Primary afferent projections to the central octavus nuclei in the elasmobranch, Rhinobatos sp., as demonstrated by nerve degeneration.

Nerve degeneration studies were performed to determine the central projections of the octavus nerve in the guitarfish. Rhinobatos sp. Most first-order octavus nerve afferents are confined to the ventral longitudinal column of the medulla, which consists of four octavus nuclei: the nucleus octavus anterior (NOA), nucleus octavus magnocellularis (NOM), nucleus octavus descendens (NOD); and nucleus octavus posterior (NOP). Central projections of the octavus nerve outside of the ventral octavus nuclear column are to the vestibulolateral lobe of the cerebellum, the large multipolar cells that lie d

Sensory denervation of the kidney attenuates renovascular hypertension in the rat.

To determine the role of the renal afferent nerves in the pathogenesis of one-kidney, one-clip renovascular hypertension, the renal afferent nerves were selectively lesioned by dorsal rhizotomy, a procedure that eliminates renal sensory input to the spinal cord but does not directly damage the sympathomotor innervation of the kidney. One week after denervation, the proximal left renal artery was clipped in denervated and sham control rats. Blood pressure of the sham group rose progressively over the next 5 wk, to 185 mmHg (systolic). In contrast, blood pressure of the denervated rats leveled o

A projection from the periaqueductal grey to the lateral reticular nucleus in the cat.

A projection from the periaqueductal grey (PAG) to the lateral reticular nucleus (NRL) in the cat was demonstrated by means of retrograde transport of the wheat germ agglutinin-horseradish peroxidase complex. The connection has its main origin ipsilaterally in the ventral part of the caudal PAG, but scanty projections from other parts of the PAG were also found. The neurons projecting to the NRL are of varying shapes and sizes, but most cells have a maximum diameter of less than 20 micron. The findings are discussed in relation to the other afferent and efferent connections of the NRL.

Lack of sprouting and its presence after lesions of the cat spinal cord.

Degeneration methods were used to study the dorsal root and descending projections after chronic partial denervation of adult cat spinal cord. Conventional mapping methods were used, supplemented in some cases by densitometric measurements of the amounts of degeneration present. The amount of shrinkage of spinal gray matter in some sections was estimated by planimetric measurement. Two preparations were used: (1) partial unilateral rhizotomy in which all dorsal roots caudal to T4 were cut except L6 (spared root preparation); (2) complete unilateral deafferentation. The projection of L6 dorsal

Properties of intraoral mechanoreceptors represented in the mesencephalic nucleus of the fifth nerve in the cat.

1. The activity of neurones in the mesencephalic nucleus of the fifth nerve that respond to forces applied to the teeth were recorded using extracellular microelectrodes; the properties of these neurones have been studied. 2. Electrophysiological evidence consistent with the view that primary afferent intraoral mechanoreceptor fibres have their cell bodies in the trigeminal mesencephalic nucleus is presented. 3. Two groups of intraoral mechanoreceptor neurones were found. The first group, the periodontal mechanoreceptor neurones, which have been described by previous workers, responded to elec

Tactile projections to granule cells in caudal vermis of the rat's cerebellum.

We discovered a small tactile area in a single a folium of the uvula of the cauday vermis of the rat's cerebellum. Gentle mechanical stimulation of relatively small cutaneous receptive fields (RFs) activated multiple units in the granule cell (GC) layer in a portion of a single folium in rats anesthetized with sodium pentobarbital. The total size of this area on each side of the midline is about 1.5 mm2, yet micromapping within this tiny region using tungsten ball microelectrodes and a high puncture sampling density (about 75 punctures/mm2) revealed a highly differentiated pattern of cutaneous

Direct accessory optic projections to the vestibulo-cerebellum: a possible channel for oculomotor control systems.

The nucleus of the basal optic root (nBOR) receives direct retinal projections in all classes of vertebrates. This nucleus is also known as the medial terminal nucleus of the accessory optic tract, or as the nucleus extomamillaris. Following a series of HRP injections into the uvula and flocculonodular lobe of pigeons, both large and small cells of the nBOR were labelled bilaterally with the marker. No significant transport of HRP to nBOR was observed following injections of more rostral folia of the posterior or anterior lobes of the cerebellum. In view of the prominence of the tract from ret

Rac1 and Nectin3 are essential for planar cell polarity-directed axon guidance in the peripheral auditory system.

Spiral ganglion neurons (SGNs) carry sound information from the cochlea to the hindbrain, and innervate either inner or outer hair cells. Type II SGNs (SGNIIs) extend peripheral afferents towards outer hair cells, which make a characteristic 90° turn towards the cochlear base and innervate multiple outer hair cells. It has been shown that the planar cell polarity (PCP) pathway acts non-autonomously in the cochlear epithelium to guide SGNII peripheral afferent turning. However, the underlying mechanisms are unknown. Here, we show that PCP signaling regulates junctional localization of the small

Cholinergic and neurogenic mechanisms in obstructive airways disease.

Although primary neural control of airway function is through parasympathetic pathways, more recent evidence indicates that there are important adrenergic and non-adrenergic, non-cholinergic neural mechanisms that may also influence respiratory function. The parasympathetic nervous system component includes neural receptors in the airways as well as afferent and efferent pathways that travel in the vagus nerves. Afferent vagal sensory receptors mediate the response to irritant or rapidly adapting receptor activation, Hering-Breuer, and the unmyelinated "C" fibers or "J" receptor pathways. The

Practical Perspectives in the Treatment of Nausea and Vomiting.

Nausea and vomiting result from complex interactions between afferent and efferent pathways of the gastrointestinal tract, central nervous system, and autonomic nervous system. Afferent pathways from the vagus nerve, vestibular system, and chemoreceptor trigger zone project to nucleus tractus solitarius, which in turn relays signals to the central pattern generator to initiate multiple downstream pathways resulting in symptoms of nausea and vomiting. There is increasing evidence that the central pathway of chronic nausea is different from that of acute nausea and vomiting-and closely resembles…

Effects of hip joint angle changes on intersegmental spinal coupling in human spinal cord injury.

Pathological expression of movement and muscle tone in human upper motor neuron disorders has been partly associated with impaired modulation of spinal inhibitory mechanisms, such as reciprocal or presynaptic inhibition. In addition, input from specific afferent systems contributes significantly to spinal reflex circuits coupled with posture or locomotion. Accordingly, the objectives of this study were to identify the involved afferents and their relative contribution to soleus H-reflex modulation induced by changes in hip position, and to relate these effects with activity of spinal interneur

Peripheral neural circuits regulating IOP? A review of its anatomical backbone.

The peripheral nervous system is classically separated into a somatic division containing both afferent and efferent pathways and an autonomic division composed of efferents only. The somatic afferent division is divided in A- and B-neurons. The B-neurons are supposed to be autonomic afferents as part of a reflex system involved in homeostasis. Recent data obtained by neuronal tracing and immunohistochemical experiments concerning the eye related peripheral nervous system endorse the existence of these peripheral reflex systems. Somatic afferents of trigeminal origin synaptically innervate par

Proteomic analysis of exosomes from lymphatic affluents reveals their implications in developing premetastatic niche in melanoma.

Melanoma is an aggressive form of skin cancer that often spreads via lymphatic pathways to regional and distant sites. Melanoma-derived lymphatic exosomes play a crucial role in forming a tumor-supportive environment for metastasis, or premetastatic niche, within the first tumor draining lymph node, also known as the sentinel lymph node (SLN). Therefore, analyzing the proteomic content of tumor-draining lymphatic exosomes that deliver oncogenic molecules to the SLN is important in understanding the premetastatic niche. To reveal the proteomic landscape of lymphatic exosomes, we performed multi

Transneuronal pathways to the vestibulocerebellum.

The alpha-herpes virus (pseudorabies, PRV) was used to observe central nervous system (CNS) pathways associated with the vestibulocerebellar system. Retrograde transneuronal migration of alpha-herpes virions from specific lobules of the gerbil and rat vestibulo-cerebellar cortex was detected immunohistochemically. Using a time series analysis, progression of infection along polyneuronal cerebellar afferent pathways was examined. Pressure injections of > 20 nanoliters of a 10(8) plaque forming units (pfu) per ml solution of virus were sufficient to initiate an infectious locus which resulted in…

Inflammation of some visceral sensory systems and autonomic dysfunction in cardiovascular disease.

The sensitization and hypertonicity of visceral afferents are highly relevant to the development and progression of cardiovascular and respiratory disease states. In this review, we described the evidence that the inflammatory process regulates visceral afferent sensitivity and tonicity, affecting the control of the cardiovascular and respiratory system. Some inflammatory mediators like nitric oxide, angiotensin II, endothelin-1, and arginine vasopressin may inhibit baroreceptor afferents and contribute to the baroreflex impairment observed in cardiovascular diseases. Cytokines may act directl

Nitric oxide as an endogenous peripheral modulator of visceral sensory neuronal function.

Nitric oxide (NO) plays important roles in CNS and smooth muscle function. Here we reveal an additional function in peripheral sensory transmission. We hypothesized that endogenous NO modulates the function of gastrointestinal vagal afferent endings. The nonselective NO synthase (NOS) inhibitor N(G)-nitro-L-arginine methyl ester hydrochloride increased responses to tactile mechanical stimuli of mucosal afferent endings in two species, in some cases severalfold. This was mimicked by a neuronal NOS inhibitor but not an endothelial NOS inhibitor. NOS inhibitors did not affect the responsiveness o

Triggering genetically-expressed transneuronal tracers by peripheral axotomy reveals convergent and segregated sensory neuron-spinal cord connectivity.

To better understand the mechanisms through which non-painful and painful stimuli evoke behavior, new resources to dissect the complex circuits engaged by subsets of primary afferent neurons are required. This is especially true to understand the consequences of injury, when reorganization of central nervous system circuits likely contributes to the persistence of pain. Here we describe a transgenic mouse line (ZWX) in which there is Cre-recombinase-dependent expression of a transneuronal tracer, wheat germ agglutinin (WGA), in primary somatic or visceral afferent neurons, but only after trans
