G Protein-Mediated Allosteric Modulation of Ligand Binding in Class A GPCRs: Receptor-Ligand-Transducer Ensembles in Disease and Drug Discovery.

G protein-coupled receptors (GPCRs) are dynamic allosteric proteins whose signaling properties are governed by reciprocal communication between extracellular ligand-binding sites and intracellular transducer interfaces. Although classical pharmacological models established the concept that ligand binding and G protein coupling are thermodynamically linked, recent structural, biophysical, and computational studies have revealed a far more complex picture in which GPCRs exist as ensembles of interconverting conformational states. Accumulating evidence indicates that G proteins function not only

Modeling and optimizing trust-mediated influence in temporal heterogeneous networks.

Public health communication increasingly depends on networked interactions in which attitudes and behavioral intentions emerge through repeated peer exchange and institutional contact, yet trust remains weakly represented in many computational diffusion and graph-learning frameworks, particularly when privacy shocks abruptly reshape credibility. We present a proof-of-concept, mechanism-grounded framework that treats trust as a bounded, directed, and diffusible state on a temporal heterogeneous graph and couples that state to learned influence pathways and budget-constrained intervention optimi

Deep learning to convert unstructured CT pulmonary angiography reports into structured reports.

BACKGROUND: Structured reports have been shown to improve communication between radiologists and providers. However, some radiologists are concerned about resultant decreased workflow efficiency. We tested a machine learning-based algorithm designed to convert unstructured computed tomography pulmonary angiography (CTPA) reports into structured reports. METHODS: A self-supervised convolutional neural network-based algorithm was trained on a dataset of 475 manually structured CTPA reports. Labels for individual statements included "pulmonary arteries," "lungs and airways," "pleura," "mediastin

Organosulfur scaffolds as quorum sensing and biofilm modulators in Gram-negative bacteria.

Quorum sensing (QS) is a cell-density-dependent communication system that coordinates bacterial virulence, biofilm formation, motility, and stress adaptation, making it a compelling target for antivirulence intervention. Due to the possibility of sulfonyl (-SO₂-) and sulfinyl (-S = O-) functional groups to impart specific electronic and steric properties that can modulate bacterial signaling and receptor interactions, organosulfur compounds are increasingly gaining attention. This review highlights recent advances in the development of sulfonyl- and sulfinyl-containing organosulfur compounds a

Investigating potency of TMC-126 against wild-type and mutant variants of HIV-1 protease: a molecular dynamics and free energy study.

A detailed computational study was performed to investigate the conformational changes of flap region and the mechanism underlying the binding of the inhibitor TMC-126 to HIV-1 protease (PR1) and its mutant variants through molecular dynamics simulations in conjunction with the molecular mechanics Poisson-Boltzmann (MM-PBSA) free energy calculation. Further, we have studied the effectiveness of the inhibitor against HIV-2 protease (PR2). The MM-PBSA calculation suggests that TMC-126 loses its potency against mutant variants and PR2 compared to wild-type PR1 mainly due to the loss in intermolec

A Parametric Targetability Evaluation Approach for Vitiligo Proteome Extracted through Integration of Gene Ontologies and Protein Interaction Topologies.

Vitiligo is a well-known skin disorder with complex etiology. Vitiligo pathogenesis is multifaceted with many ramifications. A computational systemic path was designed to first propose candidate disease proteins by merging properties from protein interaction networks and gene ontology terms. All in all, 109 proteins were identified and suggested to be involved in the onset of disease or its progression. Later, a composite approach was employed to prioritize vitiligo disease proteins by comparing and benchmarking the properties against standard target identification criteria. This includes sequ

Ultralow Energy Optoelectronic Synapse Using Halide Perovskite/Organic Semiconductor Heterostructure for Neuromorphic Computing, Optical Logic and Wireless Communication.

With the advancement of artificial intelligence, the emulation of biological neural processes through neuromorphic computing has gained significant attention. Artificial optoelectronic synapses have emerged as promising components for neuromorphic computing due to their simple structure, low energy consumption, and ability to overcome the von Neumann bottleneck. Here, we design a multifunctional, energy-efficient optoelectronic synapse based on a formamidinium cesium lead iodide (FAxCs1-xPbI3)/Poly(3-hexylthiophene) (P3HT) heterojunction in a two-terminal vertical structure. The synaptic devic

Spatiotemporal analysis of Crohn's disease reveals PECAM2 signaling at the basis of the inflammation-to-fibrosis transition.

BACKGROUND AND AIMS: Crohn's disease (CD) is a chronic inflammatory disease of the bowel, often complicated by fibrotic strictures, for which medical treatment is lacking, and surgery is commonly required. The mechanisms underlying the progression from chronic inflammation to fibrosis are not yet defined. We aim to unravel CD pathogenesis using a cutting-edge computational pipeline combining several available tools. METHODS: Spatial transcriptomics was performed on 13 surgical specimens, including inflamed and fibrotic CD tissues and healthy controls. The resulting spatial data were integrate

Developmental adaptation of rod photoreceptor number via photoreception in melanopsin (OPN4) retinal ganglion cells.

Photoreception, a form of sensory experience, is essential for normal development of the mammalian visual system. Detecting photons during development is a prerequisite for visual system function - from vision's first synapse at the cone pedicle and maturation of retinal vascular networks, to transcriptional establishment and maturation of cell types within the visual cortex. Consistent with this theme, we find that the lighting environment regulates developmental rod photoreceptor apoptosis via OPN4-expressing intrinsically photosensitive retinal ganglion cells (ipRGCs). Peripheral HMGB1 Downregulation Is Associated With the S100-IL-17 Axis in Parkinson's Disease: An Integrated Bulk and Single-Cell Transcriptomic Analysis.

BACKGROUND: Accumulating evidence implicates peripheral immune dysregulation in the pathogenesis of Parkinson's disease (PD). High-mobility group box 1 (HMGB1), a key alarmin released during mitochondrial stress, has been reported to be upregulated in the peripheral blood of PD patients, yet its precise role remains unclear. METHODS: We performed an integrated analysis of multiple transcriptomic datasets from peripheral blood mononuclear cells (PBMCs) of PD patients. Fecal microbiota transplantation in murine models of colitis and short bowel syndrome: lessons learned, limitations, and translational perspectives.

Fecal microbiota transplantation (FMT) has become a powerful experimental tool for dissecting microbiota-driven mechanisms in murine models of gastrointestinal and systemic disease. This review provides a comprehensive methodological and translational overview of FMT in mice, focusing on lessons learned from inflammatory bowel disease (IBD) research and emerging perspectives in short bowel syndrome (SBS). We first outline the fundamental role of the gut microbiota in immune regulation, metabolic homeostasis, and maintenance of epithelial barrier integrity, establishing the rationale for modula

PIM-1L Kinase Binds to and Inactivates SRPK1: A Biochemical and Molecular Dynamics Study.

SR/RS dipeptide repeats vary in both length and position, and are phosphorylated by SR protein kinases (SRPKs). PIM-1L, the long isoform of PIM-1 kinase, the splicing of which has been implicated in acute myeloid leukemia, contains a domain that consists largely of repeating SR/RS and SH/HS dipeptides (SR/SH-rich). In order to extend our knowledge on the specificity and cellular functions of SRPK1, here we investigate whether PIM-1L could act as substrate of SRPK1 by a combination of biochemical and computational approaches. Our biochemical data showed that the SR/SH-rich domain of PIM-1L was

Early identification of cognitive impairment and dementia: Results from four years of the community consultation center.

The community consultation center was established as the core facility for a project entitled "Community Support Network for Citizens with Mild Cognitive Impairment (MCI) and Dementia." This study reports on our center's activity and user outcomes. Users consulted with medical staff regarding their memory problems and were self-screened using a touch-panel computer assisted screening tool (TPST). Dementia was suspected when the TPST score was 12 points or below, and the Mini-Mental State Examination (MMSE) was conducted by our onsite clinical psychologists, which served as the gold standard. A dual-stream deep learning framework for continuous sign language recognition to enhance communication accessibility in the Ha'il region.

Human-Computer Interaction (HCI) depends critically on hand gesture recognition, which also provides the basis of sign language communication for those with hearing problems. In automatic sign language identification systems, however, the contribution of non-manual elements like head posture and facial emotions has sometimes been disregarded. In order to fully capture the diversity of sign language modalities and to enhance the quality of life of deaf people in the region of Ha'il, we present in this study a dual-stream convolutional neural network (TS-CNN) that separately models hand motions

Profiling lateral gene transfer events in the human microbiome using WAAFLE.

Lateral gene transfer (LGT), also known as horizontal gene transfer, facilitates genomic diversification in microbial populations. While previous work has surveyed LGT in human-associated microbial isolate genomes, the landscape of LGT arising in personal microbiomes is not well understood, as there are no widely adopted methods to characterize LGT from complex communities. Here we developed, benchmarked and validated a computational algorithm (WAAFLE or Workflow to Annotate Assemblies and Find LGT Events) to profile LGT from assembled metagenomes. WAAFLE prioritizes specificity while maintain

Graph Convolution and Self-attention Enhanced CNN with Domain Adaptation for Multi-site COVID-19 Diagnosis.

In 2019, coronavirus disease (COVID-19) is an acute disease that can rapidly develop into a very serious state. Therefore, it is of great significance to realize automatic COVID-19 diagnosis. However, due to the small difference in the characteristics of computed tomography (CT) between community acquire pneumonia (CP) and COVID-19, the existing model is unsuitable for the three-class classifications of healthy control, CP and COVID-19. The current model rarely optimizes the data from multiple centers. Therefore, we propose a diagnosis model for COVID-19 patients based on graph enhanced 3D con

Monte Carlo Dropout for Uncertainty Estimation and Motor Imagery Classification.

Motor Imagery (MI)-based Brain-Computer Interfaces (BCIs) have been widely used as an alternative communication channel to patients with severe motor disabilities, achieving high classification accuracy through machine learning techniques. Recently, deep learning techniques have spotlighted the state-of-the-art of MI-based BCIs. These techniques still lack strategies to quantify predictive uncertainty and may produce overconfident predictions. In this work, methods to enhance the performance of existing MI-based BCIs are proposed in order to obtain a more reliable system for real application s

Improving Signal-Strength Aggregation for Mobile Crowdsourcing Scenarios.

Due to its huge impact on the overall quality of service (QoS) of wireless networks, both academic and industrial research have actively focused on analyzing the received signal strength in areas of particular interest. In this paper, we propose the improvement of signal-strength aggregation with a special focus on Mobile Crowdsourcing scenarios by avoiding common issues related to the mishandling of log-scaled signal values, and by the proposal of a novel aggregation method based on interpolation. Our paper presents two clear contributions. First, we discuss the misuse of log-scaled signal-st
