MABGEL 1: first phase 1 trial of the anti-HIV-1 monoclonal antibodies 2F5, 4E10 and 2G12 as a vaginal microbicide.

BACKGROUND: Monoclonal antibodies (mAbs) which potently neutralize a broad range of HIV isolates are potential microbicide candidates. To date, topical application of mAbs in humans and their stability in vaginal secretions has not been studied. OBJECTIVES: To assess the pharmacokinetics and safety of the mAbs 2F5, 4E10 and 2G12 when applied vaginally in women. DESIGN: A randomized, double-blind, placebo-controlled phase 1 trial. METHODS: Twenty-eight healthy, sexually abstinent women administered 2.5 g of gel daily for 12 days containing either 10 or 20 mg/g of each mAb (MABGEL) or placebo

A prospective, randomized, double-blind study of vaginal microflora and epithelium in women using a tampon with an apertured film cover compared with those in women using a commercial tampon with a cover of nonwoven fleece.

Healthy women with normal menstrual cycles were randomly assigned to use either a test tampon during cycle 1 and a reference tampon during c … clinically significant. It is noteworthy, however, that declines in the prevalence and abundance of Lactobacillus during the menstrual periods were less pronounced during the use of both test and reference tampons than those reported from previous studies. Colposcopy showed no abnormal findings with either tampon and no changes in vaginal or cervical epithelial …

[The lactobacilli-protection system of pregnant women--efficient prevention of premature births by early detection of disturbances].

BACKGROUND: The most frequent cause of preventable early premature births (< 32 + 0 gestational weeks) is urogenital tract infection (UTI). Screening for UTI and Chlamydia are now standard examinations. Detection of ascending genital infections, especially bacterial vaginosis, plays an ever-increasing role. Lactobacilli, which produce, e. g., lactic acid, H2O2, biosurfactants and coaggregation molecules, are particularly important for the vaginal biosystem. Some evaluations suggest that early diagnosis and therapy within this "lactobacilli-protection system" apparently leads to better results

Maternal separation disrupts the integrity of the intestinal microflora in infant rhesus monkeys.

The integrity of the indigenous microflora of the intestines after maternal separation was investigated in infant rhesus monkeys to determine whether psychological stress may lead to an internal environment conducive to pathogen infection. The stability of the indigenous microflora were estimated by enumeration of total and gram-negative aerobic and facultatively anaerobic bacterial species, specifically Lactobacilli, from coprocultures taken before and after maternal separation. In addition, behavioral and cortisol responses to separation were correlated to the microflora. A significant decre

Prophylactic co-trimoxazole and lactobacilli preparation in neutropenic patients.

A randomized study of intestinal decontamination was undertaken in 68 children with leukemia and solid tumours. Framycetin, colymycin, nystatin, and metronidazole were given in 35 neutropenic episodes in 33 children, while co-trimoxazole and lactobacilli preparation were administered in 35 episodes in 35 children. The diseases, severity of neutropenia, and incidence of infection at entry into study were comparable in the two groups. There was no significant difference in the incidence of infections developing during the phase of neutropenia. The median and range of time required to recover fro

Metabolite-mediated crosstalk: unraveling the interactions between gut microbiota and host in fatty liver hemorrhagic syndrome of laying hens.

Fatty liver hemorrhagic syndrome (FLHS) in laying hens is a metabolic disorder characterized by excessive hepatic lipid accumulation, inflam … ammation, and oxidative stress, driving the development of FLHS. Key microbes, such as Bacteroides, Lactobacillus, and Akkermansia muciniphila, are considered potential therapeutic targets due to their involvement in metabolite production. Research advancement on the correlation between gut microbiota and chronic kidney disease.

Chronic kidney disease (CKD) represents a significant global health challenge, with its progression and complications associated with dysbiosis of the gut microbiota. Patients with CKD demonstrate stage-dependent alterations in the composition of gut microbiota and a reduction in diversity, which is characterized by a decline in beneficial bacteria (e.g., Bifidobacterium, Lactobacillus) and an increase in pathogenic species. This dysbiosis disrupts the gut-kidney axis, resulting in a depletion of protective metabolites such as short-chain fatty acids (SCFAs) and secondary bile acids, while fac…

Effects of Early Fructooligosaccharides Intervention on Growth Performance and Jejunal Development in Suckling Piglets.

Fructooligosaccharides (FOSs) are a class of functional prebiotics that effectively modulate the composition of the intestinal microbiota an … genes in the Wnt/β-catenin signaling pathway. This may be related to increased abundance of jejunal Lactobacillus and the concentrations of metabolites such as lactate and short-chain fatty acids (SCFAs). Early FOS intervention drove jejunal development through Wnt/β-catenin activation, a process mediated by microbiota-metabolite shifts that augmented intestina …
