Mechanistic insights into glufosinate ammonium‑induced hepatotoxicity in grass carp: Integrating network toxicology, transcriptomics, experimental validation and intervention strategies.

Glufosinate ammonium (GLA) residues in water threaten aquatic organisms and pose ecological risks to aquaculture. Current understanding of the molecular mechanisms driving GLA-induced hepatotoxicity in aquatic species remains incomplete, with effective intervention strategies still lacking. This study aims to elucidate the molecular mechanisms underlying hepatotoxicity GLA in grass carp, and to evaluate the intervention effects of activating the cholinergic anti-inflammatory pathway (CAP). Herein, the toxicokinetic characteristics revealed that GLA is highly hydrophilicity and suitable intesti

Aeromonas hydrophila induces mitophagy for intracellular survival through its secreted effectors in the splenic macrophages of grass carp (Ctenopharyngodon idella).

Aeromonas hydrophila, a Gram-negative bacterium ubiquitously distributes in aquatic environments, represents a prevalent pathogenic threat to aquatic animals. The present study investigated how A. hydrophila induced mitophagy to promote its own survival in the splenic macrophages of grass carp (Ctenopharyngodon idella). Our results revealed that A. hydrophila impaired mitochondrial integrity by reducing mitochondrial membrane potential and ATP levels, as well as increasing mitochondrial ROS (mtROS) production. Meanwhile, this bacterium decreased the expression of the TOM20, a mitochondrial out

Genome-Wide Identification of the TRAF Gene Family in Common Carp (Cyprinus carpio) and Analysis of Their Expression in Response to CyHV-3 Challenge.

Tumor necrosis factor receptor-associated factors (TRAFs) are key signal transduction molecules in innate immunity and are involved in multiple immune pathways, such as TNF receptor and Toll-like receptor signaling. In this study, 16 CcTRAFs were identified from the common carp genome, assigned to six subfamilies (TRAF1/2/3/4/6/7), with no TRAF5 found. Significant paralogous expansion was observed in the TRAF2/4 subfamilies. All CcTRAF proteins were predicted to be unstable and hydrophilic by ProtParam analysis; most contain conserved RING, TRAF and MATH domains, whereas CcTRAF7 carries seven

R-spondin1 plays an indispensable role in ovarian development of Qi River crucian carp (Carassius auratus) by regulating estrogen synthesis.

R-spondin1 (Rspo1) is a member of the secreted furin-like domain-containing protein family, and it is recognized for its significance in mammalian ovarian development. However, its role in teleost ovarian development remains largely uninvestigated. The Qi River crucian carp (Carassius auratus) is a species capable of gynogenesis, and it encounters challenges of premature ovarian maturation in aquaculture settings. Previous research established the essential involvement of Rspo1 in oocyte growth in Qi River crucian carp, but the precise molecular mechanisms underlying its role remain poorly und

Caffeic acid phenethyl ester improves high-carbohydrate diet utilization by promoting adipocyte hyperplasia in grass carp (Ctenopharyngodon idellus).

Enhancing the ability of fish to consume a high-carbohydrate diet (HCD) is a key focus of aquaculture research. The propolis extract, caffeic acid phenethyl ester (CAPE) has anti-inflammatory, hepatoprotective, and glycolytic-promoting properties, but its potential to mitigate metabolic disorders in fish fed a HCD remains uncertain. This study investigated the effects of CAPE on the adaptability and utilization of a HCD in the herbivorous grass carp (Ctenopharyngodon idellus), focusing on growth performance, tissue and organ health, and nutrient metabolism. A total of 270 grass carp with an in

The role of β-catenin in ovarian differentiation by upregulation cyp19a1a in a natural triploid teleost, Qi River crucian carp (Carassius auratus).

The Qi River crucian carp (Carassius auratus), a gynogenetic all-female fish, reproduces both unisexually and sexually, but the underlying molecular mechanisms remain unclear. The Wnt/β-catenin pathway influences mammalian ovarian development via cytochrome P450 family 19 subfamily A member 1 (CYP19A1) activation, however, its role in teleosts is poorly understood. Here, we investigated β-catenin's involvement in gonadal differentiation through histological and molecular analyses. Histological examination with Hematoxylin and Eosin (HE) staining and immunohistochemistry showed that oogonia eme

Ferulic acid alleviates long-term avermectin-induced damage to the spleen of carp and restores its inflammatory response and oxidative balance.

Extensive use of avermectin (AVM) in agriculture can seriously contaminate fish in aquaculture. Ferulic acid (FA) is a strong antioxidant sourced from plants and is widely applied in food and medicine, but its protective function on the immunological damage caused by AVM is still lacking. In this study, carp were chronically exposed to (1/10 96 hr LC50) AVM for 30 day(s), with a dietary supplement of FA (400 mg/kg) to assess its effects on carp spleen. The experimental groups (n = 10) included: control, AVM-exposed (2.404 µg/L), FA + AVM co-treated, and FA alone. Long-term AVM exposure altered

HSC70 functions as a negatively regulator in IFN signaling pathway via suppressing K63-linked ubiquitination of RIG-I in black carp.

Heat shock cognate 70 (HSC70), a highly conserved molecular chaperone in the heat shock protein 70 (HSP70) family, plays an essential role in maintaining the homeostasis of the cellular environment. Furthermore, although previous studies have investigated potential function of HSC70 in innate antiviral immunity, further research is still required to fully elucidate its role. In this study, we cloned and characterized the HSC70 homolog gene from black carp (Mylopharyngodon piceus), which consists of 1950 nucleotides encoding 650 amino acids, migrates at approximately 71 kDa on SDS-PAGE, and is

Perfluorooctane sulfonate causes pyroptosis and lipid metabolism disorders through ROS-mediated NLRP3 inflammasome activation in grass carp hepatocyte.

The surfactant perfluorooctane sulfonate (PFOS) is widely produced worldwide. It is a persistent organic pollutant in the aquatic environment and poses a serious threat to aquatic organisms, as PFOS exposure can cause liver injury in a wide range of organisms. However, it is unclear whether PFOS exposure-induced hepatocellular injury in fish is associated with ROS-mediated activation of NLRP3 inflammasome. In this study, various PFOS concentrations were applied to L8824 cells, a cell line of grass carp hepatocytes. The detrimental impacts of PFOS on oxidative stress, pyroptosis, lipid metaboli

Silybin prevented avermectin-induced cardiotoxicity in carp by modulating oxidative stress, inflammation, endoplasmic reticulum stress, mitochondrial apoptosis, and autophagy.

Avermectin is one of the widely used anthelmintics in aquaculture and exhibits substantial toxicity to aquatic organisms. Silybin is extensively used for its anti-inflammatory, antioxidant and anti-apoptotic biological properties. Heart is essential for the survival of fish and plays a vital role in pumping blood oxygen and nutrients. Residual avermectin in water poses harm to carp. However, there is still insufficient research on whether silybin can mitigate the toxicity of avermectin to carp heart tissues. In this research, we established a model involving carp subjected to acute avermectin

Use of Cryopreserved Sperm of Grass Carp (Ctenopharyngodon idella) for Seed Production at the Hatchery Level of Bangladesh-A Need for Development of Germplasm Repositories.

Grass carp (Ctenopharyngodon idella), a freshwater nonnative fish species, is a potential aquaculture candidate in Bangladesh. The seed of the species is produced in the hatcheries by hypophysation, but the quality of seedstock of grass carp is deteriorated due to inbreeding, negative selection, and interspecific introgression among fishes. To increase the availability of quality seed and best genetic traits of grass carp, this study dealt with finding suitable conditions of sperm cryopreservation protocols and evaluated the effectiveness of cryopreserved sperm through breeding trials of C. id

An Oral Microencapsulated Vaccine Loaded by Sodium Alginate Effectively Enhances Protection Against GCRV Infection in Grass Carp (Ctenopharyngodon idella).

Grass carp reovirus (GCRV) is highly infectious and lethal to grass carp, causing huge economic losses to the aquaculture industry annually. Currently, vaccination is the most effective method against viral infections. Among the various vaccination methods, the oral vaccination is an ideal way in aquaculture. However, low protective efficiency is the major problem for oral vaccination owing to some reasons, such as antigen degradation and low immunogenicity. In our study, we screened the antigenic epitopes of GCRV-II and prepared an oral microencapsulated vaccine using sodium alginate (SA) as
