Detection of Delta9-tetrahydrocannabinol and amphetamine-type stimulants in oral fluid using the Rapid Stat point-of-collection drug-testing device.

The Rapid Stat assay, a point-of-collection drug-testing device for detection of amphetamines, cannabinoids, cocaine, opiates, methadone, and benzodiazepines in oral fluid, was evaluated for cannabis and amphetamine-type stimulants. The Rapid Stat tests (n = 134) were applied by police officers in routine traffic checks. Oral fluid and blood samples were analyzed using gas chromatography-mass spectrometry (GC-MS) for Delta(9)-tetrahydrocannabinol, amphetamine, methamphetamine, methylenedioxymethamphetamine, methylenedioxyethylamphetamine, and methylenedioxyamphetamine. The comparison of GC-MS

Stimulant use and increased severity of opioid use disorder and mental illness in patients with co-occurring disorders.

BACKGROUND: Among individuals with opioid use disorder (OUD) and co-occurring mental illness, stimulant co-use may be associated with more severe symptoms and less treatment utilization. We sought to understand the association of stimulant use with social needs and OUD and mental illness severity and treatment. METHODS: This cross-sectional study used baseline data from a randomized controlled trial of collaborative care for primary care patients with OUD and co-occurring depression and/or posttraumatic stress disorder (PTSD). We categorized stimulant use (cocaine/methamphetamine) into daily

Social Homeostasis and Psychoactive Drugs: What Can We Learn From Opioid and Amphetamine Drug Challenge Studies in Humans?

Social disequilibrium, or disrupted social homeostasis, underlies many behavioral disorders, including problematic drug use. One way to study the relationship between drug use and social homeostasis is to determine whether single doses of psychoactive drugs relieve some of the discomfort of social isolation and promote social connection. In this narrative review, we discuss challenges and opportunities in studying the relationship between psychoactive drugs and social homeostasis. Using the examples of opioids and amphetamines, we discuss the evidence that drugs alleviate dysphoria related to

Prescription amphetamines in people with opioid use disorder and co-occurring psychostimulant use disorder initiating buprenorphine: an analysis of treatment retention and overdose risk.

BACKGROUND: Attention-deficit and hyperactivity disorder (ADHD) is frequently diagnosed in patients with substance use disorders (SUDs), including opioids. There remains concern about the safety and efficacy of prescription amphetamines (PAs) and their impact on effectiveness of opioid use disorder (OUD) treatment with buprenorphine. OBJECTIVES: To assess the effect of PAs on OUD buprenorphine treatment retention and/or SUD-related emergency admission or drug-related poisonings. METHODS: We used a retrospective cohort design with a secondary analysis of data from Merative MarketScan Commerci

Hippocampal transcranial temporal interference stimulation reduced craving in methamphetamine use disorder.

BACKGROUND: Craving is a key challenge in treating methamphetamine use disorder (MUD). The memory reconsolidation theory indicates that interference with MUD after drug memory retrieval helps to reduce craving. Previous studies have demonstrated the importance of the hippocampus in memory consolidation, but traditional non-invasive interventions cannot effectively locate and intervene in the hippocampus. Although tTIS can accurately intervene in deep brain regions, it is still unknown whether tTIS intervention on the MUD hippocampus is effective in reducing craving. METHOD: A total of 40 male

Beyond addiction: exploring the factors behind suicidal thoughts among methamphetamine users in Guangdong, China.

BACKGROUND: Methamphetamine is one of the most abused drugs worldwide. Methamphetamine use disorders (MAUD) not only affect the physical and mental health of addicts but also increase the risk of suicide. Although the suicide rate of MAUD patients is high, there is little research on the clinical characteristics and risk factors of suicide risk in MAUD patients. So, it is necessary to investigate the related risk factors of suicidal ideation of MAUD. METHODS: Using response-driven sampling (RDS), a total of 10,225 MAUDs were randomly selected from April to May 2023 in Guangdong, China. MAUD w

A Case Report: Fatal Mesenteric Microvascular Compromise Following Methamphetamine use and Insulin Overdose Treated with Octreotide.

INTRODUCTION: Severe hypoglycemia from insulin overdose is a medical emergency that can be complicated by comorbidities and concurrent substance use. This case highlights a rare but fatal outcome of acute mesenteric ischemia in a patient with insulin overdose treated with Octreotide (Sandostatin, Novartis), compounded by recent methamphetamine use. The interplay of these factors may contribute to microvascular compromise and ischemia. CASE PRESENTATION: A 44-year-old male with chronic kidney disease stage 3b, body-mass-index of 23.23 kg/m2, insulin-dependent diabetes mellitus, and recent meth

Ketamine-Assisted Recovery (KARE): protocol for an open-label pilot trial of ketamine-assisted psychotherapy for publicly insured patients with methamphetamine use disorder and HIV risks.

INTRODUCTION: Methamphetamine use disorder (MeUD) is a debilitating condition with no FDA-approved pharmacotherapies that has been associated with poor neurological, psychiatric and cardiovascular outcomes, particularly among low-income populations. The use of methamphetamine also increases risks for sexually transmitted infections (STIs) by reducing behavioural inhibitions while enhancing sexual libido, disproportionately affecting sexual and gender minorities. The overlap of MeUD with HIV risks and psychological trauma underscores the need for innovative, accessible and culturally responsive

Substance use and spine density: a systematic review and meta-analysis of preclinical studies.

The elucidation of synaptic density changes provides valuable insights into the underlying brain mechanisms of substance use. In preclinical studies, synaptic density markers, like spine density, are altered by substances of abuse (e.g., alcohol, amphetamine, cannabis, cocaine, opioids, nicotine). These changes could be linked to phenomena including behavioral sensitization and drug self-administration in rodents. However, studies have produced heterogeneous results for spine density across substances and brain regions. Identifying patterns will inform translational studies given tools that no

Fatal α-PVP and amphetamine poisoning during a sauna and autoerotic practices.

We describe the sudden death of a middle-aged man while having a sauna under the influence of α-pyrrolidinovalerophenone (α-PVP) (PM blood concentration: 0.8 mg/L), amphetamine (0.34 mg/L), and other drugs (buprenorphine, benzodiazepines), and engaging in solitary sexual activities. The drugs' effects on the cardio-circulatory system and on body thermoregulation combined with the high temperatures are likely to have been central mechanisms leading to death. The high levels of adrenaline triggered by sexual arousal and the respiratory depression caused by buprenorphine, in association with benz

Drugs currently in Phase II clinical trials for cocaine addiction.

INTRODUCTION: There are no FDA-approved pharmacotherapies for treating cocaine addiction; thus, developing drugs to treat cocaine dependence is an unmet critical need. Fortunately, there are a number of drugs that are currently in Phase II clinical trial/s. This is due in part to the advances from in vivo imaging in humans which provided a roadmap of the neurochemistry of the cocaine-dependent brain. Most drugs currently in Phase II clinical trials attempt to modulate the disturbed neurochemistry in cocaine dependents to resemble those of healthy individuals. These predominantly modulate dopam

Self-reports of consumption of amphetamines, cocaine and heroin in a survey among marginalized drug users.

BACKGROUND AND AIM: Scientific literature offers few measurements of the quantities consumed by individual drug users. Such measurements are used for calculating the total drug consumption by the quantity-frequency method, and are extremely important for the comparison with waste water derived consumption estimates. The aim of this study was to measure quantities of amphetamines, cocaine and heroin consumed by marginalized drug users, using a multi-city questionnaire survey design. Variation by gender, age, frequency of use, main drug used and city was explored. RESULTS: The self-reported qua

Randomized, placebo-controlled trial of bupropion in methamphetamine-dependent participants with less than daily methamphetamine use.

AIMS: Two previous randomized trials found an effect for bupropion in reducing methamphetamine use in the subgroup with lower frequency of methamphetamine use at baseline. This study aimed to replicate these results by comparing bupropion versus placebo in methamphetamine-dependent participants with less than daily methamphetamine use at baseline. METHODS: Methamphetamine-dependent volunteers reporting methamphetamine use on ≤29 of past 30 days were randomized to bupropion 150 mg twice daily (n = 41) or placebo (n = 43) and out-patient counseling for 12 weeks. The primary outcome was the prop

Crystal methamphetamine use and HIV medical outcomes among HIV-infected men who have sex with men accessing support services in New York.

BACKGROUND: Drug use poses multiple challenges to maintaining physical health among HIV-infected individuals, particularly with regard to disease progression. Few studies, however, have examined the association between the use of crystal methamphetamine ("crystal meth") and HIV disease progression specifically among HIV-infected men who have sex with men (MSM). Understanding this relationship among HIV-infected MSM is particularly critical because of the high rates of crystal meth use reported in the population. METHODS: Associations between recent crystal meth use and poor HIV medical outcom

"High on my own supply": correlates of drug dealing among heterosexually identified methamphetamine users.

Although rates of methamphetamine use continue to increase throughout the United States, little is known about the individuals who sell methamphetamine at the street level. This exploratory study examined the prevalence and correlates of drug-dealing behavior in a sample of 404 heterosexually identified methamphetamine users who were participants in a sexual risk reduction intervention in San Diego, California. Twenty-nine percent of participants (N = 116) reported "dealing" methamphetamine in the past 2 months. In a multivariate logistic regression, methamphetamine dealing was associated with
