Activation of STING signaling accelerates skin wound healing.

BACKGROUND: The process of repair after skin injury is precisely regulated by a variety of mediators such as cytokines and chemokines. Recent reports demonstrated that cytoplasmic DNA-sensor cyclic GMP-AMP synthase (cGAS) activates the stimulator of interferon genes (STING) via production of cyclic GMP-AMP (cGAMP) and subsequently induces inflammatory cytokines, including type I interferon (IFN). OBJECTIVE: We examined whether activation of the STING pathway by cGAMP affects the process of skin wound repair. METHODS: The skin wound repair model was established using wild-type (WT) mice. Two …

Brucella abortus Cyclic Dinucleotides Trigger STING-Dependent Unfolded Protein Response That Favors Bacterial Replication.

Brucella abortus is a facultative intracellular bacterium that causes brucellosis, a prevalent zoonosis that leads to abortion and infertility in cattle, and undulant fever, debilitating arthritis, endocarditis, and meningitis in humans. Signaling pathways triggered by B. abortus involves stimulator of IFN genes (STING), which leads to production of type I IFNs. In this study, we evaluated the pathway linking the unfolded protein response (UPR) and the endoplasmic reticulum-resident transmembrane molecule STING, during B. abortus infection. We demonstrated that B. abortus infection induces the

Loss of relaxations, metabolic failure and increased calcium permeability of smooth muscle during chronic cerebral vasospasm.

In the canine basilar artery during chronic vasospasm following subarachnoid hemorrhage, endothelium-dependent relaxations were diminished. Release of endothelium-derived relaxing factor (EDRF), as measured by a bioassay method, was unchanged. Relaxation to nitric oxide (NO) in preparations without endothelium was smaller in the spastic arteries. Production of cyclic GMP, measured by radioimmunoassay, was reduced in the spastic arteries; the impaired production was accompanied by decrease in GTP, the substrate for the production of cyclic nucleotide. The contents of other high-energy phosphate…

Anemia and splenomegaly in cGKI-deficient mice.

To explore the functional significance of cGMP-dependent protein kinase type I (cGKI) in the regulation of erythrocyte survival, gene-targeted mice lacking cGKI were compared with their control littermates. By the age of 10 weeks, cGKI-deficient mice exhibited pronounced anemia and splenomegaly. Compared with control mice, the cGKI mutants had significantly lower red blood cell count, packed cell volume, and hemoglobin concentration. Anemia was associated with a higher reticulocyte number and an increase of plasma erythropoietin concentration. The spleens of cGKI mutant mice were massively enl

Effects of cyclic nucleotide phosphodiesterase IV inhibitor, Ro20,1724, on pancreatic exocrine secretion in dog.

1. The effects of the cyclic nucleotide phosphodiesterase (PDE) inhibitors, Ro20,1724, 3-isobutyl-1-methylxanthine (IBMX), trifluoperazine ( … and IBMX were additive. 5. Ro20,1724 and IBMX increased cyclic AMP concentration but did not affect cyclic GMP concentration. 6. These results suggest that Ro20,1724 and IBMX have secretory properties on pancreatic exocrine glands of the dog, which may be mediated through an increase in cyclic AMP subsequent to inhibition of PDE activity. Furthermore, pancre …

Effect of some beta-adrenergic blocking agents on tissue guanylate cyclase and cyclic nucleotides in the rat.

Pretreatment of rats with the beta-adrenergic blocking agents, atenolol, practolol, pronethalol and propranolol, at a dosage of 150 mg/kg/day for 5 days, produced marked increases in guanylate cyclase activity in the liver, gastric and intestinal mucosae, but with concomitant decreases in cyclic GMP levels. The ratios of cAMP/cGMP in each of the tissues were not changed significantly from control values after pretreatment with these drugs. These observations indicate that any oncogenicity seen with beta-blocking agents is not due to the common pharmacological action of beta-adrenoreceptor bloc…

Physiological calcium ion concentration and a cholinergic drug on activity of particulate guanylate cyclase in guinea pig taenia caecum.

The microsomal fraction derived from the taenia caecum of guinea pigs was used as particulate guanylate cyclase preparation. The guanylate cyclase activity was not influenced by butyltrimethylammonium bromide and Ca2+ (4.0 to 24 X 10-7 M). The results suggest that the particulate guanylate cyclase is not activated by the drug-receptor interaction and also by the increase of intracellular Ca2+ concentration. Therefore, the increase of tissue level of cyclic GMP brought about in the guinea pig taenia caecum might be due to activation of soluble guanylate cyclase following the increase in the int…

Plasmodium falciparum Guanylyl Cyclase-Alpha and the Activity of Its Appended P4-ATPase Domain Are Essential for cGMP Synthesis and Blood-Stage Egress.

Guanylyl cyclases (GCs) synthesize cyclic GMP (cGMP) and, together with cyclic nucleotide phosphodiesterases, are responsible for regulating levels of this intracellular messenger which mediates myriad functions across eukaryotes. In malaria parasites (Plasmodium spp), as well as their apicomplexan and ciliate relatives, GCs are associated with a P4-ATPase-like domain in a unique bifunctional configuration. P4-ATPases generate membrane bilayer lipid asymmetry by translocating phospholipids from the outer to the inner leaflet. Here, we investigate the role of Plasmodium falciparum guanylyl cycl…

Antibiofilm Peptides Revisited: From Biofilm Models and Metrics to Mechanisms and Translation.

Biofilm-associated infections contribute substantially to the antimicrobial resistance-related burden by promoting persistent, difficult-to-treat infections. Antibiotic failure often reflects biofilm recalcitrance (tolerance and persistence) and, in some contexts, the enhanced selection and dissemination of resistance. To overcome this burden, antibiofilm peptides (ABPs) are emerging as a versatile solution. ABPs can inhibit biofilm development, weaken established biofilms, and synergize with existing antimicrobials. This review highlights current biofilm models and susceptibility end points,

Enhancement of neutrophil chemotaxis and alteration of levels of cellular cyclic nucleotides by levamisole.

Levamisole, an antihelminthic agent reported to enhance nonspecifically various parameters of the immune response, was examined for its effect on chemotaxis of human neutrophils and on levels of cellular cyclic nucleotides. This agent was found, in most instances, to enhance chemotactic responses of neutrophils to a bacterial chemotactic factor derived from Escherichia coli. At similar concentrations, levamisole produced increases in levels of guanosine 3':5'-cyclic phosphate in neutrophils. In contrast, a decrease in concentrations of adenosine 3':5'-cyclic phosphate was observed when neutrop

The NO/cGMP/PKG pathway in platelets: The therapeutic potential of PDE5 inhibitors in platelet disorders.

Platelets are the "guardians" of the blood circulatory system. At sites of vessel injury, they ensure hemostasis and promote immunity and vessel repair. However, their uncontrolled activation is one of the main drivers of thrombosis. To keep circulating platelets in a quiescent state, the endothelium releases platelet antagonists including nitric oxide (NO) that acts by stimulating the intracellular receptor guanylyl cyclase (GC). The latter produces the second messenger cyclic guanosine-3',5'-monophosphate (cGMP) that inhibits platelet activation by stimulating protein kinase G, which phospho

TPx Protein of Cysticercus cellulosae Regulates Macrophage M2 Polarization via the cGMP-PKG Signaling Pathway.

Cysticercosis, caused by the larval stage of Taenia solium (Cysticercus cellulosae), is a neglected tropical disease threatening public health. Thioredoxin peroxidase (TPx) is a key antioxidant protein secreted by the parasite, but its role in macrophage polarization remains unclear. In this study, THP-1-derived macrophages were treated with TPx protein for 24 h and 48 h. Flow cytometry, reverse transcription-quantitative polymerase chain reaction (RT-qPCR), and Western blot were employed to assess reactive oxygen species (ROS) levels, M1/M2 cell proportions, mRNA expression of tumor necrosis

Activation of HIV-1 expression and replication by cGMP dependent protein kinase type 1-beta (PKG1beta).

The effect of cGMP (cyclic GMP) dependent protein kinase 1-beta (PKG1-beta) and cGMP analogues on transcriptional activity and replication of human immunodeficiency virus type 1 (HIV-1) was investigated. Transfection of PKG1beta expression plasmid increased expression from an HIV-1 LTR-reporter as well as from an infectious HIV-1 molecular clone, pNL4-3. Treatment of HIV-1 AD8-infected monocyte derived macrophages (MDMs) with cGMP agonists and cGMP antagonists caused respectively increased and decreased virus replication. These findings provide evidence that cGMP and PKG serve to regulate HIV-…

A divergent cyclic nucleotide binding protein promotes Plasmodium ookinete infection of the mosquito.

Colonisation of mosquitos by the malarial parasite is critically reliant on the invasive ookinete stage. Ookinete invasion of mosquito is coordinated by the apical complex, a specialised parasite structure containing components for secretion, attachment and penetration. While studies have investigated cytoskeletal and secretory elements, it is currently unknown if signalling modules are present or functional at the apical complex. Here we elucidate the role of a cryptic cyclic nucleotide-binding protein which we name CBP-O. PbCBP-O showed a marked localisation to the ookinete apex and disrupti

[The content of cyclic nucleotides in the blood of patients with chronic type-A and -B gastritis].

A study of the content of cyclic nucleotides in patients with chronic gastritis indicates that the level of cAMP and cGMP undergoes various changes. The cGMP/cAMP ratio was in gastritis of the A type higher than in the healthy and in the B gastritis type lower. It is concluded that chronic gastritis is accompanied not only by changes of absolute values of the level of cyclic nucleotides but also by changes of the cGMP/cAMP ratio.
