Proteomics-Based Identification of the Pyroptosis-Related Biomarker PCSK9 and Its Association With the Pathogenesis of Rheumatoid Arthritis.

Rheumatoid arthritis (RA) is a common autoimmune disease, and early diagnosis is critical for effective treatment. This study aims to identify potential biomarkers related to pyroptosis through serum proteomics analysis, offering new insights for the early diagnosis of RA. We enrolled 100 participants, including 50 patients with RA and 50 healthy controls. Serum samples were collected and analyzed using high-resolution liquid chromatography-tandem mass spectrometry (LC-MS/MS) for proteomics profiling. Differential protein expression analysis and functional annotation revealed significant upreg

Immunonutritional biomarkers in osteoarthritis: mechanistic insights and prognostic potential.

Osteoarthritis (OA) is the most prevalent degenerative joint disease worldwide. Beyond structural injury to cartilage, synovium, and subchondral bone, OA is increasingly recognized as an immunometabolic disorder characterized by low-grade inflammation and dysregulated innate and adaptive immune responses. Nutritional biomarkers-derived from macro- and micronutrient status and metabolite signatures-can shape the OA inflammatory microenvironment by modulating macrophage polarization, T-cell subset balance, cytokine networks, and key signaling programs (e.g., NF-kappaB, JAK/STAT, NLRP3 inflammaso

Cigarette and Electronic Cigarette Exposure in Osteoarthritis: Immune Dysregulation and Inflammatory Signaling Pathways.

Osteoarthritis (OA) was once seen mainly as a degenerative joint disease, but more studies now show that inflammation also has an important role in its development and progression. Smoking may be one factor in this process. Findings from population studies are still not fully consistent, but cigarette exposure is known to affect immune function and inflammatory signaling. This suggests that its role in OA may be more complex than a simple risk factor. At the same time, the fast growth of electronic cigarettes (EC) use has raised new concerns, because its effects on joint tissues are still not

Autoinflammation with infantile enterocolitis induced by a heterozygous variant (c.1357C > T) in the NLRC4 gene: a case report.

BACKGROUND: Necrotizing enterocolitis (NEC) is s a life-threatening inflammatory intestinal disorder primarily affecting preterm infants, though rare in full-term neonates. NLR-family CARD domain-containing protein 4 (NLRC4), a cytosolic inflammasome component driving IL-1β-mediated inflammation, is linked to autoinflammatory syndromes via germline gain-of-function mutations, though its role in term infant NEC remains underexplored. CASE PRESENTATION: A full-term male infant developed NEC shortly after birth, requiring emergency surgical interventions including ileostomy and intestinal resect

Overcoming cholesterol-induced pulmonary toxicity: plant sterols and natural saponins as bioactive alternatives in inhalable liposomes for active carrier-mediated synergistic therapy.

Exogenous cholesterol is routinely incorporated into inhalable liposomes to enhance membrane stability under nebulization-induced shear stress. However, excessive cholesterol deposition may trigger lipid overload in alveolar macrophages, activate the NLRP3 inflammasome, and induce pulmonary immunotoxicity, thereby severely restricting the clinical translation of such liposomal formulations. Given their ethnopharmacological relevance and structural homology, natural plant sterols and bioactive saponins derived from medicinal herbs have emerged as promising substitutes for cholesterol. These two

Nucleotide metabolism reprogramming in obesity-associated cardiovascular inflammation: a new perspective.

Obesity drives cardiovascular disease primarily through chronic meta-inflammation, yet the precise molecular convergence linking nutrient excess to sustained NLRP3 inflammasome activation in macrophages has remained unclear. Obesity induces inhibitory phosphorylation of SAMHD1, resulting in cytosolic dNTP accumulation, mitochondrial import through SLC25 transporters, uncontrolled mtDNA synthesis and oxidation, and consequent NLRP3 hyperactivation. This SAMHD1-dNTP-mtDNA-NLRP3 axis is supported by emerging evidence as a potential upstream checkpoint that may set macrophage inflammatory tone and

Prevotella copri Exacerbates Psoriasis Through Keratinocyte-Neutrophil Crosstalk via NF-κB Activation.

Psoriasis is a chronic inflammatory skin disorder driven by systemic immune dysregulation and recent studies have implicated the gut-skin axis in its pathogenesis. However, the specific role of individual gut microbes remains poorly understood. In this study, the impact of Prevotella copri (P. copri) on psoriasis development was examined and the underlying mechanisms were investigated. In an imiquimod (IMQ)-induced mouse model, oral administration of P. copri exacerbated skin inflammation, increased epidermal thickness, promoted neutrophil infiltration and aggravated disease severity. Pyoderma gangrenosum caused by the molecular uncoupling of OTULIN catalytic activity and LUBAC binding.

The pathogenic mechanisms underlying pyoderma gangrenosum (PG) remain unclear. Here we report three patients with PG from two unrelated kindreds with homozygous R57C mutation of the linear deubiquitinase OTULIN. The patients have isolated, pediatric-onset, OTULIN-related PG (ORP). In contrast to OTULIN-related autoinflammatory syndrome (ORAS), caused by mutations affecting the catalytic domain, R57C affects the PUB-interacting motif with distinct biochemical, immunological and clinical consequences. OTULIN-R57C is catalytically active but is unable to bind the linear ubiquitin assembly complex

Natural flavonoids in multiple sclerosis: molecular insights and emerging therapeutic strategies.

Multiple sclerosis is a chronic immune mediated disease in which current disease modifying therapies reduce inflammatory relapses but incompletely address neurodegeneration and remyelination. Natural flavonoids are pleiotropic polyphenols that can modulate immune and glial signaling, oxidative stress, and mitochondrial function. This review synthesizes evidence from experimental models and human studies on flavonoids relevant to multiple sclerosis, emphasizing mechanisms involving NF-κB, Nrf2, inflammasome signaling, and microglia and macrophage polarization that shape oligodendrocyte precurso

Macrophage-Mediated Transport of Insoluble Indirubin Induces Hepatic Injury During Intestinal Inflammation.

Many plant-derived bioactive molecules with low solubility and permeability can cause hepatocyte injury. However, the mechanism by which they induce hepatic damage without passive diffusion into the hepatic circulatory system remains unclear. This study demonstrate that indirubin, the main component of indigo naturalis with poor aqueous solubility, predisposes mice with chronic colitis to hepatic injury. This closely mimics the hepatic damage commonly observed in ulcerative colitis patients treated with indigo naturalis. Upon administration, indirubin is detected in the plasma, Peyer's patches

Nucleoside reverse transcriptase inhibitors as a therapeutic opportunity to counteract inflammaging and age-related diseases: New evidence from epidemiological data.

Nucleoside Reverse Transcriptase Inhibitors (NRTIs), widely used to treat HIV and hepatitis B, have recently been shown to possess anti-inflammatory properties by inhibiting inflammasome activation. Epidemiological studies have revealed a significantly reduced incidence of age-related diseases, such as Alzheimer's disease (AD) and type 2 diabetes mellitus (T2DM), among patients chronically treated with NRTIs, but not with other classes of antiretroviral drugs. In this short review, we explore the mechanistic and clinical evidence linking NRTIs to reduced inflammaging, with particular attention

Protein regulatory network mediated by palmitoylation modifications in the pathological progression of Parkinson's disease: a narrative review.

Palmitoylation is a reversible lipid modification regulated by palmitoyl transferases and acyl-protein thioesterases, in which palmitic acid is attached to protein cysteine residues. This modification plays a pivotal role in modulating membrane localization and protein stability, and its dysregulation is closely associated with various neurodegenerative diseases, including Parkinson's disease (PD). In PD, synaptotagmin-11, encoded by the PD risk gene SYT11, has been shown to reduce physiological α-synuclein (α-syn) tetramer formation while promoting the aggregation-prone monomeric form in a pa

Poliumoside Exhibits Neuroprotective Effects against Cerebral Ischemia-Reperfusion Injury by Relieving Microglia-Mediated Neuronal Damage and Astrocytic Activation.

Excessive activation of microglia contributes to neuronal damage and astrocytic activation during cerebral ischemia and hypoxia. Poliumoside (Pol) is a caffeoylated phenylpropanoid glycoside with significant anti-inflammatory and antioxidant functions. However, whether Pol can mediate microglia-mediated neurotoxicity in the ischemic brain remains nebulous. Here, a cerebral ischemia-reperfusion injury (CI/RI) mouse model was conducted to investigate Pol's role in microglial activation and neurotoxicity. We found that Pol significantly reduced neurological deficits, cerebral infarction volume, a

Adjuvant conditioning enhances neutrophil function while inducing a suppressive peritoneal macrophage phenotype.

Adjuvants are widely used to boost the immune response during vaccination protocols. Our group has previously reported that repeated intraperitoneal administration of alum in mice, known as adjuvant conditioning (AC), creates an immunosuppressive environment that delays allogeneic graft rejection through NLRP3-dependent MDSC expansion. However, little is known about the effects of AC on the reprogramming of peritoneal cavity cells, particularly the different peritoneal macrophage populations, and the impact on the adaptive immune response. We found a population-specific immune response to alum

Oxidative Stress and Survival of Leishmania spp.: A Relationship of Inverse Proportionality for Disease Outcome.

SEARCH RESULTS: Reactive oxygen species (ROS) play a dual role in leishmaniasis by contributing to both host defence and parasite survival mechanisms. In the host, ROS promote parasite clearance through induction of apoptosis, activation of pro-inflammatory signalling pathways (e.g., MAPK, JNK), inflammasome assembly, and M1 macrophage polarisation. Conversely, Leishmania species have evolved multiple strategies to neutralize ROS, including the upregulation of host antioxidant enzymes like HO-1, inhibition of ROS-producing pathways, and expression of parasite-derived antioxidants such as SOD,
