Brain tissue mechanics is governed by microscale relations of the tissue constituents.

Local mechanical tissue properties are a critical regulator of cell function in the central nervous system (CNS) during development and disorder. However, we still don't fully understand how the mechanical properties of individual tissue constituents, such as cell nuclei or myelin, determine tissue mechanics. Here we developed a model predicting local tissue mechanics, which induces non-affine deformations of the tissue components. Using the mouse hippocampus and cerebellum as model systems, we show that considering individual tissue components alone, as identified by immunohistochemistry, is

A bioengineered peripheral nerve construct using aligned peptide amphiphile nanofibers.

Peripheral nerve injuries can result in lifelong disability. Primary coaptation is the treatment of choice when the gap between transected nerve ends is short. Long nerve gaps seen in more complex injuries often require autologous nerve grafts or nerve conduits implemented into the repair. Nerve grafts, however, cause morbidity and functional loss at donor sites, which are limited in number. Nerve conduits, in turn, lack an internal scaffold to support and guide axonal regeneration, resulting in decreased efficacy over longer nerve gap lengths. By comparison, peptide amphiphiles (PAs) are mole

Stem-Cell-Derived Biologic Ventricular Assist Tissue in Heart Failure.

BACKGROUND: Biologic ventricular assist tissue (BioVAT) is formulated from engineered heart muscle composed of cardiomyocytes and stromal cells derived from allogeneic induced pluripotent stem cells for cardiac remuscularization in patients with heart failure and a reduced left ventricular ejection fraction. METHODS: We conducted an open-label, phase 1-2 study of tissue-engineered heart repair by means of BioVAT transplantation. Patients with heart failure and a left ventricular ejection fraction of 35% or less and at least one hypokinetic or dyskinetic left ventricular segment were treated w

Engineered Regulatory T Lymphocytes Promote Infarcted Heart Repair.

BACKGROUND: Myocardial infarction (MI) initiates a dysregulated healing process characterized by excessive fibrosis and unresolved inflammation, resulting in suboptimal cardiac repair in clinical settings. Regulatory T lymphocytes (Tregs) naturally orchestrate cardiac repair after MI, but their therapeutic potential is limited by inefficient homing to ischemic myocardium. We hypothesize that FAP (fibroblast activation protein)-specific CAR (chimeric antigen receptor) engineering overcomes this barrier by enabling precise delivery of Tregs to FAP⁺-enriched infarct zones, thereby focally amplify

Loss of sarcomeric proteins via upregulation of JAK/STAT signaling underlies interferon-γ-induced contractile deficit in engineered human myocardium.

The level of circulating interferon-γ (IFNγ) is elevated in various clinical conditions including autoimmune and inflammatory diseases, sepsis, acute coronary syndrome, and viral infections. As these conditions are associated with high risk of myocardial dysfunction, we investigated the effects of IFNγ on 3D fibrin-based engineered human cardiac tissues ("cardiobundles"). Cardiobundles were fabricated from human pluripotent stem cell-derived cardiomyocytes, exposed to 0-20 ng/ml of IFNγ on culture days 7-14, and assessed for changes in tissue structure, viability, contractile force and calcium

Arterial reconstruction with human bioengineered acellular blood vessels in patients with peripheral arterial disease.

OBJECTIVE: Vascular conduit is essential for arterial reconstruction for a number of conditions, including trauma and atherosclerotic occlusive disease. We have developed a tissue-engineered human acellular vessel (HAV) that can be manufactured, stored on site at hospitals, and be immediately available for arterial vascular reconstruction. Although the HAV is acellular when implanted, extensive preclinical and clinical testing has demonstrated that the HAV subsequently repopulates with the recipient's own vascular cells. We report a first-in-man clinical experience using the HAV for arterial r

BMP-2 gene activated muscle tissue fragments for osteochondral defect regeneration in the rabbit knee.

BACKGROUND: Previously published data indicate that BMP-2 gene activated muscle tissue grafts can repair large bone defects in rats. This innovative abbreviated ex vivo gene therapy is appealing because it does not require elaborative and time-consuming extraction and expansion of cells. Hence, in the present study, we evaluated the potential of this expedited tissue engineering approach for regenerating osteochondral defects in rabbits. METHODS: Autologous muscle tissue grafts from female White New Zealand rabbits were directly transduced with an adenoviral BMP-2 vector or remained unmodifie…

Variability in responses observed in human white adipose tissue models.

Obesity is a risk factor for a myriad of diseases including diabetes, cardiovascular dysfunction, cirrhosis, and cancer, and there is a need for new systems to study how excess adipose tissue relates to the onset of disease processes. This study provides proof-of-concept patient-specific tissue models of human white adipose tissue to accommodate the variability in human samples. Our 3D tissue engineering approach established lipolytic responses and changes in insulin-stimulated glucose uptake from small volumes of human lipoaspirate, making this methodology useful for patient specific sample s…

Targeting, Mutagenesis, and Functional Testing Strategies for Large Collagens.

Collagens comprise a homogeneous family of cell-surface or extracellular proteins. Many of them are fundamental for health, and consequently, their deficiency or dysregulation occurs in a wide range of diseases, from tissue fragility to neoplastic and fibrosing diseases, to name a few. With the advent of new omics approaches, an increasing number of variants of specific collagens have been discovered in various conditions. Some of these variants lead to a loss of expression, highlighting the need to assess the functions and loss of specific collagens in such conditions, while other variants ma

New frontiers in modeling tuberous sclerosis with human stem cell-derived neurons and brain organoids.

Recent advances in human stem cell and genome engineering have enabled the generation of genetically defined human cellular models for brain disorders. These models can be established from a patient's own cells and can be genetically engineered to generate isogenic, controlled systems for mechanistic studies. Given the challenges of obtaining and working with primary human brain tissue, these models fill a critical gap in our understanding of normal and abnormal human brain development and provide an important complement to animal models. Recently, there has been major progress in modeling the

Development of Injectable Amniotic Membrane Matrix for Postmyocardial Infarction Tissue Repair.

Ischemic heart disease represents the leading cause of death worldwide. Heart failure following myocardial infarction (MI) is associated with severe fibrosis formation and cardiac remodeling. Recently, injectable hydrogels have emerged as a promising approach to repair the infarcted heart and improve heart function through minimally invasive administration. Here, a novel injectable human amniotic membrane (hAM) matrix is developed to enhance cardiac regeneration following MI. Human amniotic membrane is isolated from human placenta and engineered to be a thermoresponsive, injectable gel around

Prospects for gene-engineered T cell immunotherapy for solid cancers.

Adoptive transfer of receptor-engineered T cells has produced impressive results in treating patients with B cell leukemias and lymphomas. This success has captured public imagination and driven academic and industrial researchers to develop similar 'off-the-shelf' receptors targeting shared antigens on epithelial cancers, the leading cause of cancer-related deaths. However, the successful treatment of large numbers of people with solid cancers using this strategy is unlikely to be straightforward. Receptor-engineered T cells have the potential to cause lethal toxicity from on-target recogniti

Extracellular vesicles as biomimetic platforms for photo-assisted cancer therapy: evolution from synthetic nanocarriers to engineered therapeutic systems.

Photo-assisted therapies have emerged as promising strategies for cancer treatment by combining localized tumor ablation with the induction of systemic antitumor immune responses. Despite encouraging preclinical and clinical advances, their therapeutic efficacy remains limited by the unfavorable pharmacokinetics, poor tumor selectivity, restricted tissue penetration, and off-target toxicity of conventional photosensitizer formulations. Although synthetic nanocarriers have substantially improved photosensitizer delivery, their limited biological functionality has driven the development of biomi

3D printed porous tantalum loaded with quercetin-curcumin-piperine PLGA nanoparticles for bone defect repair.

The repair of irregular bone defects poses a significant challenge for orthopedic surgeons. This study presents a customizable, novel bone tissue engineering scaffold capable of promoting bone regeneration, which is composed of curcumin-quercetin-piperine-loaded poly(lactic-co-glycolic acid) nanoparticles (Cur-Que-Pip-PLGA NPs) loaded onto 3D-printed porous tantalum (3DPTa). The Cur-Que-Pip-PLGA NPs were prepared via the emulsification-solvent evaporation method, and subsequently loaded onto the 3DPTa scaffold via the physical impregnation method. The morphology and structure of the composite …

Biomimetic 3D-printed bilayered scaffold with stem cell delivery for rabbit vaginal regeneration.

Traditional biological scaffolds for vaginal reconstruction often fail to restore the physiological bilayered structure of vaginal tissue, leading to post-operative complications such as contraction, stenosis, and fibrosis. To address this, we developed a biomimetic bilayered vaginal scaffold using decellularized porcine vaginal extracellular matrix (vECM), gelatin methacryloyl (GelMA), and silk fibroin (SF) hydrogels. The optimized bioink (vECM/GelMA/SF) was characterized via SEM, rheology, FTIR, swelling tests, and H&E staining, confirming its morphology, stability, and biocompatibility. Rab

The effect of human umbilical cord mesenchymal stem cells combined with concentrated growth factor on repairing necrotic pulp caused by dental caries.

This study investigated the impact of combining human umbilical cord mesenchymal stem cells (hUC-MSCs) with concentrated growth factor (CGF) on regenerating necrotic pulp. Ten-month-old male Bama miniature pigs were divided into control and caries groups. The experimental teeth were randomly divided into three groups: caries untreated, Ca(OH)2, and engineering dental pulp-like tissue (EDPT). hUC-MSCs and CGF scaffold were combined to construct EDPT, and the histological structure was observed. Odontoblasts and dental pulp cells were counted in each group. The results showed that hUC-MSCs adher
