Mitochondrial genome of Protobothrops jerdonii (Squamata: Viperidae: Crotalinae).

Protobothrops jerdonii is a common venomous snake that is widely distributed in southwestern China and other adjacent countries of Asia. In this study, the complete mitochondrial genome of P. jerdonii was determined. The circle genome with the 17,239 bp total length contained 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes, and 2 control regions. Overall base composition of the complete mtDNA was 33.13% A, 25.07% T, 29.31% C, and 12.50% G. All the genes in P. jerdonii were distributed on the H-strand, except for the ND6 subunit gene and eight tRNA genes which were encoded

Is the mitochondrial complex I ND5 gene a hot-spot for MELAS causing mutations?

We identified two novel heteroplasmic mitochondrial DNA point mutations in the gene encoding the ND5 subunit of complex I: a 12770A-->G transition identified in a patient with MELAS (mitochondrial encephalomyopathy with lactic acidosis and stroke-like episodes) and a 13045A-->C transversion in a patient with a MELAS/Leber's hereditary optic neuropathy/Leigh's overlap syndrome. Biochemical analysis of muscle homogenates showed normal or very mildly reduced complex I activity. Histochemistry was normal. Our observations add to the evidence that mitochondrial ND5 protein coding gene mutations fre

Purification and characterization of glutathione transferase of human thyroid.

An anionic (pI 4.6) isoenzyme of glutathione transferase was purified to homogeneity from human thyroid by affinity chromatography followed by isoelectric focusing. The content of enzyme was calculated to constitute about 0.2% of soluble proteins. The enzyme is formed by two identical subunits of 23,000 daltons approximately. The thyroid transferase did not catalyze the reduction of peroxides. Physical, catalytic and immunological analyses demonstrated extensive similarities between the thyroid transferase and the transferase from placenta, erythrocytes and breast. On the other hand, the thyro

[Characteristics of a low-molecular nonvirion ("soluble") antigen from the tick-borne encephalitis virus].

Pig embryo kidney and BHK-21 cells infected with tick-borne encephalitis virus synthesize a nonvirion antigen differing in its immunological properties from the virion antigens. The antigen has a high thermostability. According to the results of ultrafiltration and gel filtration, its molecular weight is approximately 70-100 kilodaltons. Electrophoretic analysis in polyacrylamide gel showed the molecular weight of a polypeptide isolated from the precipitation band formed by this low molecular antigen to differ from molecular weights of virion and high molecular virus-specific nonvirion protein

Engineering a dimeric porcine deltacoronavirus S-RBD subunit vaccine for enhanced immunogenicity and neutralizing antibody responses.

BACKGROUND: Porcine deltacoronavirus (PDCoV) is an emerging swine enteric coronavirus that has caused global epidemics and substantial economic losses, posing a potential risk of cross-species transmission, including to humans. However, commercial vaccines are currently scarce. The receptor binding domain (RBD) of the spike (S) protein is a key target for inducing neutralizing antibodies, but the RBD monomer typically exhibits low molecular weight and weak immunogenicity. This study aimed to develop a tandem dimeric RBD subunit vaccine and evaluate its immunogenicity and protective efficacy in

cAMP/PKA signaling in endocrine hypertension: genetic mechanisms and pathophysiological insights.

The cyclic adenosine monophosphate (cAMP)-protein kinase A (PKA) signaling pathway plays a central role in adrenal function, steroidogenesis, and blood pressure regulation. Increasing evidence suggests that dysregulation of this pathway contributes to several forms of hypertension, both endocrine and non-endocrine. A growing number of germline and somatic alterations affecting components of the cAMP/PKA axis have been implicated as key drivers of hypertensive disorders. Among these, activating pathogenic variants (PV) in GNAS, which encodes the stimulatory G protein α-subunit (Gsα) responsible

Neuronal overexpression of Kcnn1 in A53T α-synuclein mice suppresses phospho-serine 129 α-synuclein formation and doubles survival time.

Synucleinopathies, including idiopathic Parkinson's Disease, are driven by misfolding and aggregation of the 140 residue α-synuclein protein that plays a role in presynaptic vesicle regulation. We describe effects of a modifier, neuronal overexpression of the mouse calcium-activated potassium channel subunit Kcnn1, on a mouse model in which transgenic Thy1.2-driven human A53T α-synuclein directs fully penetrant lethal motor disease. Kcnn1 overexpression increased median survival of these mice from 8.5 to 18 mo, associated with an altered presentation from rapidly progressive dystonic-like beha

A novel albumin-based nanoparticle vaccine including the globular knob domain of the egg drop syndrome virus fiber protein for egg drop syndrome: Immunogenicity and preliminary structural analyses.

Egg drop syndrome (EDS), caused by the avian adenovirus known as EDS virus (EDSV), is a viral disease characterized by reduced egg production and abnormal egg quality in laying hens, resulting in substantial economic losses. While conventional vaccines can have some safety issues, the subunit vaccines circumvent these concerns. The globular knob domain of the EDSV fiber protein (EDSVFP) harbors critical neutralizing epitopes, making it an ideal vaccine target. In this study, chicken serum albumin nanoparticles (CSA-NPs) were employed as a delivery vehicle. The coding sequence for the knob doma

Development and evaluation of a multi-epitope subunit vaccine against Salmonella Enteritidis infection.

Salmonella Enteritidis (S. Enteritidis) is a major pathogen causing foodborne diseases worldwide, posing a serious threat to public health security and the sustainable development of the poultry industry. To overcome the limitations of conventional vaccines in terms of efficacy, safety, and cross-protection, a novel multi-epitope subunit vaccine (SEMV) was designed and constructed using an immunoinformatics-based strategy. Through systematic screening of epitopes from key immunogenic proteins of S. Enteritidis (SseB, SipC, OmpD and TolC), a total of 8 B-cell epitopes, 4 MHC-I-restricted epitop

Lavandula angustifolia essential oil attenuates chronic prostatitis by inhibiting PI3K/AKT and NF-κB signaling pathways.

Although lavandula angustifolia essential oil (LEO) has demonstrated anti-inflammatory properties, its mechanism in alleviating chronic prostatitis (CP) remains unclear. This study aimed to explore the pharmacological basis and molecular mechanisms of LEO's effect on CP. The chemical composition of LEO was analyzed using Gas Chromatography-Mass Spectrometry (GC-MS), identifying 15 active ingredients. A carrageenan-induced rat CP model was used to evaluate the impact of LEO on prostate index, inflammatory markers, and histopathological changes. The results showed that LEO significantly reduced

Enhancing humoral and mucosal immune response of PED vaccine candidate by fusing S1 protein to nanoparticle multimerization.

Porcine epidemic diarrhea virus (PEDV) is a highly infectious pathogen with a high mortality rate, which poses a serious threat to newborn piglets. A rapid, safe and effective vaccine is necessary for protecting pigs from PED infection. Nanoparticles have become molecular scaffolds for displaying soluble antigens due to their unique physical and chemical properties. Here, a vaccine candidate was based on the display of PEDV S1 protein on a mi3 nanoparticle platform using SpyTag/SpyCatcher technology. The size, zeta potential and microstructure of the S1-mi3 NPs were investigated, and their eff

Autophagy-dependent hepatocyte secretion of DBI/ACBP induced by glucocorticoids determines the pathogenesis of Cushing syndrome.

DBI/ACBP is a phylogenetically ancient hormone that stimulates appetite and lipo-anabolism. In response to starvation, DBI/ACBP is secreted through a noncanonical, macroautophagy/autophagy-dependent pathway. The physiological hunger reflex involves starvation-induced secretion of DBI/ACBP from multiple cell types. DBI/ACBP concentrations subsequently increase in extracellular fluids to stimulate food intake. Recently, we observed that glucocorticoids, which are endogenous stress hormones as well as anti-inflammatory drugs, upregulate DBI/ACBP expression at the transcriptional level and stimula

Protein Kinase CK2 Modulates the Calcium Sensitivity of Type 3 Small-conductance Calcium-activated Potassium Channels in Colonic Platelet-derived Growth Factor Receptor Alpha-positive Cells From Streptozotocin-induced Diabetic Mice.

BACKGROUND/AIMS: The gastrointestinal symptom of diabetes mellitus, chronic constipation, seriously affects patients' life. Whereas, the mechanism of chronic constipation is still ambiguous, resulting in a lack of effective therapies for this symptom. As a part of the smooth muscle cells, interstitial cells of Cajal, and platelet-derived growth factor receptor alpha-positive (PDGFRα+) cells syncytium (SIP syncytium), PDGFRα+ cells play an important role in regulating colonic motility. According to our previous study, in PDGFRα+ cells in colons of diabetic mice, the function of the P2Y1 puriner

Berberine ameliorates pulmonary inflammation in mice with influenza viral pneumonia by inhibiting NLRP3 inflammasome activation and gasdermin D-mediated pyroptosis.

Viral pneumonia is a common complication caused by Influenza A virus infection and is characterized by severe pulmonary inflammation. A previous study showed that berberine (BBR) significantly ameliorated the pulmonary inflammation in mice with influenza viral pneumonia but its underlying mechanism is not entirely understood. In this study, we reproduced the mouse model of influenza viral pneumonia through intranasal infection of A/Puerto Rico/8/34 (H1N1), to further investigate the anti-inflammatory mechanism of BBR based on nucleotide-binding oligomerization domain-like receptor protein (NLR

Proof of Concept for a Quick and Highly Sensitive On-Site Detection of SARS-CoV-2 by Plasmonic Optical Fibers and Molecularly Imprinted Polymers.

The rapid spread of the Coronavirus Disease 2019 (COVID-19) pandemic, caused by the Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) pathogen has generated a huge international public health emergency. Currently the reference diagnostic technique for virus determination is Reverse Transcription Polymerase Chain Reaction (RT-PCR) real time analysis that requires specialized equipment, reagents and facilities and typically 3-4 h to perform. Thus, the realization of simple, low-cost, small-size, rapid and point-of-care diagnostics tests has become a global priority. In response to the

Genomic Epidemiology, Evolution, and Transmission Dynamics of Porcine Deltacoronavirus.

The emergence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has shown once again that coronavirus (CoV) in animals are potential sources for epidemics in humans. Porcine deltacoronavirus (PDCoV) is an emerging enteropathogen of swine with a worldwide distribution. Here, we implemented and described an approach to analyze the epidemiology of PDCoV following its emergence in the pig population. We performed an integrated analysis of full genome sequence data from 21 newly sequenced viruses, along with comprehensive epidemiological surveillance data collected globally over the l
