Prevalence of PCV2 in Austrian and German boars and semen used for artificial insemination.

Since epidemiologically-based science on PCV2 in porcine semen is patchy, we investigated 806 Austrian (A) and German (G) AI boars from five studs, and boars from Austrian farms used for on-farm semen collection, for the presence for IgG/IgM in blood by ELISA (n=754) as well as for PCV2 DNA in semen (n=472) and if positive, also in blood of a few boars by nested PCR and sequencing. A total of 420 boars were tested for both PCV2 in semen and antibodies in blood. Boars were aged between 8 and 82 months at sampling. None of the boars tested positive for IgM but 60.1% did for IgG. PCV2 DNA was det

Effect of PCV-2 Vaccination on Cytokines Gene Expression Profile in Wild Boar Peripheral Blood Mononuclear Cells after Stimulation with Mycobacteria Antigens.

Wild boar (Sus scrofa) is a common wild ungulate known as the most important reservoir of tuberculosis (TB) in Spain. The severity of TB lesions in this species and the high prevalence of porcine circovirus type 2 (PCV-2) have been related. PCV-2 is ubiquitous in swine populations, being usual for the free-living ones the contact with this agent. Recent studies found a correlation between a decrease of generalised TB prevalence in wild boar populations and the PCV-2-vaccination. The aim of this study was to find out if PCV-2 vaccination modulates the gene expression of cytokines from immune ce…

Diet Metabarcoding Reveals Correlations Between the Consumption of Slugs and Hepatic Toxicants in European Hedgehogs (Erinaceus europaeus).

The European hedgehog (Erinaceus europaeus) is currently designated as near threatened by the International Union for the Conservation of Na … gehogs. We found no relation between diet and infection with pathogens (Corynebacterium ulcerans or circovirus). However, diet richness was negatively correlated with hepatic concentration in brodifacoum, a widely used rodenticide. Moreover, the relative abundance of Stylommatophora in the hedgehog diet was positively correlated with their hepatic concentrat …

Development of a recombinase polymerase amplification isothermal assay for rapid visual and lateral flow detection of porcine parvovirus-7.

Porcine parvoviruses (PPVs) are globally recognized as significant contributors to reproductive failure in swine, primarily due to their ass … biting no cross-reactivity with other common porcine pathogens such as porcine sapelovirus, porcine circovirus and classical swine fever virus. Of the 167 field samples tested, 23 were positive for PPV-7, corresponding to a positivity rate of 13.77%. Operating at a low and constant temperature, the assay eliminates the need for advanced laboratory equipment, …

Subunit vaccine of PCV3 capsid protein produced by sf9 cells with double knockout of Caspase-1 and Dronc induces strong immune response in mice.

Porcine circovirus type 3 (PCV3) associated with multisystemic clinicopathological diseases in swine herds has caused economic losses and there is no available commercial vaccine. Production of PCV3 capsid protein (Cap) by Spodoptera frugiperda 9 (sf9) cells using baculovirus expression vector system (BEVS) is a valid strategy to develop vaccines. Here, we report that subunit vaccine of PCV3 produced by sf9 cells with double knockout of Caspase-1 and Dronc genes induces strong immune response in mice. Three kinds of knockout sf9 cells aimed at Caspase-1 gene, Dronc gene and both genes were suc…

Engineering a bivalent nanoparticle vaccine with PCV2 capsid protein and PRRSV epitopes.

BACKGROUND: Coinfection with multiple viruses is a common occurrence in the pig industry. Porcine reproductive and respiratory syndrome virus (PRRSV) and porcine circovirus type 2 (PCV2) are two significant pathogens, with a notable prevalence of their coinfection observed in clinical settings. However, reports on corresponding combination vaccines against PRRSV and PCV2 are scarce. The capsid (Cap) protein encoded by PCV2 is highly immunogenic, and recombinant Cap expressed in vitro can self-assemble into virus-like particles (VLPs), providing a promising strategy for developing bivalent vacc…
