Macrophages of the splenic marginal zone are essential for trapping of blood-borne particulate antigen but dispensable for induction of specific T cell responses.

Rapid removal of pathogens from the circulation by secondary lymphoid organs is prerequisite for successful control of infection. Blood-borne Ags are trapped mainly in the splenic marginal zone. To identify the cell populations responsible for Ag trapping in the marginal zone, mice were selectively depleted of marginal zone macrophages and marginal metallophilic macrophages. In the absence of these cells, trapping of microspheres and Listeria monocytogenes organisms was lost, and early control of infection was impaired. Depletion of marginal zone macrophages and marginal metallophilic macropha

The 'teabag method': tick feeding protocol and the effects of tick feeding on hematological parameters in the canine host.

BACKGROUND: Hard tick infestation occurs naturally in humans, domestic animals, and livestock species. Upon feeding, ticks transmit a wide variety of pathogens that may result in serious diseases with severe public health and economic impacts. While tick-borne diseases significantly impact human and animal health and agricultural production worldwide, as ectoparasites, ticks can also cause serious tissue injury, tick paralysis, or exsanguination from mass infestation. Experimental tick feeding is necessary to study tick-borne diseases and effectively test novel vaccines and therapeutics. Such

First molecular evidence of vertical transmission of Ehrlichia canis in naturally infected female dogs in Brazil.

Ehrlichia canis is an obligate intracellular bacterium transmitted by the tick Rhipicephalus sanguineus sensu lato. This pathogen is also the etiological agent of canine monocytic ehrlichiosis, a disease with widespread global distribution and recognized zoonotic potential. Although vector-borne transmission is well established, evidence supporting the occurrence of vertical transmission remains limited. This study aimed to investigate the potential transplacental transmission of E. canis in 51 naturally infected pregnant bitches using molecular diagnostic techniques, genetic sequencing, and p

Molecular identification and characterization of Rickettsia spp. and other tick-borne pathogens in cattle and their ticks from Huambo, Angola.

Ticks are one of the most common vectors of a broad variety of pathogenic agents that significantly affects cattle production causing reduced productivity and important economic losses, while simultaneously having an impact on human health due to the zoonotic risk. In much of the territory of Angola urban population has grown rapidly in recent decades, sharing today close contact with large farms that are generally owned by city residents, providing the ideal conditions for vector-borne pathogens (VBP) transmission between animals and humans. Here we studied the occurrence of Rickettsia, Anapl

Attributable testing for abnormal prion protein, database linkage, and blood-borne vCJD risks.

CONTEXT: National prospective collection of tonsillar tissue to be tested anonymously for abnormal lymphoreticular accumulation of prion protein (PrP) was approved to begin in the UK in 2004. The UK is not, however, testing autopsy specimens attributably for abnormal PrP (PrP(SC)) so that recipients at risk after a blood transfusion from, or exposed to surgical instruments from, a deceased carrier of variant Creutzfeldt-Jakob disease (vCJD) can be followed up to quantify transmission risks. In Switzerland, surveillance for subclinical vCJD includes unconsented testing in autopsies: consented t

[Assessment of biological risk of blood transmission among health workers at a Bari university hospital].

We have examined all occupational exposures to biological materials, occurred from September 2001 to December 2002, among health workers of Bari polyclinic. The reported exposures, through an appropriate questionnaire expressly predisposed, have been a total of 114. The nurses were the most exposed category (46.5%) followed by physicians (27.2%). The highest load of accidents happened in surgical area (53%) and at a greater extent after the first three hours of the work shift (60%). The instruments that have caused endermic lesions have been more frequently the hollow bone needles (53.2%). The

Infectious contamination of healthcare workers.

BACKGROUND: Infectious diseases represent a major component of biological hazards encountered by healthcare workers. Occupational exposure may occur through multiple transmission pathways including airborne particles, direct contact, contaminated environments, or blood exposure accidents. Understanding these mechanisms is essential to guide effective prevention strategies in healthcare settings. METHODS: This narrative review synthesises current epidemiological data and preventive recommendations regarding occupational infectious risks in healthcare and related professional environments. The effect of riboflavin and ultraviolet light on the infectivity of arboviruses.

BACKGROUND: Arboviruses are an emerging threat to transfusion safety and rates of infection are likely to increase with the increased rainfall associated with climate change. Arboviral infections are common in Australia, where Ross River virus (RRV), Barmah Forest virus (BFV), and Murray Valley encephalitis virus (MVEV), among others, have the potential to cause disease in humans. The use of pathogen reduction technology (PRT) may be an alternative approach for blood services to manage the risk of arboviral transfusion transmission. In this study, the effectiveness of the Mirasol PRT (Terumo B

Tick Saliva Collection.

Tick saliva contains a complex mixture of biologically active compounds that enable prolonged blood feeding by modulating host hemostasis, inflammation, and immune responses. In addition, saliva serves as a key interface for the transmission of tick-borne pathogens, with specific salivary molecules facilitating pathogen survival and host infection. Tick saliva may also contain glycosylated proteins bearing galactose-α-1,3-galactose (α-Gal), which have been implicated in the development of α-Gal allergy in humans, as well as neurotoxic components responsible for tick paralysis. Given the centra

Lipid abnormalities in a healthcare worker receiving HIV prophylaxis.

Healthcare workers (HCWs) worldwide risk occupational exposure to HIV and other blood-borne pathogens. Post-exposure prophylaxis (PEP) may decrease the risk of seroconversion after occupational or sexual exposure. Current guidelines recommend immediate PEP with at least 2 drugs following HIV exposure. In high-risk exposures, the guidelines recommend the use of a protease inhibitor (PI) as well as 2 reverse transcriptase inhibitors. Protease inhibitors have been associated with dyslipidaemias, other metabolic abnormalities and lipodystrophy syndromes in AIDS patients. We report a case of new tr…

Role of enterococcus in intraabdominal sepsis.

Although enterococcus is well recognized as a pathogen in endocarditis, urinary tract infections, and biliary sepsis, its role in other forms of intraabdominal sepsis remains controversial. Antibiotics that lack activity against enterococcus can often be employed successfully in intraabdominal infections, even when enterococci are present as part of the polymicrobial flora. Furthermore, the enterococcus rarely emerges as a blood borne pathogen in such a setting. Breakthrough enterococcal septicemia may occur, however, in the immunodepressed host, particularly in the face of broad-spectrum anti

Demonstration of transplacental transmission of a human isolate of Anaplasma phagocytophilum in an experimentally infected sheep.

Anaplasma phagocytophilum, first identified as a pathogen of sheep in Europe, more recently has been recognized as an emerging tick-borne pathogen of humans in the U.S. and Europe. Transmission of A. phagocytophilum is reported to be by ticks, primarily of the genus Ixodes. While mechanical and transplacental transmission of the type genus organism, A. marginale, occur in addition to tick transmission, these modes of transmission have not been considered for A. phagocytophilum. Recently, we developed a sheep model for studying host-tick-pathogen interactions of the human NY-18 A. phagocytophil

Wild boar (Sus scrofa) - reservoir host of Toxoplasma gondii, Neospora caninum and Anaplasma phagocytophilum in Slovakia.

In Central Europe the wild boar population is permanently growing and consequently Cf foodborne infections. In this study serological and molecular detection of Toxoplasma gondii and Neospora caninum in wild boars was evaluated. Moreover, same samples were screened for the presence and genetic variability of tick-borne bacterium Anaplasma phagocytophilum. Blood samples collected from 113 wild boars from Southern Slovakia were examined for antibodies to T. gondii by indirect and to N. caninum by competitive ELISA. The presence of parasitic DNA in blood samples was determined by standard or real

Response rate of Malaysian blood donors with reactive screening test to transfusion medicine unit calls.

Blood donors with reactive screening test results are requested to come in for counseling by letter and telephone call. It has been noticed many donors responded to neither the letters nor the telephone calls. We evaluated 589 cases with reactive screening test results (208 positive for hepatitis C, 209 for hepatitis B, 85 for VDRL and 87 for HIV). In the hepatitis C positive group 61 donors (29.3%) did not respond and 4.7% missed their follow-up appointment. Similarly low response rates were noted with the HBV (58.9%) and VDRL (67.1%) positive groups. Among HIV positive donors 46.0% failed to

Tattoos and transfusion-transmitted disease risk: implications for the screening of blood donors in Brazil.

Having a tattoo has been associated with serological evidence of hepatitis B and C viruses, as well as human immunodeficiency virus infections and syphilis; all of these are known to be transmissible by blood transfusion. These associations are of higher magnitude for individuals with nonprofessionally-applied tattoos and with two or more tattoos. Tattoos are common among drug addicts and prisoners, conditions that are also associated with transfusion-transmitted diseases. We examined the implications of these associations for the screening of blood donors in Brazil. Numbers of individuals who

Nestling sex and behaviour determine the host preference of insect vectors in avian nests.

Sexual differences in pathogen prevalence in wildlife often arise from varying susceptibility influenced by factors such as sex hormones and exposure to pathogens. In the case of vector-borne pathogens, host selection by insect vectors determines the exposure of hosts to infections, largely affecting the transmission of these infectious diseases. We identify the blood-feeding patterns of insect vectors in Blue Tit (Cyanistes caeruleus) nestlings in a 3-year study. Blood from both nestlings and insect vectors (Culicoides spp. and Simuliidae) captured inside nest-boxes were used to molecularly d
