Extracellular signal-regulated kinase 5 promotes acute cellular and systemic inflammation.

Inflammatory critical illness is a syndrome that is characterized by acute inflammation and organ injury, and it is triggered by infections and noninfectious tissue injury, both of which activate innate immune receptors and pathways. Although reports suggest an anti-inflammatory role for the mitogen-activated protein kinase (MAPK) extracellular signal-regulated kinase 5 (ERK5), we previously found that ERK5 mediates proinflammatory responses in primary human cells in response to stimulation of Toll-like receptor 2 (TLR2). We inhibited the kinase activities and reduced the abundances of ERK5 an

Disseminated intravascular coagulation with congestive heart failure and left ventricular thrombus: a case report with literature review of 7 cases.

BACKGROUND: Coagulation abnormalities can accompany acute congestive heart failure (CHF). However, disseminated intravascular coagulation (DIC) is rarely documented in such patients. DIC is characterized by generalized excessive activation of coagulation pathways followed by their depletion with secondary activation of anticoagulation and fibrinolysis. Treatment of the cause is an integral part of management of DIC; thus, recognition of the cause is critical. CASE REPORT: A 55-year-old previously healthy man presented with breathlessness, swelling of both legs, and left leg pain. His physical

Defects in phosphate acquisition and storage influence virulence of Cryptococcus neoformans.

Nutrient acquisition and sensing are critical aspects of microbial pathogenesis. Previous transcriptional profiling indicated that the fungal pathogen Cryptococcus neoformans, which causes meningoencephalitis in immunocompromised individuals, encounters phosphate limitation during proliferation in phagocytic cells. We therefore tested the hypothesis that phosphate acquisition and polyphosphate metabolism are important for cryptococcal virulence. Deletion of the high-affinity uptake system interfered with growth on low-phosphate medium, perturbed the formation of virulence factors (capsule and

Mycobacterium tuberculosis WhiB3: a novel iron-sulfur cluster protein that regulates redox homeostasis and virulence.

SIGNIFICANCE: Mycobacterium tuberculosis (Mtb), the causative agent of tuberculosis (TB), can persist in a latent state for decades without causing overt disease. Since latent Mtb is refractory to current antimycobacterial drugs, the discovery and characterization of the biological mechanisms controlling the entry, maintenance, and emergence from latent infection is critical to the development of novel clinical therapies. RECENT ADVANCES: Recently, Mtb WhiB3, a member of the family of intracellular iron-sulfur (Fe-S) cluster proteins has emerged as a redox sensor and effector molecule control

[Intraoperative monitoring of brainstem auditory evoked potentials during microvascular decompression of cranial nerves in cerebellopontine angle].

Brainstem auditory evoked potentials (BAEP) monitoring is a useful tool to decrease the danger of hearing loss during pontocerebellar angle surgery, particularly in microvascular decompression (MVD). Critical complications arising during MVD surgery are the stretching of the VIII nerve - the main cause of hearing loss - labyrinthine artery manipulation, direct trauma with instruments, or a nearby coagulation, and at end of the surgery neocompression of the cochlear nerve by the prosthesis positioned between the conflicting vessel(s) and the VIIth-VIIIth nerve complex. All these dangers warrant

UVB-induced ERK/AKT-dependent PTEN suppression promotes survival of epidermal keratinocytes.

Ultraviolet (UV) radiation in sunlight is the major environmental cause of skin cancer. PTEN (phosphatase and tensin homolog deleted on chromosome 10) is a proven critical tumor suppressor. We report here that UVB downregulates PTEN in primary human keratinocytes, human HaCaT keratinocytes and mouse skin. As compared with normal skin, PTEN levels are reduced in human actinic keratosis, a precancerous skin lesion caused by solar UV. PTEN downregulation is mediated by two mechanisms: (1) PTEN is cleaved by active caspase in apoptotic cells in which AKT activation is reduced; and (2) PTEN transcr

Environmental estrogens and obesity.

Many chemicals in the environment, in particular those with estrogenic activity, can disrupt the programming of endocrine signaling pathways that are established during development and result in adverse consequences that may not be apparent until much later in life. Most recently, obesity and diabetes join the growing list of adverse consequences that have been associated with developmental exposure to environmental estrogens during critical stages of differentiation. These diseases are quickly becoming significant public health issues and are fast reaching epidemic proportions worldwide. In t

The critical role of vascular endothelial growth factor in pulmonary vascular remodeling after lung injury.

The pulmonary vascular endothelial cell plays a crucial role in the regulation of the pulmonary vascular tone and in the maintenance of the barrier function and integrity of the alveolar-capillary membrane. It also plays a major role in coagulation, fibrinolysis, and angiogenesis and participates in inflammatory reactions. Vascular endothelial growth factor (VEGF) is a central growth and survival factor for the endothelial cell. Particularly high levels of VEGF are expressed in the lungs, reflecting the critical role of VEGF for lung development and structural integrity of the adult lung. Vasc

Regulated expression of a cell division control-related protein kinase during development.

Protein kinases are important signaling molecules that are known constituents of cellular pathways critical for normal cellular growth and development. We have recently identified a new protein kinase, p58, which contains a large domain that is highly homologous to the cell division control p34cdc2 protein kinase. This new cell division control-related protein kinase was originally identified as a component of semipurified galactosyltransferase; thus, it has been denoted galactosyltransferase-associated protein kinase. In vitro, this protein kinase has been shown to phosphorylate a number of s

Integrated multi-omics profiling reveals phenotype- and tissue-specific host-microbiota interactions in paired tumor and peritumoral tissues of advanced gastric cancer patients from Northwest China.

BACKGROUND: Advanced gastric cancer (AGC) exhibits a high incidence in Northwest China, largely attributed to region-specific dietary patterns and environmental exposures. Its pathogenesis involves complex host-microbiota crosstalk, which has not yet been comprehensively elucidated through integrated multi-omics approaches. Herein, we employed trasncriptomic and shotgun metagenomic sequencing on paired tumoral and peritumoal mucosal tissues from 88 AGC patients in Northwest China. Our aim was to systematically characterize host gene expression profiles, the composition and functional potential

Epimedium brevicornu flavonoids alleviate neuroinflammation and Alzheimer's disease pathology via immune-related pathways.

BACKGROUND: With global population aging, Alzheimer's disease (AD) has become a critical clinical challenge. This multifactorial neurodegenerative disorder is characterized by amyloid-β aggregation, tau hyperphosphorylation, and neuroinflammation. The lack of effective disease-modifying therapies highlights the urgent need for multi-target strategies. Epimedium brevicornu flavonoids (EF), derived from a traditional medicinal plant used to support cognitive function, exhibit significant neuroprotective potential; however, the underlying mechanisms remain to be fully elucidated. The multifaceted role of cathepsin a in glioblastoma pathogenesis: from molecular scaffolding to immunological evasion.

Glioblastoma (GBM) is a highly aggressive primary central nervous system malignancy characterized by pronounced intratumoral heterogeneity, rapid progression, and resistance to standard therapeutic approaches. Increasing evidence indicates that cathepsin A (CTSA), a lysosomal serine carboxypeptidase with multifunctional roles, is associated with aggressive GBM phenotypes and may contribute to tumor progression through multiple, context-dependent mechanisms. CTSA is an established structural component of the elastin receptor complex (ERC), where it stabilizes NEU1 and supports receptor complex

A dual methane production mechanism mediated by enzymatic catalysis and the Fenton reaction in the wood-rotting fungus Schizophyllum commune.

Understanding biological sources of methane is critical for elucidating the global methane cycle and addressing climate change. While traditional research has predominantly focused on prokaryotic methanogens, emerging evidence suggests that eukaryotes, particularly fungi, can produce methane through both enzymatic and non-enzymatic pathways. However, no eukaryotic organism has been documented to possess both pathways simultaneously. Here, we provide preliminary evidence suggesting that the wood-decaying fungus Schizophyllum commune strain 20R-7-F01, isolated from sediment approximately 2 km be

Placental Methylation Profile in Women Exposed to Fluoride and Its Association with Kidney Volume at Birth.

KEY POINTS: Placental DNA methylation patterns were associated with neonatal kidney volume in a population with high fluoride exposure. Newborns with high fluoride exposure and low kidney volume showed methylation changes in genes involved in kidney development pathways. These placental epigenetic patterns revealed early markers of altered kidney development, potentially linked to reduced nephron endowment. BACKGROUND: Mexico has a high burden of CKD. The state of Aguascalientes is among the regions with the highest reported prevalence of ESKD worldwide. In this setting, CKD of unknown etiolo
