Structure of human aldehyde oxidase under tris(2-carboxyethyl)phosphine reducing conditions.

The importance of human aldehyde oxidase (hAOX1) has increased in recent decades due to its involvement in drug metabolism. Inhibition studies involving hAOX1 are extensive and a common reducing agent, dithiothreitol (DTT), was recently found to inactivate the enzyme. However, in previous crystallographic studies of hAOX1, DTT was found to be essential for crystallization. To surpass this concern, another reducing agent was used in crystallization trials. Using tris(2-carboxyethyl)phosphine (TCEP), a sulfur-free reducing agent, it was possible to obtain well ordered crystals of wild-type hAOX1

Aryl hydrocarbon receptor as a molecular mediator in the induction of human aldehyde oxidase gene expression.

Relatively little is known about receptor-mediated regulation of aldehyde oxidase (AOX). 2,3,7,8-Tetrachlorodibenzo-p-dioxin, an aryl hydrocarbon receptor (AHR) agonist, increases mouse Aox expression. However, given the species differences in ligand-dependent AHR activation and AOX regulation, it remains to be investigated whether human AHR regulates the expression of AOX1 (the human AOX isoform). Therefore, the present study was designed to determine the effect of modulating the functionality of human AHR on AOX1 gene expression in cell culture models (HepG2 and MCF-7) known to express AHR a

A glyoxal-specific aldehyde signaling axis in Pseudomonas aeruginosa that influences quorum sensing and infection.

The universally conserved α-oxoaldehydes glyoxal (GO) and methylglyoxal (MGO) are toxic metabolic byproducts whose accumulation can lead to cell death. In the absence of a known, natural inducer of the GO-specific response in prokaryotes, we exploited RNA-seq to define a GO response in the bacterial pathogen Pseudomonas aeruginosa. The highest upregulated operon consisted of the known glyoxalase (gloA2) and an antibiotic monooxygenase (ABM) domain of unknown function - renamed here Aldehyde responsive quorum-sensing Inhibitor (ArqI). The arqI-gloA2 operon is highly specific to GO induction and…

Aldehyde metabolism in Maotai-flavor Baijiu: insights from integrated metagenomic and metaproteomic analyses.

Acetaldehyde and acetal are crucial to the flavor of Maotai-flavor Baijiu, affecting aroma release and sauce-aroma traits. Targeted control of acetaldehyde metabolic flux is critical for stabilizing base liquor quality. However, the diversity of acetaldehyde-metabolizing enzymes and their microbial drivers remain uncharacterized, critically impeding precision control. This study selected the third production round to systematically investigate acetaldehyde accumulation dynamics and metabolic mechanisms. Time-resolved profiling showed that acetaldehyde and acetal concentrations display an initi

A General Direct Aldehyde C-H Alkylation via TBADT-Nickel Synergistic Catalysis.

Herein, we present a nickel/tetrabutylammonium decatungstate (TBADT)-catalyzed protocol for general C-H alkylation of aldehydes, enabling the efficient synthesis of aliphatic ketones through ligand-controlled cross-coupling. This mild and cost-effective methodology demonstrates broad substrate compatibility with various commercially available aldehydes and both activated and unactivated alkyl bromides, delivering target products in high yields. Notably, the practical utility of this catalytic system has been highlighted through a concise two-step synthesis of the commercially valuable musk odo…

Protective Effect of Indole-3-Aldehyde in Murine COVID-19-Associated Pulmonary Aspergillosis.

Aspergillus fumigatus is an environmental fungus recently included in the fungal high-priority pathogens by the World Health Organization. While immunodeficiency and/or pre-existing lung damage represent a well-recognized fertile ground for fungal growth, it is increasingly being recognized that severe viral infections may similarly favor A. fumigatus colonization and infection, as recently experienced in the Coronavirus disease 2019 (COVID-19) pandemic. Herein, in a murine model of COVID-19-associated pulmonary aspergillosis (CAPA), obtained by the concomitant exposure to the Severe Acute Res

The Intestinal Barrier Protective Effect of Indole Aldehyde Derivatives on Acute Toxoplasma gondii Infection.

Toxoplasmosis, a zoonotic infection caused by Toxoplasma gondii (T. gondii), poses a significant risk to human health and public safety. Despite the availability of clinical treatments, none effectively mitigate the intestinal barrier damage, which is the primary defense against T. gondii invasion. This study introduced aldehyde groups into the indole scaffold of a peptide-like structure to investigate the protective effects of these indole aldehyde derivatives on the intestinal barrier in mice with acute T. gondii infection. This approach leveraged the propensity of peptides and aldehyde grou

Aldehyde End-Capped Degradable Polyethylenes From Hydrogen-Controlled Ethylene/CO Copolymerization.

To access degradable polyolefin plastic, non-alternating copolymerization of ethylene (E) and carbon monoxide (CO) for producing polyethylene (PE) with in-chain ketones is particularly appealing; however, it still presents significant challenges such as molecular weight modulation (hydrogen response) and chain endgroup control (functional terminal). In this study, we achieved hydrogen-controlled E/CO non-alternating copolymerization using late transition metal catalysts. This process results in linear PEs containing the desired non-alternating in-chain keto groups (1.0-9.3 mol %) and with tuna

Gut Microbiota-Derived Tryptophan Metabolite Indole-3-aldehyde Ameliorates Aortic Dissection.

Tryptophan, an essential dietary amino acid, is metabolized into various metabolites within both gut microbiota and tissue cells. These metabolites have demonstrated potential associations with panvascular diseases. However, the specific relationship between tryptophan metabolism, particularly Indole-3-aldehyde (3-IAId), and the occurrence of aortic dissection (AD) remains unclear. 3-IAId showed an inverse association with advanced atherosclerosis, a risk factor for AD. In this study, we employed a well-established β-aminopropionitrile monofumarate (BAPN)-induced AD murine model to investigate

Characterization of N-Acetyl Cysteine Adducts with Exogenous and Neutrophil-Derived 2-Chlorofatty Aldehyde.

Hypochlorous acid is produced by leukocyte myeloperoxidase activity. 2-Chlorofatty aldehydes (2-ClFALDs) are formed when hypochlorous acid attacks the plasma membrane phospholipid plasmalogen molecular subclass and are thus produced following leukocyte activation as well as in the lungs of mice exposed to chlorine gas. The biological role of 2-ClFALD is largely unknown. Recently, we used an alkyne analog (2-ClHDyA) of the 2-ClFALD molecular species, 2-chlorohexadecanal (2-ClHDA), to identify proteins covalently modified by 2-ClHDyA in endothelial cells and epithelial cells. Here, we demonstrat…

Aldehyde oxidase 1 activity and protein expression in human, rabbit, and pig ocular tissues.

Aldehyde oxidase (AOX) is a cytosolic drug-metabolizing enzyme which has attracted increasing attention in drug development due to its high hepatic expression, broad substrate profile and species differences. In contrast, there is limited information on the presence and activity of AOX in extrahepatic tissues including ocular tissues. Because several ocular drugs are potential substrates for AOX, we performed a comprehensive analysis of the AOX1 expression and activity profile in seven ocular tissues from humans, rabbits, and pigs. AOX activities were determined using optimized assays for the

Highly Sensitive and Selective Detection of Formaldehyde via Bio-Electrocatalysis over Aldehyde Dehydrogenase.

Formaldehyde (HCHO), as one of the prominent indoor pollutants, causes many health-related problems. Although the detection of HCHO is a widespread concern and a variety of detection methods have been continuously developed, the volatile organic chemical (VOC) interference remains to be solved. Here, we report a highly sensitive and selective method for HCHO detection, relying on the selective electrochemical oxidation of formaldehyde catalyzed by aldehyde dehydrogenases (ALDHs) on a Cu electrode. The detection signal exhibits a standard power law relationship against the analytes with a broad

Intermolecular amide and aldehyde interactions: rotational spectroscopy of the complexes of formaldehyde with 2-azetidinone and formamide.

The binary intermolecular complexes of amides and formaldehyde can be taken as suitable models to investigate the non-covalent interactions of a peptide with the carbonyl group. We herein studied the rotational spectra of the model complexes of 2-azetidinone-H2CO and formamide-H2CO generated in a helium supersonic jet. For each complex, one rotational spectra featuring hyperfine structures caused by the 14N quadrupole coupling effect was observed and assigned to its global minimum conformation. The detected isomers of both studied complexes are stabilized by a dominant amide hydrogen bond N-H⋯

Synthesis of Benzoxaboroles by ortho-Oxalkylation of Arylboronic Acids with Aldehydes/Ketones in the Presence of Brønsted Acids.

Herein we describe a simple and efficient synthesis of benzoxaboroles from arylboronic acids and aldehydes or ketones in the presence of a Brønsted acid. This method greatly simplifies the starting materials and reduces the number of reaction steps. The reaction can also be accomplished with acetals and ketals. The reaction has a wide substrate scope and high practicability.

Genotoxicity of aldehyde mixtures: profile of exocyclic DNA-adducts as a biomarker of exposure to tobacco smoke.

Electrophilic compounds present in humans, originating from endogenous processes or pollutant exposures, pose a risk to health though their reaction with nucleophilic sites in protein and DNA. Among this chemical class, aldehydes are mainly present in indoor air and they can also be produced by endogenous lipid peroxidation arising from oxidative stress. Known to be very reactive, aldehydes have the ability to form exocyclic adducts to DNA that, for the most if not repaired correctly, are mutagenic and by consequence potential agents involved in carcinogenesis. The aim of this work was to esta…

CD36 involvement in the olfactory perception of oleic aldehyde, an odour-active volatile compound, in mice.

Cluster of differentiation 36 (CD36) is a broadly expressed transmembrane receptor that has multiple ligands. It has been found to occur abundantly on the surface of the olfactory epithelium in mice and postulated to play a role in mammalian olfaction. However, there have been no ethological analyses of the mammalian behaviour showing CD36 involvement in the olfactory perception of a distinct odour-active volatile compound. In this study, we aimed to assess whether mammals perceive oleic aldehyde, an odour-active volatile that serves as a potential CD36 ligand, and if so, whether CD36 is invol

Aldehyde dehydrogenase activity is a cancer stem cell marker of tongue squamous cell carcinoma.

Accumulating data reveal that aldehyde dehydrogenase (ALDH) activity is a cancer stem cell marker in several types of human cancers. Whether ALDH also defines cancer stem cells of tongue squamous cell carcinoma is unknown. In the present study, we performed the Aldefluor assay to detect ALDH enzymatic activity, and used flow cytometry to isolate ALDH+ and ALDH- cells based on their ALDH activity in the Tca8113 tongue squamous cell carcinoma cell line. We found that Tca8113 cells harbored 1.3% of ALDH+ cells, which exhibited higher proliferation capacity than their ALDH- counterparts and parent
