Investigating the Pathology-Related Astroglial Plasticity in the Human Cerebral Cortex.

Evaluation of astrocyte proliferation in human brain pathology plays a key role in the diagnostic and prognostic assessment of diseases, especially in cases of benign or malignant brain neoplasms. However, the proliferative potential of nonneoplastic astrocytes within the affected human brain parenchyma has not been well defined. Given the beneficial functions of proliferating reactive astrocytes for brain repair in experimental models of stroke and trauma, investigating the context-specific potential of human astrocytes to proliferate will be a major step forward in exploiting their roles in …

Lifetime walking and Alzheimer's pathology: A longitudinal study in older adults.

IMPORTANCE: While many studies have shown that greater amounts or longer durations of walking are associated with a lower risk of Alzheimer's disease (AD) or cognitive decline in older adults, the neuropathological basis for this is not yet fully understood. OBJECTIVE: To examine the relationship between walking intensity and duration and longitudinal changes in Alzheimer's disease (AD)-related brain pathologies, including Aβ and tau accumulation, neurodegeneration, and white matter hyperintensity (WMH). DESIGN: Data were drawn from the Korean Brain Aging Study for the Early Diagnosis and Pr

Health disparities among incarcerated populations: a crucial laboratory and pathology review.

BACKGROUND: The United States notoriously has one of the highest rates of incarceration in the world, yet scant attention to the health care needs of those incarcerated exists within laboratory medicine and pathology training and education. This article explores health disparities among incarcerated and released individuals regarding diagnostic laboratory testing and pathology services. METHODS: A literature search was conducted for articles published between 2002 and 2023 using keywords including "healthcare," "incarcerated," "laboratory services," "pathology services," and "health insurance…

The Genetic Mechanisms and Pathology of Atrial Fibrillation: A Narrative Review.

Atrial fibrillation (AF), the most prevalent tachyarrhythmia worldwide, is a complex condition influenced by genetic, structural, and environmental factors. While AF in the elderly is often associated with underlying cardiac disease, early-onset or "lone" AF (LAF) exhibits a stronger genetic predisposition. Studies have identified both monogenic and polygenic contributors to AF risk. Monogenic mutations, inherited in Mendelian patterns, often affect ion channels and regulatory proteins, while polygenic variants modulate susceptibility and interact with environmental factors. Genome-wide associ

High-grade astrocytoma in a sheep: clinical, pathology, and genetic investigations.

Primary neoplastic diseases of the CNS in livestock are rare compared to companion animals. Here we describe the clinical, pathologic, and genetic findings in an adult sheep with a high-grade cerebral astrocytoma. Clinically, apathy, ataxia, bilateral amaurosis, and horizontal resting nystagmus were observed. The signs were suggestive of a space-occupying lesion affecting the vestibular and ascending reticular activating systems. The CSF had high total protein, hypercellularity, and pleocytosis. Macroscopically, a friable, gray-pink, ovoid mass (6 × 3 × 4 cm) was observed within most of the ri

Artificial intelligence-based quantitative bone marrow pathology analysis for myeloproliferative neoplasms.

The evaluation of bone marrow pathology is essential for diagnosing and classifying myeloproliferative neoplasms (MPN). However, morphological assessments of bone marrow trephine (BMT) sections by hematopathologists are inherently subjective; thus, an accurate and objective diagnostic system is needed. Based on U2-Net, UNeXt, and ResNet, we developed an automated quantitative analysis platform for BMT sections from MPN patients and non-neoplastic cases (N=342 total) to enhance the accuracy of diagnosis and classification of MPN. Bone marrow metrics, including marrow cellularity, the myeloid-to…

The role of m6A methylation in female reproductive physiology and pathology.

N6-methyladenosine (m6A) is a critical regulator of female reproductive physiology, yet existing reviews have focused predominantly on oocytes. The objective of this review is to systematically evaluate the regulatory effects of m6A throughout the pregnancy process. This review covers aspects such as oocyte maturation, granulosa cell dynamics, endometrial receptivity, immune homeostasis, and systemic adaptations, aiming to demonstrate the comprehensive regulatory capacity of m6A in female reproduction. Dysregulated m6A modifications in infertility-associated pathologies, including endometriosi

Traumatic brain injury. Rethinking pharmacological clinical trials in an orphan pathology.

INTRODUCTION: Every year more than 50 million people in the world experience a traumatic brain injury (TBI). In its more severe form the mortality is high, and survivors can be very disabled. Nevertheless, there are currently no approved pharmacological treatments that definitely improve the prognosis in humans, and given the consistently disappointing results, pharmaceutical companies are reluctant to invest further. AREAS COVERED: We reviewed relevant PubMed-indexed studies on pharmacological trials conducted during the acute phase of TBI. The potential reasons for the observed lack of effi

Forensic pathology and toxicological analyses: A case of fatal xylazine poisoning.

Xylazine, an α2-adrenergic receptor agonist commonly used as a veterinary sedative, has gained notoriety for its increasing misuse in combination with opioids, such as fentanyl, to potentiate its effects. This trend has led to a rise in cases of xylazine poisoning. Although timely diagnosis and intervention have reduced fatalities, the postmortem pathological manifestations and lethal blood concentration of xylazine alone remain poorly characterized due to its common co-occurrence in toxicology with fentanyl. This study presented a case of homicide involving fatal intoxication solely from xyla

Tau pathology differs by sex in Alzheimer's disease in Down syndrome.

INTRODUCTION: Alzheimer's disease (AD), the leading cause of dementia, is more common in females. Although sex differences in tau pathology have been reported in AD, findings remain inconsistent. Down syndrome (DS), caused by trisomy 21, is the most common genetic cause of AD (DS-AD) and features tau pathology, but sex effects in DS-AD remain unclear. METHODS: We examined post mortem brain samples from individuals with DS-AD, DS without AD, and a rare partial trisomy 21 (PT) case with only two amyloid precursor protein (APP) gene copies. PHF1 tau, total tau, and sarkosyl-soluble and insoluble…

Interplay and intervention of epigenetic dysregulation in traumatic brain injury pathology.

Traumatic brain injury (TBI) occurs when an external mechanical force damages brain tissue, leading to temporary or lasting disturbances in brain structure and function. The heterogeneous molecular and phenotypic nature of TBI poses a major challenge to translating basic research discoveries into clinically effective interventions. Emerging evidence indicates that epigenetic and epitranscriptomic mechanisms, including histone modifications, DNA methylation, and RNA modifications, play pivotal roles in the molecular response to TBI. In this review, we discuss post-TBI epigenomic alterations wit

Investigating and quantifying selection bias in research using placental pathology samples.

INTRODUCTION: Increasing research interest has focused on placental pathology and pregnancy outcomes. Placental pathology examinations that are requested by the delivering provider are often used for research due to convenience, potentially introducing selection bias. We leveraged a large cohort of prospectively collected and examined placentas to compare prevalences of placental pathology and to quantify potential selection bias. METHODS: All placentas were prospectively collected from participants and pathology examinations were completed in the Stress, Pregnancy, and Health (SPAH) study, r…

Editorial Commentary: Endoscopic Treatment of Hamstring Pathology Shows Positive Patient Outcomes.

Hamstring pathology is both a common, and commonly missed, source of pain. Acute traumatic tears can be missed or underappreciated, and hamstring tendinopathy and partial-thickness tears are often atraumatic with insidious onset. Partial-thickness hamstring tears or tendinopathy can be underappreciated on magnetic resonance imaging or missed in conjunction with concomitant pathology. Ultrasound-guided diagnostic injections are invaluable tools when sorting through concomitant pathologic processes. If noninvasive treatment options are unsuccessful, injections and surgery are options. Corticoste…
