Influence of decreased and increased magnesium supply on the cardiotoxic effects of epinephrine in rats.

Female Sprague-Dawley rats kept on a standard chow or on a magnesium (Mg)-deficient diet during 6 days received s.c. injections of 0, 25, 50, 100 and 200 micrograms of epinephrine (Ep). 1 h before, they were orally treated with 0, 125 or 250 mg of Mg/kg b.w. given as magnesium aspartate hydrochloride. Drug-induced changes were studied by analyzing serum and cardiac tissue samples (Mg, Ca, K, Na and in addition glucose, FFA, cholesterol and creatine kinase in serum) taken at 7 different times (15 to 420 min) after treatment with Ep; effects were evaluated by considering the respective areas und

Mitochondrial dysfunction-driven PANoptosis in doxorubicin-induced cardiotoxicity: mechanistic insights and intervention strategies.

Doxorubicin (DOX) is a broad-spectrum anthracycline chemotherapeutic agent, and its clinical application is severely limited by dose-dependent cardiotoxicity (DIC), for which there are currently no effective clinical interventions. Mitochondria are the central organelles regulating myocardial energy metabolism and cell survival, and mitochondrial dysfunction is considered the initiating and core mechanism underlying DIC. DOX disrupts the mitochondrial quality control (MQC) system and induces mitochondrial metabolic reprogramming, thereby leading to mitochondrial dysfunction. This results in ex…

Integration of network toxicology and zebrafish models to elucidate bisphenol BP-induced cardiotoxicity.

Bisphenol BP (BPBP), a structural analog of bisphenol A (BPA), is increasingly utilized in industrial applications; however, its potential hazards to the cardiovascular system remain unexplored. This study employed an integrated strategy combining network toxicology, molecular docking, and in vivo zebrafish models to systematically elucidate the cardiovascular toxicity of BPBP. Network toxicology identified several candidate hub genes and pathways potentially associated with BPBP-induced cardiotoxicity. Molecular docking further suggested favorable predicted interactions between BPBP and selec…

AKT (Ser473) suppression mediates ceritinib cardiotoxicity through autophagic flux impairment and mitochondrial injury.

Ceritinib, an anaplastic lymphoma kinase (ALK) inhibitor, is associated with cardiovascular adverse events, yet the mechanisms remain incomp … 3β (Ser9) phosphorylation. Together, these findings establish that AKT suppression drives ceritinib cardiotoxicity through autophagic flux impairment and mitochondrial injury, and position AMPK-driven, TFEB-associated lysosomal restoration as a mechanism-based cardioprotective strategy independent of AKT recovery.

Targeting the Apelin-APJ Axis: A Promising Strategy to Mitigate Anthracycline-Induced Cardiotoxicity.

Anthracycline-induced cardiotoxicity remains a major clinical challenge, often progressing to heart failure years after therapy. Conventional cardioprotective agents, including angiotensin-converting enzyme inhibitors, angiotensin receptor blockers, and beta-blockers, are widely used to preserve cardiac function; however, their effectiveness is limited by their inability to comprehensively address the complex, multifactorial pathophysiology of anthracycline-induced cardiotoxicity. This underscores the critical need for more effective and mechanism-based cardioprotective strategies that directl…

Integrated stress response inhibition decouples cardiotoxicity from anthracycline chemotherapy while enhancing antitumor efficacy.

The clinical efficacy of anthracycline chemotherapeutics is severely limited by their dose-dependent cardiotoxicity, a condition lacking effective preventive strategies. We identify the maladaptive ATF4-dependent integrated stress response (ISR) program as a central driver of this pathology. In murine models, ISR inhibition achieved by either cardiomyocyte-specific knockout of the transcription factor ATF4 or the ISR inhibitor (ISRIB), provides robust protection from doxorubicin (DOX)-induced cardiac dysfunction, remodeling, and injury. Mechanistically, we delineate a regulatory pathway in whi…

Methionine is an essential amino acid in doxorubicin-induced cardiotoxicity through modulating mitophagy.

Doxorubicin (Dox) is a widely used anticancer drug. However, its time- and dose-dependent side effects, particularly severe cardiotoxicity, limit its clinical use. Understanding the molecular mechanisms underlying Dox-induced cardiotoxicity has become a research focus in recent years. Among these, impaired mitophagy which participated in the process of damaged mitochondria clearance, is considered one of the key mechanisms in Dox-induced cardiomyopathy. Methionine (Met) is an essential amino acid that plays a crucial role in various biological processes. This study aims to investigate the role…

Targeting the NLRP3 by Natural Compounds: Therapeutic Strategies to Mitigate Doxorubicin-Induced Cardiotoxicity.

Doxorubicin (DOX), a widely utilized anthracycline chemotherapy agent, is known for its potent anticancer efficacy across various malignancies. However, its clinical use is considerably restricted due to the risk of dose-dependent cardiotoxicity, which can lead to long-term heart dysfunction. The underlying mechanism of DOX-induced cardiotoxicity has been associated with the formation of reactive oxygen species (ROS) and disrupting cellular signaling pathways. This is particularly relevant to the activation of the NLRP3 inflammasome, which triggers inflammation and pyroptosis in cardiac cells.…

Targeting ligand PDL1 for cardiotoxicity assessment and cardiac protection in immune-related myocarditis.

AIMS: The role of cardiac programmed cell death ligand 1 (PDL1) in immune checkpoint inhibitor (ICI) - related myocarditis (irMyocarditis) remains unclear. We aimed to investigate whether ligand PDL1 could serve as an early indicator and a potential therapeutic target for irMyocarditis. METHODS: Cardiac PDL1 expression was assessed using single-nucleus RNA sequencing and multiplex immunohistochemistry in human patients and mouse models of irMyocarditis. A PDL1-targeted magnetic resonance imaging (MRI) nanoprobe was developed for noninvasive imaging of irMyocarditis. Additionally, an adeno-ass

Enhancing cardiac safety: Liposomal ciprofloxacin mitigates anthracycline-induced cardiotoxicity without compromising anticancer efficacy.

Anthracyclines, such as doxorubicin and epirubicin (EPI), are integral in the treatment of solid tumors and hematological malignancies but are associated with cardiotoxicity, potentially leading to heart failure. The underlying mechanisms involve the generation of reactive oxygen species (ROS), alterations in iron metabolism, and the inhibition of topoisomerase 2β (Top2β), resulting in mitochondrial dysfunction and cell death. Fluoroquinolones, including ciprofloxacin (CPX), enhance the efficacy of anthracyclines by inhibiting topoisomerase II and inducing apoptosis, thereby indicating a promi…

Neurotoxicity and cardiotoxicity alleviation by polystyrene microplastics in cadmium sulfide-exposed zebrafish larvae.

Our recent study confirmed the polystyrene microplastics (PS MPs) promote the release of Cd2 + from cadmium sulfide (CdS) pigment under sunlight irradiation, potentially increasing environmental risk. However, despite the anticipated increase in toxicity due to Cd2+ release, PS MPs were found to mitigate the toxicity of CdS pigment, although the underlying mechanisms remain unclear. In this study, sunlight irradiation induced significant neurotoxicity and cardiotoxicity in zebrafish larvae exposed to CdS pigment, as evidenced by reduced spontaneous movement, decreased total locomotor distance,…
