational risk, sociocultural behavior, gender roles and practices. These parasitic diseases confer some social stigma, which affects the health seeking behavior of women. Women are therefore important in the control of these parasitic diseases and they are key agents of change, if they are included in community control programs.

Detection of T.b. rhodesiense trypanosomes in humans and domestic animals in south east Uganda by amplification of serum resistance-associated gene.

The human serum resistance-associated (SRA) gene was identified in 28 (80%) of the 35 T.b. rhodesiense trypanosomes from parasitologically confirmed sleeping sickness cases, using the primers designed by Radwanska and in 27 (77.1%) of the same 35 T.b. rhodesiense trypanosomes using the primers designed by Gibson. However, about 20% of the 35 T.b. rhodesiense trypanosomes could not be detected by SRA-polymerase chain reaction (PCR) even when an aliquot of the first PCR was used in the second PCR, indicating that the gene may be absent in those trypanosomes or the trypanosomes could be having an

Spatial and temporal patterns of human African trypanosomosis (HAT) transmission risk in the Bipindi focus, in the forest zone of southern Cameroon.

Vector control is an effective and cost-efficient way to disrupt the transmission of human African trypanosomosis (HAT); it has nonetheless been little used to date in the disease's foci. With the aim to target trapping more precisely and to develop an optimized vector control system, a transmission risk index was used in the HAT focus of Bipindi, in the forest zone of southern Cameroon. The authors used a simplified version of the index originally developed by Laveissière et al. in 1994. The calculation of this new index only requires knowledge of the proportion of teneral flies and the propo

Evaluation of antigen and antibody rapid detection tests for Trypanosoma evansi infection in camels in Kenya.

The card agglutination test for Trypanosoma evansi (CATT/T. evansi) for the detection of antibodies, and Suratex for the detection of circulating antigens were compared in a cross-sectional study involving camels in eastern and central parts of Kenya. Of the 2227 camels screened, 2038 were owned by nomadic pastoralists in T. evansi endemic areas in eastern Kenya. A herd of 86 camels were from a ranch in Mugwoni. In Athi River area, 35 camels belonged to Kenya Trypanosomiasis Research Institute, and 68 were slaughter animals. Diagnostic sensitivity estimates were obtained by testing sera from 5…

Performance of enzyme-linked immunosorbent assays for detection of antibodies against T. congolense and T. vivax in goats.

Indirect ELISAs using denatured antigen preparations of Trypanosoma (T.) congolense (TcAGd) and T. vivax (TvAGd) for detection of anti-trypanosome antibodies in bovine serum (I-TAB ELISAs), were adapted for serodiagnosis in goats. The diagnostic proficiency, the cross-reactivity with sera from heterologous trypanosome infections and the operational performance of the assays were evaluated on experimentally trypanosome-infected goats. The I-TAB ELISA (TcAGd) detected antibodies in all T. congolense infected goats (100% overall sensitivity) from 2 to 4 weeks post-infection (p.i.) until the end o

Production of hydrogen peroxide by peripheral blood monocytes and specific macrophages during experimental infection with Trypanosoma cruzi in vivo.

Acute Chagas' disease triggers potent inflammatory reaction characterized by great increase of peripheral blood monocyte (PBM) and macrophage numbers. We studied the respiratory burst responses of PBM and peritoneal and splenic macrophages to in vivo infection (rats). The ultrastructure of heart inflammatory macrophages was also investigated. The infection increased the hydrogen peroxide (H2O2) production by PBM and splenic macrophages but not by peritoneal macrophages. Accordingly, the PBM and spleen cell numbers increased but the total number of peritoneal cells was similar to controls. Hear

Determination of melarsoprol in biological fluids by high-performance liquid chromatography and characterisation of two stereoisomers by nuclear magnetic resonance spectroscopy.

The analysis of melarsoprol in whole blood, plasma, urine and cerebrospinal fluid is described. Extraction was made with a mixture of chloroform and acetonitrile followed by back-extraction into phosphoric acid. A reversed-phase liquid chromatography system with ultraviolet detection was used. The relative standard deviation was 1% at concentrations around 10 mumol/l and 3-6% at the lower limit of determination (9 nmol/l in plasma, 93 nmol/l in whole blood, 45 nmol/l in urine and 10 nmol/l in cerebrospinal fluid). Melarsoprol is not a stable compound and samples to be stored for longer periods

A novel cultivation technique for long-term maintenance of bloodstream form trypanosomes in vitro.

We used an axenic cultivation system to grow African trypanosomes in vitro. Long-term cultivation for more than 60 days has been achieved by replacing the culture medium at regular intervals between 6 and 48 h. In contrast to a control culture without medium replacement, increasing amounts of maximum cell concentrations have been obtained, ranging from 5 x 10(6) to 2 x 10(7) trypanosomes ml-1, whereas the generation doubling time remained constant (about 6 h). Higher cell concentrations have only been obtained by total medium replacement; neither addition of fresh medium nor serum led to a hig

Zebu-taurine variation in Y chromosomal DNA: a sensitive assay for genetic introgression in west African trypanotolerant cattle populations.

Owing to increasing scientific and agricultural interest in the disease-resistant (trypanotolerant), indigenous cattle breeds of West and Central Africa, there is a need for a rational genetically based description of populations in the region. The greatest threat to the invaluable genetic resource represented by these animals is that of extensive genetic introgression of distantly related zebu cattle from northern populations which do not share their inherited tolerances. Southern blotting with a chromosome Y-specific probe, btDYZ-1 (locus DYZ1) is shown to be a sensitive assay to detect such

Chemotherapy and delivery systems: haemoparasites.

Chemotherapy of haemoparasitic diseases in domestic animals is dependent on a limited number of compounds, many of which are chemically closely related. In this review, a summary is given of each of the drugs currently available for treatment and prophylaxis of trypanosomosis and the tick-borne diseases theileriosis, babesiosis, anaplasmosis and cowdriosis. In contrast to the situation with the drugs used for tick-borne diseases, drug resistance appears to be becoming an increasing problem associated with the compounds used for trypanosomosis. The literature that has been reviewed, therefore,

Effects of infection with Trypanosoma brucei brucei on different trimesters of pregnancy in ewes.

The effects of Trypanosoma brucei brucei infection during the first, second or third trimesters of pregnancy in 13 ewes were studied. All infected ewes were anaemic with the anaemia being most severe, moderate and least in ewes infected in the second, third and first trimesters, respectively. Weight loss occurred in all infected ewes but was most severe in ewes infected in the third trimester. Three of the four ewes infected in the first trimester died without aborting while one aborted and later died. Of the four ewes infected in the second trimester, three died without aborting while one lam

[Treatment of sleeping disease caused by trypanosoma brucei gambiense with alpha-difluoromethylornithine (DFMO) in a rural hospital in Zaire].

The authors report the results of 32 patients with sleeping sickness due to Trypanosoma brucei gambiense treated with DFMO (DL-alpha-difluoromethylornithine), an inhibitor of polyamine biosynthesis. Between those patients, there were 5 new cases, 1 reinfection, and 26 cases with a primary, a secondary resistance or a relapse. 26 cases got DFMO only per os, but six others received first DFMO for two weeks by the intravenous way, followed by three weeks of DFMO per os. The secondary effects were never very severe and never prompted a definitive discontinuation of treatment. 12 cases were followe

The molecular epidemiology of parasites.

The explosion of new techniques, made available by the rapid advance in molecular biology, has provided a battery of novel approaches and technology which can be applied to more practical issues such as the epidemiology of parasites. In this review, we discuss the ways in which this new field of molecular epidemiology has contributed to and corroborated our existing knowledge of parasite epidemiology. Similar epidemiological questions can be asked about many different types of parasites and, using detailed examples such as the African trypanosomes and the Leishmania parasites, we discuss the t

Production of metacyclic forms by cyclical transmission of west African Trypanosoma (T.) brucei isolates from man and animals.

Fifteen West African Trypanosoma (T.) brucei isolates from man and animals were cyclically transmitted. Five stocks, belonging to the non-gambiense group, could easily be transmitted through Glossina morsitans morsitans or Glossina m. centralis infected on mice, whereas successful transmission of the 10 isolates, identified as Trypanosoma brucei gambiense, was performed using G. palpalis gambiensis as vector. Glossina p. gambiensis was infected with culture-derived procyclic trypanosomes by repeated membrane feeding. In both cases, metacyclic forms could normally be detected in saliva samples

Sleeping sickness and the factors affecting it in Botswana.

From the first appearance of sleeping sickness in Botswana in 1934 outbreaks increased in severity up to 1971. All populated areas around the fly belt were affected. Rates of infection are highest in the hot wet season and males aged between 30 and 50 years most affected. Rates vary considerably between tribes and have greatly increased in one tribe since 1966. Between 5 and 20% of cases die each year. Increase both in area of tsetse fly infestation and in human population size can account for the increase in severity of outbreaks. Timing of outbreaks correlates with years of low rainfall when
