[Alteration of cholera toxin biosynthesis in Vibrio cholerae 01 as a result of temperate phage 139 integration into bacterial chromosome].

Infection of V. cholerae 01 (classical and eltor biovars) cells with the temperate cholera phage 139 derived from V. cholerae serogroup 0139 followed by integration of the phage genome into the bacterial chromosome significantly increased the production of cholera toxin, the main virulence factor. The level of toxin biosynthesis in the lysogenic V. cholerae classical strain increased 3-fold and that in V. eltor thirty times in comparison with the parental strains. Increased production of cholera toxin was not associated with an increase in the number of copies of genes involved in its biosynth…

Evolution of pandemic cholera at its global source.

The seventh pandemic of cholera, caused by the seventh pandemic El Tor lineage of Vibrio cholerae, was previously shown to have emanated in three global waves from the Bay of Bengal, bordering Bangladesh and India1. However, the respective roles of the Ganges Delta and Basin regions in seeding these global pandemic waves were not known. Here we show that, although transmission events occur between Bangladesh and India, V. cholerae in the two countries has largely evolved separately over the past 20 years, apparently constrained by national borders rather than by hydrological features, such as …

Repeated Extraneous Introductions of Cholera, Thailand, 2007-2025.

Genomic analyses identified 4 seventh pandemic Vibrio cholerae El Tor clades in Thailand (2007-2025). Closely related to other South Asian strains, the clades reveal that repeated cross-border introductions, rather than local persistence, drive outbreaks. Our findings highlight the importance of genomic surveillance for monitoring transmission and informing regional control strategies.

Risk factors for cholera mortality: A scoping review.

OBJECTIVES: Cholera is an easily treatable disease, but many people are still unnecessarily dying from it. To improve current case management practices and prevent mortality requires a comprehensive understanding of who is at higher risk of dying. To identify the most common risk factors, a scoping review was undertaken, to explore the literature and summarise the evidence on cholera mortality and reported risk factors. METHODS: Following the scoping review framework proposed by Arksey and O'Malley (2005), Pubmed, EMBASE, Web of Science, LILACS, Scielo, Cochrane and Open Grey and African Jour…

Cholera resurges in Zambia: Challenges and future directions.

Cholera remains a major global health challenge, disproportionately affecting low-income countries with inadequate water, sanitation, and hygiene (WASH) infrastructure. Zambia, historically prone to outbreaks, experienced a severe resurgence in 2023-2024, with over 10,887 cases and 432 deaths. The contributing factors include poor WASH access, systemic health care weaknesses, stigma, and logistical barriers. Only 32% of households have basic water services, and health care worker shortages persist, especially in densely populated areas. Misinformation and cultural practices hinder public healt…

Enhanced cholera surveillance to improve vaccination campaign efficiency.

Systematic testing for Vibrio cholerae O1 is rare, which means that the world's limited supply of oral cholera vaccines (OCVs) may not be delivered to areas with the highest true cholera burden. Here we used a phenomenological model with subnational geographic targeting and fine-scale vaccine effects to model how expanding V. cholerae testing affected impact and cost-effectiveness for preventive vaccination campaigns across different bacteriological confirmation and vaccine targeting assumptions in 35 African countries. Systematic testing followed by OCV targeting based on confirmed cholera yi…

Nanoparticles-based technologies for cholera detection and therapy.

Cholera is a waterborne disease caused by Vibrio cholerae bacteria generally transmitted through contaminated food or water sources. Although it has been eradicated in most Western countries, cholera continues to be a highly transmitted and lethal disease in several African and Southeast Asian countries. Unfortunately, current diagnostic methods for cholera have challenges including high cost or delayed diagnoses that can lead to increased disease transmission during pandemics, while current treatments such as therapeutic drugs and vaccines have limited efficacy against drug-resistant serogrou…

Rise of cholera in Iraq: A rising concern.

With over seven pandemics and four million reported cases, Cholera remains of the most prevalent acute watery diarrheal diseases in the world to date. As in other developing countries, Iraq once again combats Cholera - and its past encounter in 2015, where the death toll reached 1500, highlights the importance of quickly addressing the current outbreak. The war-torn state of the nation, malnourished public, lack of sanitation and hygiene, mass displacement and global warming all contribute to the prevalence of Cholera in Iraq. Along with the current efforts, additional strategies are recommend…

Cholera in travellers: improving vaccination guidance in Europe.

BACKGROUND: Cholera is endemic in ~50 countries worldwide and remains a disease associated with poverty, causing illness and death in the poorest and most vulnerable people. In travellers, cholera is considered a low-incidence disease, but the true impact on travellers is difficult to assess. Cholera vaccination may improve safety for certain European travellers at risk. Effective vaccines are available; however, vaccination recommendations in Europe vary considerably between countries. METHODS: In this review, a comparison of cholera vaccination recommendations from 29 advice reference bodie…
