[First report of a case of fungal keratitis due to Curvularia hominis in Spain].

BACKGROUND: Dematiaceous fungal genus Curvularia is a causal agent of keratitis, onychomycosis, and skin infections. In 2014, using DNA sequencing techniques, five new species, including Curvularia hominis, were described. In this article, a report is presented on the first clinical case of C.hominis infection in Spain. It concerns a corneal ulcer caused by this recently described species. CASE REPORT: A 46 year-old male patient with a corneal ulcer in his left eye went to the Emergency Department. Specimens were obtained from the lesion, and the patient was admitted due to the risk of cornea

Inhibition of Haemonchus contortus larval development by fungal lectins.

BACKGROUND: Lectins are carbohydrate-binding proteins that are involved in fundamental intra- and extracellular biological processes. They occur ubiquitously in nature and are especially abundant in plants and fungi. It has been well established that certain higher fungi produce lectins in their fruiting bodies and/or sclerotia as a part of their natural resistance against free-living fungivorous nematodes and other pests. Despite relatively high diversity of the glycan structures in nature, many of the glycans targeted by fungal lectins are conserved among organisms of the same taxon and some

4-Dechloro-14-deoxy-oxacyclododecindione and 14-deoxy-oxacylododecindione, two inhibitors of inducible connective tissue growth factor expression from the imperfect fungus Exserohilum rostratum.

Connective tissue growth factor (CTGF/CCN2), a member of the CCN superfamily of secreted cysteine-rich glycoproteins, is a central mediator of tissue remodeling and fibrosis. CTGF is suggested to be an important down-stream effector of transforming growth factor-beta (TGF-β) signaling and has therefore reached considerable pathophysiological relevance because of its involvement in the pathogenesis of fibrotic diseases, atherosclerosis, skin scarring, and other conditions with excess production of connective tissue. In a search for inhibitors of inducible CTGF expression from fungi, two new mac

Sensitivity of Podosphaera xanthii populations to anti-powdery-mildew fungicides in Spain.

BACKGROUND: Cucurbit powdery mildew caused by Podosphaera xanthii limits crop production in Spain, where disease control is largely dependent on fungicides. In previous studies, high levels of resistance to QoI and DMI fungicides were documented in south-central Spain. The aim of this study was to investigate the sensitivity of P. xanthii populations to other fungicides and to provide tools for improved disease management. RESULTS: Using a leaf-disc assay, sensitivity to thiophanate-methyl, bupirimate and quinoxyfen of 50 isolates of P. xanthii was analysed to determine discriminatory concent

In vitro photodynamic therapy against Foncecaea pedrosoi and Cladophialophora carrionii.

Photodynamic therapy (PDT) has been originally developed for cancer treatment, but recently, it has been successfully employed against microorganisms, including fungi. Chromoblastomycosis is a subcutaneous fungal infection that is recalcitrant to conventional antifungal drug therapy. The most frequent species involved are Foncecaea pedrosoi and Cladophialophora carrionii. The present study aimed to verify the efficacy in vitro of PDT employing methylene blue (MB) as a photosensitiser and Light emmiting diode (LED) (InGaAl) as the light source. Methylene blue at the concentrations of 16, 32 and

Chemical structure of beta-galactofuranose-containing polysaccharide and O-linked oligosaccharides obtained from the cell wall of pathogenic dematiaceous fungus Fonsecaea pedrosoi.

Fonsecaea pedrosoi is the main etiologic agent of chromoblastomycosis, usually occurring in tropical and subtropical areas. The cell wall components of pathogenic microorganisms behave as an antigen and/or ligand of the innate immune response. The cells of F. pedrosoi reacted with the α-galactopyranose-binding lectin (Griffonia simplicifolia lectin 1B4 isolectin, GSL 1B4), as well as the α-mannose-binding lectin, concanavalin A. The cell wall glycoprotein was isolated from conidial cells of F. pedrosoi, and its structure was analyzed by (1)H-nuclear magnetic resonance (NMR) and (13)C-NMR exper

Temperature-mediated patterns of local adaptation in a natural plant-pathogen metapopulation.

There have been numerous investigations of parasite local adaptation, a phenomenon important from the perspectives of both basic and applied evolutionary ecology. Recent work has demonstrated that temperature has striking effects on parasite performance by mediating trade-offs in parasite life history and through genotype x environment interactions. To test whether parasite local adaptation is mediated by temperature, I measured the performance of sympatric populations against allopatric populations of a fungal pathogen, Podosphaera plantaginis, on its host Plantago lanceolata, across a temper

Chromoblastomycosis: an overview of clinical manifestations, diagnosis and treatment.

Chromoblastomycosis is one of the most frequent infections caused by melanized fungi. It is a subcutaneous fungal infection, usually an occupational related disease, mainly affecting individuals in tropical and temperate regions. Although several species are etiologic agents, Fonsecaea pedrosoi and Cladophialophora carrionii are prevalent in the endemic areas. Chromoblastomycosis lesions are polymorphic and must be differentiated from those associated with many clinical conditions. Diagnosis is confirmed by the observation of muriform cells in tissue and the isolation and the identification of

The major chromoblastomycosis fungal pathogen, Fonsecaea pedrosoi, extracellularly releases proteolytic enzymes whose expression is modulated by culture medium composition: implications on the fungal development and cleavage of key's host structures.

We investigated the possible secretion of peptidases by F. pedrosoi, when conidial cells were cultured in two distinct media. Aspartyl proteolytic activity was detected on the Czapeck-Dox-derived supernatant, which was blocked by pepstatin, and only active in extremely acidic conditions. The supernatant obtained after conidia growth in Kauffman medium presented metallopeptidase activity, which was active over a broad pH range and sensitive to 1,10-phenanthroline and EGTA. Additionally, both culture supernatants were able to cleave a wide range of proteinaceous substrates, including important h

Morphological evidence for keratinolytic activity of Scopulariopsis spp. isolates from nail lesions and the air.

The morphological expression of human hair and nail invasion in vitro by 31 isolates of nine Scopulariopsis species was studied by light microscopy on whole material and on semi-thin sections, as well as by transmission electron microscopy. Only some isolates of Scopulariopsis brumptii, S. candida, S. carbonaria and S. koningii were keratinolytically active. They came either from nail lesions or from outdoor aerosols. The most active isolate belonged to S. koningii and was recovered from a fingernail lesion. Both hair and nail degradation followed the biochemical and morphogenetic model descri

[A case of chromomycosis arising in a Japanese-Brazilian patient].

A 41-year-old Japanese-Brazilian man, living in Japan since 1991, visited our hospital in August 1998 complaining of a scaly annular erythema which had been present on his right forearm since 1996. Granulomatous inflammation was revealed in the dermis upon skin biopsy. Sclerotic cells were present within the granulomatous lesions. Fonsecaea pedrosoi was isolated from tissue cultures. The restriction fragment length polymorphism pattern of mitochondrial DNA of the causative fungus was compatible with F. pedrosoi type 1, the commonest type in Japan. Since type 4 is usually found in South America

X-ray microanalysis of black piedra.

The elements present in the fungal structures produced by Piedraia hortae in vivo and in vitro have been investigated using electron microscopy X-ray microanalysis. Phosphorus, sulphur and calcium were detected in the nodules which developed on hair and on colonies on culture. These elements belong to the extracellular material that compacts the pseudoparenchymatous organization of the fungus. They may be present due to the capacity of melanin-like pigments to sequester ions and/or they may form part of the sulphates and phosphates of the polyanionic mucopolysaccharides that constitute the ext
