Modified repair of truncus arteriosus to maintain pulmonary artery architecture.

Neonatal surgical repair for truncus arteriosus historically involves removing the pulmonary arteries from the truncal root, closing the ventricular septal defect, and creating right ventricular to pulmonary artery continuity. Unfortunately, early reintervention is frequently required for conduit failure and proximal branch pulmonary artery stenosis. We present a technique that preserves the pulmonary artery architecture, keeping the pulmonary arteries in their native position. This technique has been applied to 16 patients and appears to decrease proximal branch pulmonary artery stenosis, the…

The bladder extracellular matrix. Part I: architecture, development and disease.

From the earliest studies with epithelial cells implanted into detrusor muscle to later experiments on smooth muscle in defined collagen gels, cell niche and extracellular matrix (ECM) have been clearly shown to orchestrate cellular behavior and fate whether quiescent, migratory, or proliferative. Normal matrix can revert transformed cells to quiescence, and damaged matrix can trigger malignancy or dedifferentiation. ECM influence in disease, development, healing and regeneration has been demonstrated in many other fields of study, but a thorough examination of the roles of ECM in bladder cell

Neuro-archaeology: pre-symptomatic architecture and signature of neurological disorders.

During brain development cells divide, differentiate and migrate to their assigned targets to form synapses and active cell assemblies. This sequence is controlled both by genetic programs and environmental factors. Alterations of this sequence by mutations or environmental insults leads to the formation of misconnected circuits endowed with a 'pre-symptomatic signature'. I propose here that early- and late-onset neurological disorders as diverse as infantile epilepsies, mental retardation, dyslexia or, in certain conditions, even Huntington's and Alzheimer's disease might be, in part, born at

Molecular architecture of the fruit fly's airway epithelial immune system.

BACKGROUND: Airway epithelial cells not only constitute a physical barrier, but also the first line of defence against airborne pathogens. At the same time, they are constantly exposed to reactive oxygen species. Therefore, airway epithelia cells have to possess a sophisticated innate immune system and a molecular armamentarium to detoxify reactive oxygen species. It has become apparent that deregulation of epithelial innate immunity is a major reason for the development of chronic inflammatory lung diseases. To elucidate the molecular architecture of the innate immune system of airway epithel…

Ablation of collagen IX and COMP disrupts epiphyseal cartilage architecture.

Chondrodysplasias are a genetically heterogeneous group of skeletal disorders. Mutations in genes coding for cartilage oligomeric matrix protein (COMP), collagen IX and matrilin-3 have been described to cause the autosomal dominantly inherited form of multiple epiphyseal dysplasia (MED). Even though there is clear evidence that these cartilage matrix proteins interact with each other, their exact functions in matrix organisation and bone development still need to be elucidated. We generated a mouse model lacking both collagen IX and COMP to study the potential complementary role of these prote

Are You MAD (Mammographic Architectural Distortion)? Introducing Subtypes of Synthetic Mammography/Tomosynthesis-Detected Architectural Distortion: Radiating Distortion and Nonradiating Tissue Alteration.

OBJECTIVE: The goals of this study are to introduce subtypes of mammographic architectural distortion (MAD), designated as radiating distortion (RD) and nonradiating tissue alteration (TA), and evaluate the clinical outcomes, interobserver agreement, and predictive capability of these newly defined subtypes. METHODS: Radiating distortion, TA, and 7 mammographic features of MAD were prospectively defined. Three breast radiologists reviewed consecutive screening synthetic mammography/tomosynthesis examinations recalled for MAD between January 1, 2021, and December 31, 2021. Reviewers reclassifi

Thin-Layer Sample-Ion-Selective Nanomembrane Architecture for Coulometric Cation Sensing.

Herein, we investigate a coulometric nanomembrane-based ISE integrated into a microfluidic cell pursuing the absolute analysis of potassium ions (K+), selected as a model example. The sensor consisted of a glass substrate coated with the conducting polymer poly(3-octylthiophene) (POT) and a nanometer-sized, potassium-selective membrane casted on top. In the microfluidic cell, the sample is confined to a thin-layer domain being sandwiched between the working electrode and the counter/reference electrode. Two electrochemical protocols are developed toward exhaustive ion transfer at the membrane-

PuWRKY46 drives root system architecture and enhances salt tolerance in poplar.

The developmental plasticity of lateral roots (LRs) is an essential adaptive strategy for optimizing root system architecture under salt stress. However, the molecular mechanisms by which salt stress modulates LR plasticity through cell wall remodeling remain largely elusive, particularly in woody species. This study investigates the abscisic acid (ABA)-responsive transcription factor PuWRKY46 as a critical regulator of LR developmental plasticity under salt stress in Populus ussuriensis. This study integrated phenotypic characterization of transgenic overexpression and RNA interference (RNAi)…

CTL1 (SLC44A1) regulates myelin lipid composition and architecture in Schwann cells.

Myelin formation requires extensive membrane lipid synthesis, yet how myelinating glia acquire the choline needed for this process remains i … all peripheral myelin production but contributes to maintaining normal myelin lipid composition and architecture, revealing an unexpected ability of Schwann cells to sustain myelination despite disruption of CTL1-dependent choline metabolism.

Disorder-specific and shared genetic architecture underlying schizophrenia and bipolar disorder.

Schizophrenia (SCZ) and bipolar disorder (BIP) share substantial genetic liability, yet exhibit heterogeneity in cognition, illness course and cardiometabolic health. Conventional disorder-level genome-wide association studies (GWASs) aggregate genetic liability obscuring this heterogeneity. We applied zero-phase component analysis whitening and common factor GWAS to SCZ and BIP summary statistics, deriving a shared component (PSYshared) and two mutually orthogonal, disorder-predominant components (SCZ~BIP and BIP~SCZ). We then examined whether the three profiles differed in locus architecture…

Hidden architecture of resistance: The extracellular matrix in melanoma's immune landscape.

Tumor heterogeneity in cutaneous melanoma (CM) is attributed to the functional and structural diversity of tumor cells, immune cells, and the tumor stroma, and determines patients' response to therapy. The tumor immune microenvironment (TIME) comprises a diverse array of cellular and non-cellular components. Cells include cancer-associated fibroblasts, T cells, NK cells, B cells, dendritic cells, myeloid-derived suppressor cells, and monocytes. Non-cellular components include cytokines, growth factors, and extracellular matrix (ECM). In this review, we specifically focus on the critical roles

Shared genetic architecture between ADHD and intelligence varies across ADHD subtypes.

BACKGROUND: Attention-deficit/hyperactivity disorder (ADHD) is a heterogeneous neurodevelopmental condition frequently accompanied by cognitive difficulties. Although previous genetic studies have demonstrated substantial overlap between ADHD and intelligence, most have treated ADHD as a single phenotype. However, whether this shared genetic architecture differs across ADHD subtypes remains unclear. METHODS: We conducted a genome-wide cross-trait analysis integrating large-scale genome-wide association study (GWAS) datasets of overall ADHD, its subtypes-childhood ADHD, persistent ADHD, and la…
