Mosaic Accumulation of Somatic Genetic Variation and Estimates of Age in the Long-Lived Reef-Building Coral Acropora palmata.

Somatic genetic variation (SoGV) accumulates during an organism's lifetime and is traditionally viewed as detrimental rather than adaptive due to links with cancer and senescence. However, in modular organisms like corals, deleterious mutations can be purged at the cellular or polyp level, while adaptive mutations may rise in frequency as polyps create genetically distinct modules. Quantifying the somatic genetic landscape in corals is necessary to understand the role these mutations may have in coral and clonal animal development and evolution. Here, we catalogue somatic genetic variation in

Tamoxifen transiently disrupts estrous cyclicity without altering long-term ovarian aging trajectories.

The ovary is both one of the earliest organs to functionally age in the body, with declines emerging well before reproductive failure and contributing to systemic aging. Since ovarian function depends on tightly regulated hormonal and inflammatory cycles, even subtle disruptions can confound aging-related phenotypes. Tamoxifen-inducible Cre systems are widely used to study ovarian biology, however, tamoxifen is a selective estrogen receptor modulator capable of disturbing ovarian physiology. This introduces a critical and often overlooked concern that tamoxifen-based models may produce lasting

Aging in the air we breathe: Mechanisms and consequences of air pollution exposure for biological aging.

Air pollution is the leading environmental cause of disease burden worldwide and is increasingly recognized as a potent driver of accelerated biological aging. Rather than acting through organ-specific toxicity alone, air pollutants appear to affect the fundamental hallmarks of aging itself, including oxidative stress, chronic inflammation, epigenetic dysregulation, telomere attrition, mitochondrial dysfunction, and cellular senescence. The pollutants of greatest concern are fine and ultrafine particulate matter (PM2.5, PM0.1), traffic-related air pollution, and household combustion emissions.…

Mechanistic redundancy and hierarchy of aging mechanisms: implications for strategies to extend healthspan and biomarker integration.

Aging involves a network of interrelated biological processes that differ in causality and impact. This review proposes a hierarchical frame … anisms, while also feeding back to accelerate upstream deterioration. Secondary mechanisms comprise cellular senescence, impaired macroautophagy, deregulated nutrient sensing, epigenetic alterations, mitochondrial dysfunction, and altered intercellular communication. These processes emerge downstream of initial damage and further reciprocally reinforce it. Tertiary m …

Selective remodeling of snoRNA and sdRNA biology in glioblastoma.

Small nucleolar RNAs (snoRNAs) are best known as guide RNAs for ribosomal RNA modification, but accumulating evidence indicates that their biology extends beyond canonical ribosome maturation. Across GBM-specific and broader glioma studies, snoRNAs, snoRNA-derived RNAs (sdRNAs), and associated small nucleolar ribonucleoprotein components are emerging as regulators of malignant cell states. This review frames GBM-associated snoRNA and sdRNA alterations as a cell-biological remodeling process that links ribosome biogenesis, metabolic adaptation, treatment response, and extracellular-vesicle outp

Study on blueberries, protein and exercise for improving frailty and cardiovascular disease (STRONG): protocol for a parallel-group randomised controlled superiority trial in older Nova Scotians.

INTRODUCTION: As frailty and cardiovascular disease are closely linked, lifestyle interventions to reduce frailty could improve cardiovascular health in at-risk individuals. Previous clinical trials have shown that exercise reduces frailty, both individually and in combination with dietary protein. Blueberries can improve vascular health and protect against cellular senescence. The objective of the study on blueberries, protein and exercise for improving frailty and cardiovascular disease (STRONG) is to determine whether a year-long treatment including blueberries, protein and exercise will re…

Therapeutic potential of xanthohumol in senile osteoporosis: mTOR-driven regulation of AKT/mTOR/p70S6K autophagy axis in D-galactose models.

BACKGROUND: Senile osteoporosis (SOP) is a bone disorder characterized by bone loss, structural deterioration, and increased fracture risk. Xanthohumol (XAN), a bioactive isoflavone derived from Humulus lupulus L., shows potential bone-protective properties. PURPOSE: This study aimed to evaluate the anti-SOP efficacy of XAN and clarify its mechanisms through multidisciplinary approaches. METHODS: A SOP mouse model was established using d-galactose (D-gal), followed by a 12-week XAN treatment. Animal procedures were approved by the Ethics Committee of Naval Military Medical University (No. 20

Unveiling pathogenesis of pelvic organ prolapse through transcriptomic and bioinformatic analyses in uterosacral ligament tissues of postmenopausal women.

Limited restoration of T cell subset distribution and immune function in older people living with HIV-1 receiving HAART.

BACKGROUND: Older people living with HIV-1 (PLWH) experience a dual burden from the combined effects of aging and HIV-1 infection, resulting in significant immune dysfunction. Despite receiving HAART, immune reconstitution is not fully optimized. The objective of this study was to investigate the impact of aging and HAART on T cell subsets and function in PLWH across different age groups, thereby providing novel insights into the prognosis of older PLWH. METHOD: This study was conducted at Yunnan AIDS Care Center, China, to explore the immunological responses of old PLWH to HAART and compared

miR-9-5p regulates Sirt1 involved in testicular development and spermatogenesis in mouse.

Testicular development and spermatogenesis are critical for male reproduction, with histone (de)acetylation playing a key role in chromatin … cell proliferation, and energy homeostasis, as well as its association with signaling pathways like cellular senescence, the FoxO signaling pathway, and the AMPK signaling pathway. High throughput screen of small molecules as potential countermeasures to galactic cosmic radiation induced cellular dysfunction.

Space travel increases galactic cosmic ray exposure to flight crews and this is significantly elevated once travel moves beyond low Earth orbit. This includes combinations of high energy protons and heavy ions such as 56Fe or 16O. There are distinct differences in the biological response to low-energy transfer (x-rays) or high-energy transfer (High-LET). However, given the relatively low fluence rate of exposure during flight operations, it might be possible to manage these deleterious effects using small molecules currently available. Virtually all reports to date examining small molecule man

Towards resolving the enigma of the dichotomy of resveratrol: cis- and trans-resveratrol have opposite effects on TyrRS-regulated PARP1 activation.

Unlike widely perceived, resveratrol (RSV) decreased the average lifespan and extended only the replicative lifespan in yeast. Similarly, although not widely discussed, RSV is also known to evoke neurite degeneration, kidney toxicity, atherosclerosis, premature senescence, and genotoxicity through yet unknown mechanisms. Nevertheless, in vivo animal models of diseases and human clinical trials demonstrate inconsistent protective and beneficial effects. Therefore, the mechanism of action of RSV that elicits beneficial effects remains an enigma. In a previously published work, we demonstrated st

[Molecular mechanism of vascular calcification.].

Intimal and medial calcification are increased with type 1 and type 2 diabetes, metabolic syndrome, chronic kidney disease, and ageing. There are several biological mechanisms through which vascular calcification increases all-cause mortality and atherosclerotic plaque rupture. Arterial medial calcification increases arterial stiffness that causes systolic hypertension and diastolic dysfunction and heart failure. In contrast, arterial intimal calcification is strongly associated with atherosclerotic plaque burden, predicting adverse arterial events. In particular, micro-calcifications within t
