MLN4924, neddylation inhibitor suppresses hypoxia induced retinal angiogenesis by targeting Human Antigen R signaling.

PURPOSE: Retinal hypoxia is a key pathological stimulus for neovascularization, leading to abnormal proliferation of blood vessels and vascular endothelial dysfunction leading to vision threatening conditions. The anti-angiogenic potential of MLN4924, a specific inhibitor of neddylation signaling has been evidenced in cancer cells, but remains abstract as therapy for ocular angiogenesis in normal retinal cells. The current work intended to delineate a novel molecular signaling cascade of combating retinal angiogenesis by inhibiting the neddylation-Human Antigen R (HuR) signaling pathway using

Defense Responses Stimulated by Bacillus subtilis NCD-2 Through Salicylate- and Jasmonate-Dependent Signaling Pathways Protect Cotton Against Verticillium Wilt.

Bacillus subtilis NCD-2 demonstrates exceptional biocontrol potential against cotton Verticillium wilt. While previous studies have established its direct antifungal activity (e.g., inhibiting Verticillium dahliae mycelial growth and spore germination), our work reveals a novel mechanism: NCD-2 primes systemic resistance in cotton by activating plant immune-signaling pathways. Firstly, transcriptional profiling uncovered that NCD-2 triggers a defense response in roots analogous to V. dahliae infection, allowing cotton to maintain a more balanced state when confronted with pathogen attacks. A herbivore-induced salicylic acid carboxyl methyl transferase produces methyl salicylate in tomato to mediate defense signaling and deter pests.

Methyl salicylate (MeSA) is a component of the herbivore-induced volatile blend of tomato (Solanum lycopersicum), and an intermediate in long distance salicylic acid (SA) signaling. To investigate the role of MeSA in interactions between tomato and the lepidopteran pests Helicoverpa zea and Manduca sexta, we used transgenic tomato lines that were antisense (AS) or overexpressors (OE) of the tomato SA carboxyl methyl transferase SlSAMT1, which under- or over-produce MeSA, respectively. Transgenic AS plants produced less MeSA and were more susceptible to both insects compared to wild-type plants

Graphene oxide enhances aphid resistance in sorghum via the miR319-SbTCP7-SbLOX3 Pathway.

The aphid (Melanaphis sacchari) has emerged as a formidable pest, devastating sorghum plants and highlighting the need for sustainable management strategies. Graphene oxide (GO), as a novel material, has garnered attention for its use in crop cultivation and management, but its effects on biotic stresses remain elusive. Here, we used 10 mg/L GO to spray aphid-stressed sorghum seedlings four times in total. GO exposure reduced 50% H2O2 from the reactive oxygen species (ROS) burst induced by the aphid. Further analysis revealed that GO within the cells acts as a nanozyme, mimicking and enhancing

Overcoming paclitaxel resistance in ovarian cancer cells using DN200434, an inverse agonist of estrogen-related receptor gamma (ERRγ).

Ovarian cancer is one of the most prevalent gynecological malignancies in women. Resistance towards frontline chemotherapy, paclitaxel is a major obstacle in the management of cancer. This intricate phenomenon reduces the treatment efficacy, leading to poor patient prognosis. Recently, the nuclear receptor estrogen-related receptor γ (ERRγ) has been implicated in cancer progression and metastasis. In particular, the receptor is reported to be overexpressed in ovarian cancer, and its overexpression is associated with reduced overall survival. However, the effect of ERRγ targeting using the sele

Transcriptome profiling implicates ANNEXIN1 in light sensitivity and hormonal balance maintenance in Arabidopsisthaliana.

Annexins are a conserved family of calcium- and phospholipid-binding proteins. Despite their ubiquity and ancient origin, their physiological functions are not fully understood. Numerous studies have shown that the ectopic expression of some annexins can ameliorate the adverse effects of numerous abiotic stresses through modulation of reactive oxygen species accumulation and/or abscisic acid signaling. To gain further insight into the molecular mechanisms of this activity, we studied transgenic Arabidopsis thaliana (Arabidopsis) plants with increased expression of ANNEXIN1 (ATANN1) in the wild

Balancing Growth and Defense: Nanoselenium and Melatonin in Tea (Camellia sinensis) Protection against Glufosinate.

Current crop stress resistance research suggests that the prominent stimulants nanoselenium (NSe) and melatonin (MT) might improve tea safety, quality, and stress resistance induced by the widely used nonselective herbicide glufosinate (GLU). Their biofortification effects on tea growth, antioxidant activity, and secondary metabolism pathways response to GLU remain unclear. Here, NSe, MT, and their combination NSe-MT effectively reduced 26.6-50.9% GLU and its metabolites in tea seedlings, balanced the photosystem, enhanced antioxidant defenses, and optimized reactive oxygen species scavenging

Lyso-phosphatidylethanolamine primes the plant immune system and promotes basal resistance against hemibiotrophic pathogens.

BACKGROUND: Lyso-phosphatidylethanolamine (LPE) is a natural phospholipid that functions in the early stages of plant senescence. Plant innate immunity and early leaf senescence share molecular components. To reveal conserved mechanisms that link-up both processes, we tried to unravel to what extent LPE coordinates defense response and by what mode of action. RESULT: We found that LPE-treatment induces signaling and biosynthesis gene expression of the defensive hormone salicylic acid (SA). However, jasmonic acid and ethylene triggered gene induction levels are indistinguishable from the contr

Virus infection induces resistance to Pseudomonas syringae and to drought in both compatible and incompatible bacteria-host interactions, which are compromised under conditions of elevated temperature and CO2 levels.

Plants are simultaneously exposed to a variety of biotic and abiotic stresses, such as infections by viruses and bacteria, or drought. This study aimed to improve our understanding of interactions between viral and bacterial pathogens and the environment in the incompatible host Nicotiana benthamiana and the susceptible host Arabidopsis thaliana, and the contribution of viral virulence proteins to these responses. Infection by the Potato virus X (PVX)/Plum pox virus (PPV) pathosystem induced resistance to Pseudomonas syringae (Pst) and to drought in both compatible and incompatible bacteria-ho

Involvement of jasmonic acid, ethylene and salicylic acid signaling pathways behind the systemic resistance induced by Trichoderma longibrachiatum H9 in cucumber.

BACKGROUND: Trichoderma spp. are effective biocontrol agents for many plant pathogens, thus the mechanism of Trichoderma-induced plant resistance is not fully understood. In this study, a novel Trichoderma strain was identified, which could promote plant growth and reduce the disease index of gray mold caused by Botrytis cinerea in cucumber. To assess the impact of Trichoderma inoculation on the plant response, a multi-omics approach was performed in the Trichoderma-inoculated cucumber plants through the analyses of the plant transcriptome, proteome, and phytohormone content. RESULTS: A novel

Postharvest treatments with γ-aminobutyric acid, methyl jasmonate, or methyl salicylate enhance chilling tolerance of blood orange fruit at prolonged cold storage.

BACKGROUND: Blood orange is sensitive to chilling injury (CI) depending on cultivar and storage temperature. Postharvest treatments with γ-aminobutyric acid (GABA), methyl jasmonate (MeJA), or methyl salicylate (MeSA) are known to alleviate CI. γ-Aminobutyric acid aqueous solution, applied at 20 and 40 mM, was vacuum-infiltrated at 30 kPa for 8 min at 20 °C. Methyl jasmonate or MeSA vapor treatments were applied separately at 50 and 100 μM by putting the fruit in 20 L plastic containers for 18 h at 20 °C. There have been no reports about postharvest treatments of GABA, MeJA, or MeSA on enhanci

A virus plays a role in partially suppressing plant defenses induced by the viruliferous vectors.

Herbivorous attack induces plant defenses. There is evidence that some pests suppress these defenses by interfering with signaling pathways. We here report that infestation by the white-backed planthopper, Sogatella furcifera, induces defense responses in rice and infection of the southern rice black-streaked dwarf virus in the planthoppers partially suppresses the planthopper-induced plant defenses. Salicylic acid (SA) levels generally showed a temporal increase pattern while jasmonic acid (JA) levels generally exhibited a decrease pattern in the planthopper-infested plants, irrespective of v

Antioxidant activity and induction of mechanisms of resistance to stresses related to the inoculation with Azospirillum brasilense.

We investigated the effects of Azospirillum brasilense strains Ab-V5 and Ab-V6 in the induction of mechanisms of systemic acquired resistance (SAR) and induced system resistance (ISR) on maize (Zea mays L.) plants. Under normal growth conditions, the treatments consisted of the standard inoculation of cells at sowing, and leaf spray of cells or their metabolites at the V2.5 growth stage; under saline stress (170 mM NaCl), the treatment consisted of standard single and co-inoculation of A. brasilense and Rhizobium tropici. The main compounds in the Azospirillum metabolites were identified as in

Role of Epichloë Endophytes in Defense Responses of Cool-Season Grasses to Pathogens: A Review.

Various cool-season grasses are infected by Epichloë endophyte, and this symbiotic relationship is always of benefit to the host grass due to an increased resistance to abiotic and biotic stresses. Fungal diseases adversely affect the yield, quality, and economic benefits of rangelands, which affects the production of animal husbandry. Therefore, it is imperative to breed resistant cultivars and to better understand the role of fungal endophytes in order to protect grasses against pathogens. The present review introduces research regarding how these endophytes affect the growth of pathogens in

Treatment of Vinca minor Leaves with Methyl Jasmonate Extensively Alters the Pattern and Composition of Indole Alkaloids.

Alkaloids extracted from mature Vinca minor leaves were fractionated by preparative HPLC. By means of HRMS and NMR data, the main alkaloids were identified as vincamine, strictamine, 10-hydroxycathofoline, and vincadifformine. Upon treatment with methyl jasmonate (MeJA), the pattern and composition of the indole alkaloids changed extensively. While 10-hydroxycathofoline and strictamine concentrations remained unaltered, vincamine and vincadifformine levels showed a dramatic reduction. Upon MeJA treatment, four other indole alkaloids were detected in high quantities. Three of these alkaloids ha

Evidence for salicylic acid signalling and histological changes in the defence response of Eucalyptus grandis to Chrysoporthe austroafricana.

Eucalyptus species are cultivated for forestry and are of economic importance. The fungal stem canker pathogen Chrysoporthe austroafricana causes disease of varying severity on E. grandis. The Eucalyptus grandis-Chrysoporthe austroafricana interaction has been established as a model system for studying Eucalyptus antifungal defence. Previous studies revealed that the phytohormone salicylic acid (SA) affects the levels of resistance in highly susceptible (ZG14) and moderately resistant (TAG5) clones. The aims of this study were to examine histochemical changes in response to wounding and inocul

Rhizosphere-associated Pseudomonas induce systemic resistance to herbivores at the cost of susceptibility to bacterial pathogens.

Plant-associated soil microbes are important mediators of plant defence responses to diverse above-ground pathogen and insect challengers. For example, closely related strains of beneficial rhizosphere Pseudomonas spp. can induce systemic resistance (ISR), systemic susceptibility (ISS) or neither against the bacterial foliar pathogen Pseudomonas syringae pv. tomato DC3000 (Pto DC3000). Using a model system composed of root-associated Pseudomonas spp. strains, the foliar pathogen Pto DC3000 and the herbivore Trichoplusia ni (cabbage looper), we found that rhizosphere-associated Pseudomonas spp.

Exogenous Abscisic Acid and Gibberellic Acid Elicit Opposing Effects on Fusarium graminearum Infection in Wheat.

Although the roles of salicylate (SA) and jasmonic acid (JA) have been well-characterized in Fusarium head blight (FHB)-infected cereals, the roles of other phytohormones remain more ambiguous. Here, the association between an array of phytohormones and FHB pathogenesis in wheat is investigated. Comprehensive profiling of endogenous hormones demonstrated altered cytokinin, gibberellic acid (GA), and JA metabolism in a FHB-resistant cultivar, whereas challenge by Fusarium graminearum increased abscisic acid (ABA), JA, and SA in both FHB-susceptible and -resistant cultivars.
