Changes in T cell effector functions over an 8-year period with TNF antagonists in patients with chronic inflammatory rheumatic diseases.

The aim of the study was to clarify the effect of long-term anti-TNF therapy on T cell function in patients with rheumatologic immune-mediated inflammatory diseases (IMID). The production of IFNγ by T cells was evaluated at baseline and after 1, 2, 4, and 8 years of anti-TNF agents by means of a QuantiFERON-TB Gold In-Tube assay. The T cell proliferation and surface co-expression of CD25/CD134 in response to phytohaemagglutinin together with the in vitro impact of anti-TNF therapy on the functional capacity of T cells were evaluated after 8 years from the onset of the biological treatment. Age

Overview and recent developments in the medical management of paediatric uveitis.

INTRODUCTION: Although rarer than its adult counterpart, non-infectious uveitis remains a significant cause of ocular morbidity in children. Owing to the chronicity of the disorder and when refractory to first-line treatment, namely corticosteroids, systemic immunosuppressive treatment may be required to control the disease. AREAS COVERED: Following a literature search using the keywords 'paediatric uveitis', 'juvenile idiopathic arthritis-associated uveitis', 'immunosuppression' and 'treatment', we reviewed the range and effectiveness of treatments employed in the management of non-infectiou

Control of blood-transmitted infections in dentistry.

As knowledge has accumulated on the blood-transmitted pathogenic agents, the contact with biological fluids (blood, plasma, saliva, etc.) from apparently healthy individuals has started to be regarded as a real professional risk for dentists. Theoretically, exposure to a contaminated biological specimen may have as a consequence transmission of infection from patient to dentist, from dentist to patient and from patient to patient via inadequately decontaminated and sterilized dental equipment. The present study is concerned with the analysis of the specific conditions that favor the occurrence

Antimicrobial action of chelating agents: repercussions on the microorganism development, virulence and pathogenesis.

Infections caused by resistant microorganisms often fail to respond to conventional therapy, resulting in prolonged illness, increased treatment costs and greater risk of death. Consequently, the development of novel antimicrobial drugs is becoming more demanding every day since the existing drugs either have too many side-effects or they tend to lose effectiveness due to the selection of resistant strains. In view of these facts, a number of new strategies to obstruct vital biological processes of a microbial cell have emerged; one of these is focused on the use of metal-chelating agents, whi

Risk of alanine aminotransferase flare in patients with previous hepatitis B virus exposure on biological modifier therapies-A population-based study.

BACKGROUND: It is uncertain whether biological therapies would increase the risk of hepatitis among patients with past hepatitis B virus (HBV) infection. This study aimed to evaluate the risk of alanine aminotransferase (ALT) flare in patients with past HBV infection while using biological therapies. METHODOLOGY: Patients who received biological therapies for ≥3 months from 2000 to 2019 were identified from a population-based database in Hong Kong. Patients with past HBV infection were compared with a control group without prior HBV exposure. The primary endpoint was development of ALT flare

Hormonal control of the neuropeptide Y system.

Neuropeptide Y (NPY) and the related receptors represent a widely diffused system that is involved in the regulation of multiple biological functions. NPY, a 36-aminoacid peptide expressed in several areas of the nervous system, is a pleiotropic factor participating to the control of some physiological processes, such as cognitive functions, eating behavior, circadian rhythms, neuroendocrine mechanisms, reproductive and cardiovascular functions. NPY acts through a series of G-protein-associated membrane receptors (NPY-Rs), characterized by different tissue distribution and affinity for the lig

Deciphering Molecular Determinants Underlying Penicillium digitatum's Response to Biological and Chemical Antifungal Agents by Tandem Mass Tag (TMT)-Based High-Resolution LC-MS/MS.

Penicillium digitatum is a widespread pathogen responsible for the postharvest decay of citrus, one of the most economically important crops worldwide. Currently, chemical fungicides are still the main strategy to control the green mould disease caused by the fungus. However, the increasing selection and proliferation of fungicide-resistant strains require more efforts to explore new alternatives acting via new or unexplored mechanisms for postharvest disease management. To date, several non-chemical compounds have been investigated for the control of fungal pathogens. In this scenario, unders

Chitosan and nematophagous fungi for sustainable management of nematode pests.

Plants are exposed to large number of threats caused by herbivores and pathogens which cause important losses on crops. Plant pathogens such as nematodes can cause severe damage and losses in food security crops worldwide. Chemical pesticides were extendedly used for nematode management. However, due to their adverse effects on human health and the environment, they are now facing strong limitations by regulatory organisations such as EFSA (European Food Safety Authority). Therefore, there is an urgent need for alternative and efficient control measures, such as biological control agents or bi…

Targeted Biologic Therapies for Hidradenitis Suppurativa.

Chronic inflammatory disorders of the apocrine gland (CIDAP), such as hidradenitis suppurativa (HS), are characterized by painful, recurrent lesions in apocrine gland-rich areas. First-line treatments-including retinoids and antibiotics-often fail to prevent recurrence and biofilm formation, necessitating the use of targeted biologic therapies. This review evaluated U.S.-based randomized controlled trials and cohort studies published between 2014 and 2024 on the efficacy of such therapies in adult HS patients. A total of 13 studies met inclusion criteria. Agents targeting interleukins (IL-17A,

Micro-/nanorobots in nanomedicine - Guidance, imaging and the integration of AI and robotics.

The integration of robotics and artificial intelligence (AI) into nanomedicine represents a significant advancement in developing targeted therapeutic and diagnostic platforms. This field focuses on engineering micro- and nanoscale agents, such as magnetic nanoparticles (MNPs), microbots, and nanobots, for tasks like targeted therapies, sensing, and manipulation at diseased sites. MNPs are typically composed of iron oxides and serve as foundational components due to their biocompatibility, tunable surface chemistry, and responsiveness to external magnetic fields. They are used in targeted drug

Citywide pharmaceutical preparation for bioterrorism.

One community's efforts to become pharmaceutically prepared for an attack with biological agents is described. In response to recent bioterrorist activities, including a local scare in 1999 involving anthrax, the pharmacy department at Deaconess Medical Center in Spokane, Washington, was asked to develop a plan for bioterrorism preparedness. A literature search was conducted, and resources, such as the Centers for Disease Control and Prevention, were contacted. For each biological agent, information was compiled about symptom onset, treatment, postexposure prophylaxis, patient isolation precau

[Indications of anakinra].

The efficacy of anakinra on both RA-related symptoms and structural damage has been demonstrated in several randomized controlled trials. However, its interest seems limited with regards to other biologic agents. Anakinra seems promising in the treatment of childhood or adult onset Still disease, after the failure of both high dose steroids and methotrexate. The efficacy of anakinra is dramatic in several hereditary auto-inflammatory syndromes. Anakinra could be an interesting drug for the treatment of neutrophilic dermatosis or relapsing chondritis, refractory to conventional agents. Injectio

Rickettsia conorii isolated from Rhipicephalus sanguineus introduced into Switzerland on a pet dog.

A tick/rickettsial survey in a household near Geneva, Switzerland, revealed that 30 (40%) of 75 nymphs and adults of the brown dog tick, Rhipicephalus sanguineus, were infected with a rickettsial agent biologically and antigenically indistinguishable from R. conorii, the causative agent of boutonneuse fever. Introduced in 1979 from either southern France or Italy by the family's pet dog, the tick infestation had steadily increased until 1981 when control measures were initiated. During 1980 and 1981, four persons associated with the household's pet dog contracted a febrile illness diagnosed as

Pharmacological Management of Chronic Rhinosinusitis: Current and Evolving Treatments.

Chronic rhinosinusitis (CRS) is an inflammatory sinonasal condition with multiple etiologic factors that is associated with a vast economic cost. Treatment is most frequently pharmacologic and has centered on agents that ameliorate inflammation, decrease bacterial or pathogen load, and facilitate egress of mucus or purulence from the sinonasal cavity. Nasal saline irrigations, topical nasal steroids, certain antibiotics, and systemic steroids have shown some efficacy in the management of CRS. Recently, biologic therapeutics that target specific inflammatory pathways associated with subsets of

[CME: Mepolizumab, an Additional Therapeutic Agent for Severe Asthma].

CME: Mepolizumab, an Additional Therapeutic Agent for Severe Asthma Abstract. The challenging therapy of severe uncontrollable bronchial asthma aims primarily at sufficient symptom control and the minimization of the exacerbation rate. If, despite extended drug therapy, symptom control is very difficult to achieve, biologics should preferably be used instead of corticosteroid therapy. The aim of this article is to point out these different asthma therapy pathways and to describe diagnostic criteria as well as practical experiences through application in patients.

Detection of degradation products of chemical warfare agents in Halabja soil: In vitro genotoxicity and effects in exposed humans.

The town of Halabja in Iraqi Kurdistan remains a symbol of the devastating impact of chemical weapons. Almost four decades after the attack, inhabitants continue to suffer from increased cancer rates and severe respiratory, neurological, and psychiatric disorders. While the acute effects of chemical warfare agents have been widely studied, their long-term environmental persistence and associated health impacts remain poorly understood. Here, we investigated the presence of chemical warfare agent residues in the environment and assessed their potential genotoxic effects. Soil and water samples

[Therapeutic Strategies for Idiopathic Inflammatory Myopathies].

Almost all types of idiopathic inflammatory myopathy (IIM) are treated by immune-modulating therapies. Corticosteroids, such as prednisolone and methylprednisolone, are used as first-line therapy for IIM. When the symptoms cannot be sufficiently improved, immunosuppressive agents, such as azathioprine, methotrexate, or tacrolimus, should be administered approximately two weeks after starting corticosteroid therapy. Moreover, intravenous immunoglobulin is recommended for severe cases at the same time as starting immunosuppressive agents. If these therapies do not improve symptoms, biologics, su
