Type II collagen autoimmunity and Ménière's disease.

Antibodies to type II collagen have been reported by some authors to be raised in patients with Ménière's disease. In this study the antibody levels to type I and II collagen have been measured in 37 patients with Ménière's disease and 20 controls, using a solid-phase, double-antibody, enzyme-linked, immunoassay. No significant difference in antibody levels between the two groups was found. These findings do not support previously reported work which suggests that some cases of Ménière's disease are due to type II collagen autoimmunity.

Myocardial collagen matrix remodelling in arterial hypertension.

The cardiac interstitium is composed of non-myocyte cells and a structural fibrillar protein network which plays a dominant role in governing the structure, architecture, and mechanical behaviour of the myocardium. Herein we review the fibrillar collagen network, its various components, and the functions they serve in the normal and structurally remodelled myocardium in arterial hypertension. The heterogeneity in myocardial structure, created by the altered behaviour of non-myocyte cells, particularly cardiac fibroblasts, which are responsible for collagen synthesis or degradation and thereby …

[Filling cysts with type 1 bone collagen].

The good properties of bovine bone collagen type 1 for filling of bony defects was demonstrated in 56 cysts of the alveolar bone in 52 patients. 49 cysts (approx. 87%) healed without irritation. 8 months postop. complete reossification of the defects had been achieved in 20 (approx. 36%) of the cysts. Further 32 defects (approx. 57%) were markedly reduced. The discussion will compare the advantages of bone collagen and other materials.

Ureteral replacement using collagen sponge tube grafts.

The use of a collagen sponge tube graft as a material for segmental ureteral replacement was investigated. The structural design of the collagen sponge graft was achieved by cell culture on the matrix. MGH-U1 cells, derived from bladder cell carcinoma, were grown in vitro on the collagen sponge matrix with excellent biocompatibility and without evidence of cytotoxicity. The collagen sponge demonstrated biodegradability when implanted subcutaneously in dogs. However, a urine exposure test of collagen sponge in rat bladders revealed extensive salt deposits on its surface in some rats, as observe…

Can collagen metabolism be controlled: theoretical considerations.

Pharmacologic alterations of the inflammatory response and collagen metabolism will lead to control of the many disease states of fibrosis. Many of these diseases kill and cripple far more than hypertrophic scar. Far too little research is being directed in this area. The objectives of controlling fibrosis by interfering with collagen crosslinking were defined clearly by Peacock. Perhaps topical BAPN will be the first agent to bring a practical clinical reality to his predictions made over 20 years ago.

Injectable collagen: a six-year clinical investigation.

An injectable form of bovine dermal collagen has been developed for use in correcting cutaneous contour deficiencies. This material is a sterile device composed predominantly of highly purified type I collagen dispersed in phosphate-buffered physiological saline containing 0.3% lidocaine. The safety and efficacy of injectable collagen, which is now in widespread clinical use in the United States, Canada, England, Germany, France, Switzerland, Brazil, and 11 other countries, were evaluated in a 6 1/2-year investigation in which 5,109 patients were evaluated by 728 dermatologists and plastic sur…

[HLA system and collagen diseases (author's transl)].

Association of histocompatibility antigen (HLA antigens) with collagen diseases is not a casual event. Even known data, the practical conclusions have a limited usefullness because there is not a definitive correspondence between HLA antigens and rheumatic diseases. With HLA typing is possible to determine risk populations. The pauciarticular type II of juvenile reumatoid arthritis (J.R.A) is associated with HLA-B27 in the 75% of cases. Also, this antigen is associated with ankylosing spondylitis, Reiter's disease, Behçet disease, psoriasic arthritis, reactive arthritis (yersinia, shigella, sa…

Connective tissue structure: cell binding to collagen.

Established lines of fibroblasts have been shown to adhere to collagen substrates via a serum-derived glycoprotein. The attachment of various other cells to collagen types I-IV is examined here. Cells such as human skin fibroblasts, periosteum, hepatocytes, connective tissue cells, and monocytes required the serum glycoprotein and adhered equally well to all collagens, but attachment of chondrocytes, epidermal cells, and neutrophils was inhibited by the serum glycoprotein. Attachment of 2 tumorigenic cells, an osteosarcoma and a fibrosarcoma, was found to be unaffected by the serum glycoprotei…

Abnormally soluble collagen produced in fibroblasts cultures.

Abnormally soluble collagen is synthesized in vitro not only by skin fibroblasts of Marfan patients but also by those of patients with Ehlers-Danlos type V and cutis laxa. The excessive solubility of collagen is corrected by the addition to the culture medium of a synthetic flavonoid, (+)- catechin.

Targeting the D93 cryptic collagen epitope alters integrin α2β1-dependent cellular migration and collagen remodeling in metastatic breast cancer.

Targeting the tumor-associated extracellular matrix (ECM) offers a promising strategy for breast cancer therapy. During cancer progression, collagen remodeling within the ECM exposes cryptic collagen epitope sites that antibodies can selectively recognize. Here, we investigate the therapeutic potential of targeting the D93 cryptic collagen epitope in 3D human metastatic breast cancer spheroids derived from MDA-MB-231 and MCF10CA1a (M4) cell lines embedded in collagen type I hydrogels. Treatment with monoclonal antibody (mAb) D93 reduced cellular migration into collagen type I hydrogels, an eff…

Evaluation of collagen sponge and xenografts versus collagen sponge alone in alveolar ridge preservation: a randomized controlled clinical trial.

This study evaluated the clinical, radiographic, and histological outcomes of alveolar ridge preservation (ARP) using collagen sponge with xenografts (CS + Xenografts) versus collagen sponge (CS) alone, compared to spontaneous healing. 36 extraction sockets were randomly allocated into three groups: Group I (CS + Xenografts), Group II (CS alone), and Group III (control) spontaneous healing. Soft tissue assessment and CBCT imaging were conducted before tooth extraction (baseline) and 6 months post-extraction, followed by histologic and histomorphometric analysis of bone biopsies. Groups I and I…

A highly biocompatible CE-crosslinked collagen implant with exceptional anti-calcification and collagen regeneration capabilities for aging skin rejuvenation.

Skin aging, a complex and inevitable biological process, results in wrinkles, dermal laxity, and skin cancer, profoundly influencing appearance and overall health. Collagen serves as the fundamental element of the dermal matrix; nevertheless, collagen is susceptible to enzymatic degradation within the body. Crosslinking is employed to enhance the physicochemical properties of collagen. However, conventional crosslinking agents may harbor potential issues such as cytotoxicity and calcification risks, constraining their application in the biomedical field. Therefore, we have for the first time d…

Targeting HIF-activated collagen prolyl 4-hydroxylase expression disrupts collagen deposition and blocks primary and metastatic uveal melanoma growth.

Uveal melanoma (UM) is the most prevalent primary intraocular malignancy in adults, and patients that develop metastases (~50%) survive <1 year, highlighting the urgent need for new therapies. TCGA has recently revealed that a hypoxia gene signature is associated with poor UM patient prognosis. Here we show that expression of hypoxia-regulated collagen prolyl-4-hydroxylase genes P4HA1 and P4HA2 is significantly upregulated in UM patients with metastatic disease and correlates with poor prognosis, suggesting these enzymes might be key tumor drivers. We targeted hypoxia-induced expression of P4H…

Histological analysis of hyalinised keloidal collagen formation in earlobe keloids over time: collagen hyalinisation starts in the perivascular area.

Keloids grow and do not regress. They are characterised histologically by hyalinised keloidal collagen (HKC). HKC amounts vary, and the mechanism by which they form is unclear. To clarify how HKCs form and whether their formation associates with specific clinical features, we studied the histological findings of earlobe keloids and compared them with respective clinical features. A total of 50 earlobe keloids from 43 patients were used for histological analysis of keloid size (mm2 ), HKC area (mm2 ) and HKC area ratio (%). As a result, keloid durations ranged from 3 months to >13 years. Early-…

Effects of oral administration of tripeptides derived from type I collagen (collagen tripeptide) on atherosclerosis development in hypercholesterolemic rabbits.

Digestion of type I collagen with a collagenase-type protease yields a collagen tripeptide (Ctp) fraction comprising Gly-X-Y sequences that exhibit diverse biological activities. We previously demonstrated that Ctp inhibits the proliferation and migration of cultured aortic smooth muscle cells (SMCs) in vitro. These cells contribute to the pathogenesis of atherosclerosis and other cardiovascular diseases. In order to evaluate the effects of Ctp on atherosclerosis development in vivo, here we used the Kurosawa and Kusanagi-hypercholesterolemic (KHC) rabbit model of familial hypercholesterolemia…
