GSH promote renal cadmium efflux through ABCC1 of ABC family.

Accumulation of cadmium in the kidneys can lead to toxicity; therefore, it is crucial to identify methods that promote cadmium excretion to mitigate nephrotoxicity. However, the mechanisms underlying cadmium excretion from the kidneys remain unclear. The role of the ATP-binding cassette (ABC) transporter family in the renal excretion of cadmium was investigated in this study. We employed gene chip technology to identify alterations in gene and protein expression within the ABC transporter family, particularly focusing on ABCC1, in response to cadmium exposure. Utilizing CRISPR-Cas9 technology,…

mTOR-Mediated Autophagy Regulates Cadmium-Induced Kidney Injury via Pyroptosis.

The heavy metal cadmium (Cd) affects the global livestock production economy mainly through the contamination of feed raw materials and secondary contamination in feed processing, and it also poses a serious threat to food safety and human health. The nucleotide-binding oligomerization domain-like pyrin-domain-containing protein 3 (NLRP3) inflammasome is a key regulatory element of pyroptosis, which is engaged in kidney injury. Meanwhile, autophagy is also involved in renal inflammation. Mammalian target of rapamycin (mTOR) plays an important role in pyroptosis and autophagy, but its function …

Cadmium activates the innate immune system through the AIM2 inflammasome.

Cadmium (Cd) is a widely used heavy metal and has recently been recognized as a possible source of human toxicity due to its ability to accumulate in organs. Accumulation of heavy metals has several adverse effects, including inducing inflammation, in multiple organs, such as the testis. However, how Cd ions are sensed by host cells and how tissue inflammation eventually occurs remains unclear. Here, we show that Cd activates the AIM2 inflammasome by mediating genomic DNA release into the cytoplasm after DNA damage via oxidative stress, to trigger IL-1β secretion and pyroptosis. Specifically, …

Cadmium alters the cellular metabolome of human ovarian granulosa cells.

Cadmium (Cd) is a toxic heavy metal that has been extensively implicated in disordered folliculogenesis, but the mechanisms underlying the ovarian toxicity of Cd remain to be explored fully. Granulosa cells are key players in ovarian follicular development and are the primary cells affected by Cd exposure-induced damage and dysfunction. In this study, we investigated how various levels of exposure of Cd (3 and 10 μM) to human granulosa cells (KGN cells) impacted the metabolism of the KGN cells utilizing a non-targeted metabolomics methodology. In vitro cell experiments revealed that Cd exposur…

S-nitrosoglutathione reductase disfavors cadmium tolerance in shoots of Arabidopsis.

S-nitrosoglutathione reductase (GSNOR) is involved in the response to cadmium (Cd) exposure. In this study, the plants of mutant (gsnor1-3) with lossing-function of- and over-expression (GSNOROE5) of GSNOR were used to clear the role of GSNOR in Cd tolerance. GSNOR activity increased through upregulating the expression of the AtGSNOR gene and protein in Arabidopsis thaliana under Cd stress, which attenuated Cd tolerance. Oxidative damage was more serious in GSNOROE5 and was alleviated in gsnor1-3 under Cd stress, compared with Col-0. Induction of GSNOR facilitated H2O2 accumulation but inhibit…

Impaired PPARγ activation by cadmium exacerbates infection-induced lung injury.

Emerging data indicate an association between environmental heavy metal exposure and lung disease, including lower respiratory tract infections (LRTIs). Here, we show by single-cell RNA sequencing an increase in Pparg gene expression in lung macrophages from mice exposed to cadmium and/or infected with Streptococcus pneumoniae. However, the heavy metal cadmium or infection mediated an inhibitory posttranslational modification of peroxisome proliferator-activated receptor γ (PPARγ) to exacerbate LRTIs. Cadmium and infection increased ERK activation to regulate PPARγ degradation in monocyte-deri…

Impacts of cadmium on male fertility: Lessons learnt so far.

Cadmium (Cd) is one of the most dangerous heavy metals in the world. Globally, toxicities associated with cadmium and its attendant negative impact on humans and animals cannot be under-estimated. Cd is a heavy metal, and people are exposed to it through contaminated foods and smoking. Cd exerts its deleterious impacts on the testes (male reproductive system) by inducing oxidative stress, spermatogenic cells apoptosis, testicular inflammation, decreasing androgenic and sperm cell functions, disrupting ionic homeostasis, pathways and epigenetic gene regulation, damaging vascular endothelium and…

Cadmium inhibits forskolin-induced differentiation of human placental BeWo cells.

Cadmium (Cd) is an environmental pollutant. Blood Cd levels in pregnant women have been associated with premature births, infant birth size, placenta previa, and placenta accreta. There have been concerns on the reproductive developmental toxicity of Cd. The choriocarcinoma cell line BeWo, a cellular in vitro model for studying syncytial fusion, has been widely used to study the reproductive and developmental toxic effects of pollutants. Here, we examine the inhibitory effect of Cd against forskolin (FSK)-induced BeWo differentiation. Results showed that Cd exposure inhibited the FSK-induced e…

Cadmium-Induced Kidney Injury: Oxidative Damage as a Unifying Mechanism.

Cadmium is a nonessential metal that has heavily polluted the environment due to human activities. It can be absorbed into the human body via the gastrointestinal tract, respiratory tract, and the skin, and can cause chronic damage to the kidneys. The main site where cadmium accumulates and causes damage within the nephrons is the proximal tubule. This accumulation can induce dysfunction of the mitochondrial electron transport chain, leading to electron leakage and production of reactive oxygen species (ROS). Cadmium may also impair the function of NADPH oxidase, resulting in another source of…

Sulfate supply enhances cadmium tolerance in Vicia faba L. plants.

Sulfur deficiency and cadmium (Cd) pollution are two ubiquitous constraints affecting plant growth in agricultural soils. However, facing the situation of sulfur deficiency, whether surplus sulfate supply can affect Cd toxicity in plants is still unclear. Therefore, in the present study, experiments with deficient, sufficient, and excess sulfate levels under Cd stress were conducted in faba bean plants hydroponically. We found that sulfate supply significantly increased biomass of Cd-stressed plants when compared with deficient sulfate treatment. Intriguingly, sulfate application also increase…

Transformation of cadmium-associated schwertmannite and subsequent element repartitioning behaviors.

Schwertmannite is an important sink for cadmium (Cd) in acid mine drainage (AMD) environments and is unstable when environmental conditions change. However, the release and redistribution of Cd during schwertmannite transformation with respect to pre-bound Cd are poorly understood. In this work, the transformation of cadmium-associated schwertmannite and subsequent Cd repartitioning behaviors were investigated. The way of schwertmannite associated with Cd was predominant by absorption, and the diffuse layer model (DLM) showed that Cd2+ existed as monodentate complexes ≡Fe(1)OCd+ and ≡Fe(2)OCd+…

Ligustrazine modulates renal cysteine biosynthesis in rats exposed to cadmium.

The objective of this study was to determine the effect of ligustrazine (TMP) on cadmium (Cd)-induced nephrotoxicity and its relevant mechanism. TMP (50mg/kg) was injected intraperitoneally (i.p.) into rats 1h prior to CdCl2 exposure (at a Cd dose of 0.6mg/kg). TMP reversed Cd-induced nephrotoxicity, evidenced by the relatively normal architecture of the renal cortex. Additionally, TMP alleviated renal oxidative stress of rats that were exposed to Cd, evidenced by the decreased levels of malondialdehyde (MDA), 4-hydroxynonenal (4-HNE), elevated levels of glutathione (GSH) and GSH/GSSG (glutath…

An ascorbic acid sensor based on cadmium sulphide quantum dots.

We present a Förster resonance energy transfer (FRET)-based fluorescence detection of vitamin C [ascorbic acid (AA)] using cadmium sulphide quantum dots (CdS QDs) and diphenylcarbazide (DPC). Initially, DPC was converted to diphenylcarbadiazone (DPCD) in the presence of CdS QDs to form QD-DPCD. This enabled excited-state energy transfer from the QDs to DPCD, which led to the fluorescence quenching of QDs. The QD-DPCD solution was used as the sensor solution. In the presence of AA, DPCD was converted back to DPC, resulting in the fluorescence recovery of CdS QDs. This fluorescence recovery can …

Adsorptive removal of cadmium ions by Spirulina platensis dry biomass.

Cadmium is one of the most toxic substances found in aquatic ecosystems. This metal tends to accumulate in photosynthetic plants and fish and is transferred to humans causing many diseases. It has to be removed from our environment to reduce any health risks. Dry biomass of the microalga (cyanobacterium) Spirulina platensis was used as biosorbent for the removal of cadmium ions (Cd(2+)) from aqueous solutions. The effects of different levels of pH (3-9), biomass concentration (0.25-2 g), temperature (18-46 °C), metal concentration (40-200 mg/l) and contact time (30-120 min) were tested. Batch …

Kinesin-dependent motility generation as target mechanism of cadmium intoxication.

The anterograde vesicle transport within neurons critically depends on microtubules and on the activity of kinesin. The present study demonstrates that cadmium ions inhibit the in vitro assembly of microtubules from tubulin, whereby at high cadmium levels (∼500 μM) unstructured protein aggregates were formed. Cadmium ions also significantly lower both the ATPase and motility activity of neuron-specific kinesin KIF5A in concentration-dependent manner. For the inhibition of KIF5A ATPase activity, an IC50 value of 10.4±1.5 μM was determined. Inhibition could be widely compensated by addition of E…
