Mechanistic learning to predict and understand minimal residual disease.

Mechanistic modeling has long been used as a tool to describe the dynamics of biological systems, especially cancer in response to treatment. Their key advantage lies in interpretability of relationships between input parameters and outcomes of interest. Mechanistic models may also be calibrated to a cohort of patients and scaled up to generate a simulated set of virtual patients whose aggregate behavior reproduces key characteristics of the real patient population. In contrast, machine learning techniques offer strong prediction performance, especially for high dimensional datasets that are c

Effects of 5D built environment and non-built-environment factors on injury crash risk: An interpretable machine learning analysis.

To identify the key determinants of traffic injury risk and clarify the relative roles of built environment factors and crash-context factors, this study develops a traffic injury risk identification framework that integrates 5D built environment variables with non-5D factors, using traffic crash data collected in Changsha, Hunan Province, China, from 2017 to 2019. After data cleaning, screening, and spatial matching, a total of 9,743 valid samples were obtained, and injury occurrence in a crash was defined as a binary dependent variable. On this basis, three feature sets were constructed, inc

A systematic review and meta-analyses of glucagon-like peptide-1 receptor agonists in acute myocardial infarction.

BACKGROUND: Previous research has shown that glucagon-like peptide-1 receptor agonists (GLP-1 RAs) have cardioprotective effects. However, their efficacy in acute myocardial infarction (AMI) is not yet well established. We conducted a systematic review and meta-analyses to assess the impact of GLP-1 RAs in AMI. METHODS: We systematically searched major databases for studies in patients with ST-elevation (STEMI) or non-ST-elevation myocardial infarction (NSTEMI). These included MEDLINE (accessed via PubMed), Excerpta Medica dataBASE (Embase), Cochrane Central Register of Controlled Trials (CEN

Impact of ultra-low tidal volume ventilation on 1-year functional outcome in COVID-19 ARDS patients. A long-term follow-up analysis of a randomized controlled trial.

BACKGROUND: Ultra-low tidal volume ventilation (ULTV) aims to minimize ventilator-induced lung injury in acute respiratory distress syndrome (ARDS), but is associated with an increase in arterial carbon dioxide (PaCO2). This follow-up study aimed to evaluate whether ULTV impacted long-term pre-specified secondary outcomes in the VT4COVID trial. METHODS: VT4COVID was an open-label multicenter randomized controlled superiority trial performed in ten French intensive care units. Eligible patients were COVID-19 ARDS patients with arterial oxygen pressure (PaO2) over inspired oxygen fraction (FiO2

piR-27222 alleviates dexamethasone-induced osteoporosis by inhibiting ferroptosis through modulating WWP1.

BACKGROUND: Glucocorticoid-induced osteoporosis (GIOP) is the most common cause of secondary osteoporosis. Piwi-interacting RNA (piRNA) represents a novel class of small non-coding RNAs. However, the role and potential mechanism of piRNA in GIOP remain unclear. METHODS: Differentially expressed genes (DEGs) during the chondrogenic and osteogenic differentiation of human bone marrow mesenchymal stem cells (hBMSCs) and in osteoblasts treated with dexamethasone were screened. Osteogenic differentiation and adipogenic differentiation of hBMSCs were induced, and a rat GOIP model was constructed. qR

Transcatheter Versus Sutureless Aortic Valve Replacement: A Propensity-Matched Single-Center Cohort Study.

Background and Objectives: Over the past decade, transcatheter aortic valve replacement (TAVI) has evolved from a treatment for inoperable patients to an established option across all risk categories. In parallel, the Perceval sutureless valve has demonstrated safety and efficacy especially for minimally invasive surgical aortic valve replacement (AVR). Despite the advances of both TAVI and Perceval, robust long-term data and clear patient selection criteria are still lacking. This retrospective single-center study reports the outcomes of patients undergoing isolated AVR with the Perceval sutu

Low Serum C-Peptide and Albuminuria Severity in Type 2 Diabetes Mellitus: A Cross-Sectional Study.

Background/Objectives: Lower serum C-peptide, reflecting reduced beta-cell reserve, has been linked to diabetic complications, but whether these associations are independent of diabetes duration and glycaemic control is unclear. The objective of this study was to determine which microvascular and macrovascular complications of type 2 diabetes mellitus (T2DM) remain associated with fasting C-peptide after adjustment for these factors. Methods: In this single-centre, cross-sectional study with consecutive prospective enrolment, non-pregnant adults with an established diagnosis of T2DM attending

Neurological complication burden and in-hospital mortality in non-HIV tuberculous meningitis across the lifespan: a retrospective cohort study.

BACKGROUND: Tuberculous meningitis (TBM) is a severe form of extrapulmonary tuberculosis and is frequently complicated by neurological injury. Existing studies have usually evaluated individual complications or admission-based prognostic models, whereas cumulative neurological complication burden documented across a hospitalization remains poorly characterized in non-human immunodeficiency virus (HIV) cohorts spanning pediatric and adult patients. We evaluated whether neurological complications present at admission or documented later during the index hospitalization were associated with in-ho

Using Machine Learning Approaches on Dynamic Patient-Reported Outcomes to Cluster Cancer Treatment-Related Symptoms.

In patients undergoing systemic treatment for cancer, symptom tracking via electronic patient-reported outcomes (ePROs) has been used to optimize communication and monitoring, and facilitate the early detection of adverse effects and to compare the side effects of similar drugs. We aimed to examine whether the patterns in electronic patient-reported outcomes, without any additional clinician data input, are predictive of the underlying cancer type and reflect tumor- and treatment-associated symptom clusters (SCs). The data were derived from a total of 226 patients who self-reported on the pres

An Online Prediction Method for Transient Frequency Response in New Energy Grids Based on Deep Integration of WAMS Data and Physical Model.

The integration of a high proportion of renewable energy has significantly reduced the grid inertia level and markedly increased the risk of transient frequency instability in power systems. Meanwhile, the large-scale integration of diverse heterogeneous resources-such as wind power, photovoltaics, energy storage, and high voltage direct current (HVDC) transmission systems-has considerably enriched the portfolio of frequency regulation assets in modern power grids. However, the marked disparities in the dynamic response characteristics and actuation speeds among these resources introduce signi

Amoxicillin-loaded polycaprolactone-gelatin nanofiber/net (NFN) matrices for pneumothorax treatment.

The recurrence rate of spontaneous pneumothorax (collapsed lung) is 30 % after basic conservative treatments, and it can reach up to 20 % after surgery due to the limited effectiveness of current surgical techniques. This highlights the need for the development of new, effective treatment approaches supported by biomaterials. Therefore, we aimed to develop a biomaterial that does not restrict the aerodynamic movements of the lungs, is resistant to lung pressure, and can withstand the movements during inhalation and exhalation,to prevent and treat pneumothorax recurrence. For this purpose, biod
