Increased resting metabolism in neurofibromatosis type 1.

BACKGROUND & AIMS: Neurofibromatosis type 1 (NF1) is an autosomal dominant genetic disease that is characterized by neurocutaneous changes with multisystem involvement. A previous study with adults with NF1 revealed that changes in total energy expenditure were related to food consumption and body composition. Resting energy expenditure (REE), a measure of energy that the body expends to maintain vital functions, has not been assessed in NF1 populations. This study aimed to assess REE in individuals with NF1 using indirect calorimetry (IC) and evaluate its correlation with body composition and

Breast Tissue Composition-Why It Matters and How Can We Measure It More Accurately in Epidemiology Studies.

Early-life body size has been consistently associated with breast cancer risk. The direction of the association changes over time, with high birth weight, smaller adolescent body size, and adult weight gain all increasing breast cancer risk. There is also a clear positive association between larger body size and increased breast adipose tissue measured by mammograms, but less is known about how body size changes across life stages affect stromal and epithelial breast tissue. Using breast tissue slides from women with benign breast disease, Oh and colleagues applied machine learning methods to

Systolic blood pressure and albuminuria reduction mediate cardiovascular benefits of finerenone in T2 diabetes and CKD.

BACKGROUND AND HYPOTHESIS: Finerenone reduces cardiovascular events in patients with type 2 diabetes (T2D) and chronic kidney disease (CKD). The cardiovascular benefit emerges within months, before measurable kidney protection, but the early pathways carrying it are not comprehensively quantified. We hypothesized that early reductions in urine albumin-to-creatinine ratio (UACR) and systolic blood pressure (SBP) jointly mediate a substantial proportion of finerenone's long-term cardiovascular benefit, whereas changes in body weight and serum potassium do not. METHODS: We performed causal media

Weight Loss without Food Intake Suppression through Size-Dependent Retention of Anti-Inflammatory Nanomedicines.

Obesity is a risk factor for high-mortality health conditions, including cardiovascular diseases and type 2 diabetes, which makes the advancement of efficacious and safe weight loss therapies a high priority in pharmacology. The causal link between obesity and its comorbid conditions is believed to be a chronic state of inflammation originating within adipose tissue, with macrophages playing central roles, an axis that is not targeted directly by current therapies. Here, we use nanocarriers to deliver an anti-inflammatory glucocorticoid receptor agonist to adipose tissue macrophages and report

Body mass index levels and changes before and after dementia diagnosis and risk of all-cause mortality: a nationwide cohort study.

BACKGROUND: Dementia is a progressive disease associated with various health risks, including increased mortality. Although weight changes have been linked to adverse outcomes in older adults, limited research has examined the impact of body weight status after dementia diagnosis and its changes before and after diagnosis on mortality. This study aimed to address these gaps. METHODS: Using nationwide data from the Korean National Health Insurance Service, we included 37,717 individuals newly diagnosed with dementia between 2010 and 2016, comprising 29,982 with Alzheimer’s disease (AD) and 3,22

Sacha inchi (Plukenetia volubilis) seed oil alleviates Plasmodium berghei infection in mice by reducing proinflammatory cytokine levels and inhibiting parasite growth.

BACKGROUND: Sacha inchi (Plukenetia volubilis) seed is a rich source of polyunsaturated fatty acids, namely omega-3 and omega-6, which provide various health benefits, especially in the prevention of inflammatory diseases. Malaria, caused by Plasmodium species, triggers inflammasome activation, leading to the release of proinflammatory cytokines. AIM: This study aimed to reveal the immunomodulatory potential effect of Sacha inchi seed oil (SISO) on parasite growth and proinflammatory cytokine expression (Interleukin-6, Il-8, and Il-12) in Plasmodium berghei-infected mice. METHODS: Twenty-fiv

A very-low-calorie ketogenic diet normalises obesity-related enhanced levels of erythropoietin compared with a low-calorie diet or bariatric surgery.

PURPOSE: Nutritional ketosis synergistically with body-weight loss induced by a very-low-calorie ketogenic diet (VLCKD) has proven to be effective in improving obesity-related pathophysiology. Recently, growing attention has been focused on the relation between erythropoietin (EPO) and obesity. Thus, this study aims to investigate whether nutritional ketosis and weight loss induced by a VLCKD modify the circulating levels of EPO in patients with obesity in comparison with the effect of low-calorie diet (LCD) or bariatric surgery (BS). METHODS: EPO levels, iron status and body composition para

EMG activity during positive-pressure treadmill running.

Success has been demonstrated in rehabilitation from certain injuries while using positive-pressure treadmills. However, certain injuries progress even with the lighter vertical loads. Our purpose was to investigate changes in muscle activation for various lower limb muscles while running on a positive-pressure treadmill at different amounts of body weight support. We hypothesized that some muscles would show decreases in activation with greater body weight support while others would not. Eleven collegiate distance runners were recruited. EMG amplitude was measured over 12 lower limb muscles.

Real-time prediction of rapid weight change in children with cystic fibrosis who have initiated modulator therapy.

BACKGROUND & AIMS: Broad uptake of elexacaftor/tezacaftor/ivacaftor (ETI) modulator therapy has substantially improved nutritional status among children with cystic fibrosis (CF), but post-ETI weight change may be suboptimal without personalized monitoring of body composition and physical function markers. We developed a prediction model including these markers and evaluated clinical thresholds assessing individualized risk of rapid weight change over time. METHODS: A retrospective longitudinal cohort study was performed (n = 27 children with CF aged 6-11 years starting ETI) to predict age-re

The Predictive Accuracy of Methods Commonly Used for Evaluating Animal Distress.

Accurate distress assessment is essential for ensuring animal welfare and scientific validity. Although behavioral and physiological indicators are commonly used, their predictive value across disease models and severity grades remains insufficiently defined. Here, we evaluated the discriminatory performance of body weight changes, distress scoring, burrowing, and nesting across ten datasets involving transmitter implantation, bile duct ligation (BDL), and chronic pancreatitis (CP) in mice. Receiver operating characteristic analyses determined the area under the curve (AUC), and optimal cut-of…

Safety of white adipose tissue activator cyclophilin A intracellular peptide (Pep19™) in healthy dogs.

Oral consumption of Pep19™, a 10 amino acid intracellular peptide, has been associated with weight loss in rodents and humans via induction of nonshivering thermogenesis. This study aimed to test its safety and tolerance in dogs. Eight healthy adult neutered university owned beagles (4 female and 4 male) were individually housed in runs and fed an extruded kibble in a quantity historically associated with weight stability. They were administered Pep19™ (5 mg/dog/day, 0.32 - 0.49 mg/kg/day) as a once daily oral dose for 28 days. Health screening, including physical examination, body weight, bod

The Influence of the Sympathetic Nervous System on Cardiometabolic Health in Response to Weight Gain or Weight Loss.

Alterations in sympathetic nervous activity are evident in response to changes in body weight. Sympathetic nervous activity and sympathetic responses to weight change are regionalized, with alterations in end organ function dependent on the changes occurring in the brain regulatory pathways invoked and in the effector organs engaged. The obesity-induced activation of the sympathetic nervous system likely contributes to the initiation and worsening of cardiometabolic risk factors, including elevated blood pressure, cardiac dysfunction, dyslipidaemia, increased fasting blood glucose, insulin res
