The gut-kidney-brain axis in chronic kidney disease: mechanisms and nutritional interventions for cognitive protection.

Chronic kidney disease (CKD) is increasingly recognised as a systemic disorder extending beyond renal impairment to include cognitive decline. The present review focuses on the mechanisms through which CKD and cognitive impairment are connected in the gut-kidney-brain axis, especially when considering the role played by nutrition as a possible target for interventions. Sources of literature were identified in PubMed, Scopus, Web of Science, and Google Scholar up until 2026 and included in the present analysis clinical trials, systematic reviews and studies in peer-reviewed journals related to

Targeting the Gut-Brain-Ferroptosis Axis: Molecular Mechanisms and Therapeutic Potential in Alzheimer's Disease and Epilepsy.

Alzheimer’s disease (AD) and epilepsy share underlying mechanisms of oxidative stress and neuroinflammation, yet effective targeted therapies remain limited. This review presents a novel integrative model linking gut microbiota dysbiosis to ferroptosis, an iron-dependent form of regulated cell death. We propose a vicious cycle in which dysbiosis promotes systemic inflammation and disrupts cerebral iron homeostasis, impairing the glutathione/GPX4 antioxidant system and sensitizing neurons to lipid peroxidation and ferroptotic death. This neuronal damage further fuels neuroinflammation and may e

The microbiota-gut-brain axis perspective: mechanisms and intervention strategies for the comorbidity of chronic constipation and depression.

BACKGROUND: Chronic constipation and depression are highly prevalent worldwide. These two conditions frequently co-occur in clinical practice. Accumulating evidence indicates that gut microbiota dysbiosis mediates this comorbidity through the microbiota-gut-brain axis (MGBA). METHODS: This narrative review systematically summarizes current research on MGBA bidirectional communication mechanisms, gut microbiota alterations in comorbid patients, and microbiota-targeted intervention strategies. RESULTS: The MGBA facilitates bidirectional communication through four major pathways: neural pathway

Combined exposure of cold and hypoxia: a driver for hypertension and the underlying role of the microbiota-gut-brain axis.

BACKGROUND: Cold and hypoxic conditions often coexist in high-altitude environments and are associated with alterations in blood pressure during short-term exposure. Increasing evidence suggests that the microbiota-gut-brain axis may be involved in blood pressure regulation under environmental stress. However, evidence remains scarce, especially the related mechanisms. AIMS: This study aimed to investigate whether short-term combined exposure to cold and hypoxia is associated with blood pressure elevation and to explore the potential involvement of the microbiota-gut-brain axis in this proces

Immunological Mechanisms of the Gut-brain-cardiovascular Axis in Coronary Heart Disease with Comorbid Anxiety and Depression.

Coronary heart disease (CHD) is frequently accompanied by anxiety and depression, conditions that markedly worsen cardiovascular outcomes. Increasing evidence indicates that immune dysregulation, driven by gut microbiota alterations, plays a central role in linking psychological disorders with cardiovascular pathology through the gut-brain-cardiovascular axis. This narrative review systematically summarizes recent advances in the immunological mechanisms underlying CHD complicated by anxiety and depression. We focus on gut microbiota-immune interactions, inflammatory signaling pathways, immune

The gut-liver-brain axis in alcohol use disorder: The emerging role of bile acid dysregulation.

Alcohol use disorder (AUD) is a chronic, relapsing neuropsychiatric disorder with limited treatment options. The gut-liver-brain axis has emerged as an important contributor to alcohol addiction. Within this axis, bile acids, important signaling molecules that connect the gut, liver, and central nervous system (CNS), have received increasing attention, as their metabolic dysregulation is closely associated with AUD. This review systematically elucidates the physiological metabolic processes of bile acids and the mechanisms by which chronic alcohol consumption disrupts their homeostasis. We pro

Iron Homeostasis and Reproduction: Unveiling the Microbiome-Gut-Brain Axis Connection in the Mosquito Anopheles culicifacies.

Our study investigated how adult female Anopheles culicifacies mosquitoes regulate systemic iron homeostasis after blood feeding, a process essential for reproduction, and revealed striking parallels to iron deficiency disorders in mammals. This study identifies that coordinated transcriptional regulation of ferritin and transferrin plays a crucial role in follicle development and egg maturation. Silencing of both genes using ribonucleic acid interference led to severe reproductive impairment, including ovarian arrest in 50% of females, a 40% reduction in oocyte number, and a decrease in first

Lipid-mediated brain-gut axis alterations in functional diarrhea and their role in oxidative stress and neurodegeneration.

Lipid mediators control intestinal motility, epithelial signalling, and neuronal function by integrating metabolic, hormonal, and neurological interactions throughout the brain-gut axis. Recurrent loose stools without structural abnormalities are the hallmark of Functional Diarrhoea (FD), a condition marked by high oxidative stress and disruptions in this axis. The main aim of this work is to further understand the role of lipid profiles in oxidative stress and neurological abnormalities in FD using the proposed Lipid-Mediated Brain-Gut Axis (LIMBRA) model. The LIMBRA system analyzes lipid abn…

Molecular insights into physiological impact of micro- and nano-plastics on the digestive system and gut-brain axis.

Microplastics (MPs) and Nanoplastics (NPs) represent an alarming and persistent threat to global human health, owing to their resilience and ubiquity in the environment. Ingestion via contaminated food and water is the primary exposure route, resulting in the accumulation of MNPs in key organs such as the gastrointestinal tract (GI), liver, and pancreas, highlighting the urgent need to understand their potential cumulative and systemic effects. This review critically evaluates recent molecular-level insights into the physiological impacts of MNPs, with particular emphasis on the GI system and

Microbial modulation of CNS remyelination in multiple sclerosis: the missing link in gut-brain axis research.

Multiple sclerosis (MS) is a chronic, immune-mediated disorder of the central nervous system (CNS) marked by demyelination and neurodegeneration. While much attention has focused on immune dysregulation and neuroinflammation, the failure of effective remyelination is a key driver of disease progression, especially in progressive MS. Recently, the gut microbiome has emerged as a potent modulator of systemic immunity and CNS function, influencing processes such as neuroinflammation and neurogenesis. This review examines current evidence on microbiota-derived metabolites, including short-chain fa

A Comprehensive Review of the Gut-Microbiota-Brain Axis in Alzheimer's Disease: From Pathophysiology to Potential Therapies.

The gut-microbiota-brain axis (GMBA), an intricate network connecting the gastrointestinal (GI) tract and the brain, plays a pivotal role in maintaining overall health and influencing disease processes. The human gut microbiota, comprising over 3000 bacterial species, regulates immune responses, hormonal signals, and metabolite production, maintaining homeostasis under normal conditions. Dysbiosis, or microbial imbalance, has been linked to various central nervous system (CNS) disorders, including Alzheimer's disease (AD), Parkinson's disease (PD), multiple sclerosis (MS), and autism spectrum

The Emerging Role of the Brain-Gut Axis in Amyotrophic Lateral Sclerosis: Pathogenesis, Mechanisms, and Therapeutic Perspectives.

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by progressive loss of motor neurons. Although genetic and environmental factors are established contributors, recent research has highlighted the critical role of the gut-brain axis (GBA) in ALS pathogenesis. The GBA is a bidirectional communication network involving neural, immune, and endocrine pathways that connect the gut microbiota with the central nervous system. Dysbiosis in ALS disrupts this axis, leading to increased intestinal permeability, neuroinflammation, and excitotoxicity. Notably, reduction

Contribution of the Brain-Gut-Microbiome Axis to Intergenerational Abnormalities in a Rat Model of Perioperative Neurocognitive Disorder.

BACKGROUND: The brain-gut-microbiome (BGM) axis is a communication network through which the brain and gastrointestinal microbiota interact via neural, hormonal, immune, and gene expression mechanisms. Gut microbiota dysbiosis is thought to contribute to neurocognitive disorders, including perioperative neurocognitive disorder, and to various metabolic abnormalities. Recently, the authors reported that sevoflurane induces neurocognitive deficits in exposed rats as well as their future offspring, with male offspring being particularly affected (intergenerational perioperative neurocognitive dis
