Safe and efficient drug delivery system with liposomes for intrathecal application of an antivasospastic drug, fasudil.

Pharmacological treatment for cerebral ischemia and cerebral vasospasm following subarachnoid hemorrhage (SAH) cannot attain sufficiently high concentrations of the drugs in the cerebrospinal fluid (CSF) without precipitating systemic side effects. We recently developed a liposomal drug delivery system for intrathecal application that can maintain effective concentrations of cerebral vasodilator, fasudil, in the CSF. A single intrathecal injection of liposomal fasudil could maintain a therapeutic drug concentration in the CSF over a period time due to their sustained-release property, signific

Microemulsion-based drug delivery system for transnasal delivery of Carbamazepine: preliminary brain-targeting study.

This study reports the development and evaluation of Carbamazepine (CMP)-loaded microemulsions (CMPME) for intranasal delivery in the treatment of epilepsy. The CMPME was prepared by the spontaneous emulsification method and characterized for physicochemical parameters. All formulations were radiolabeled with (99m)Tc (technetium) and biodistribution of CMP in the brain was investigated using Swiss albino rats. Brain scintigraphy imaging in rats was also performed to determine the uptake of the CMP into the brain. CMPME were found crystal clear and stable with average globule size of 34.11 ± 1.

Photo-Empowered Macrophage-Based Drug Delivery System Overcomes Motility Suppression and Significantly Enhances Deep Tumor Drug Delivery.

Recently, cell-based drug delivery systems emerge as highly promising alternatives in the field of antitumor therapy. These systems are characterized by their excellent biocompatibility, robust drug-loading capacity, and the ability to effectively traverse biological barriers. However, the immunosuppressive tumor microenvironment significantly weakens the motility of immune cells, which is crucial for efficient drug delivery. A novel photo-empowered macrophage-based (PEM) drug delivery system is successfully developed in this study. The system is modified with thylakoids extracted from spinach…

Polysaccharide-based drug conjugates and related delivery systems for Alzheimer's disease: from molecular design to translational challenges.

Alzheimer's disease (AD) presents distinct extracellular, intracellular and vascular delivery targets, yet brain exposure of candidate therapeutics remains difficult to establish. This narrative review compares hyaluronic acid, chitosan, cyclodextrin, dextran, alginate and pectin systems by construct architecture, route, blood-brain barrier (BBB) evidence, cargo release and biological response. The literature includes covalent drug conjugates, loaded or coated carriers, host-guest complexes and intrinsically active polymers; evidence from one category cannot validate another. Clinical programs

In Vitro-In Vivo Correlation (IVIVC) in Transdermal Drug Delivery Systems: Models, Regulatory Perspectives and Translational Challenges.

Transdermal drug delivery systems (TDDS) represent a clinically established yet mechanistically complex platform for systemic drug administration, offering advantages such as avoidance of hepatic first-pass metabolism, sustained drug input and improved patient adherence. However, their development and regulatory evaluation remain challenging due to the physiological variability of the skin barrier, site-dependent absorption and formulation-skin interactions. In vitro-in vivo correlation (IVIVC) has therefore emerged as a critical translational framework for linking in vitro permeation characte…

Multiparticulate Drug Delivery Systems Preserve the Efficacy of Reduced-Dose Intermittent Benznidazole Therapy in Experimental Chagas Disease.

Chagas disease, caused by the Trypanosoma cruzi parasite infection, is primarily treated with benznidazole (BZ), though continuous dosing regimens are limited by adverse effects that compromise adherence. We evaluated whether a reduced-dose intermittent BZ regimen (50 mg/kg every 2 days, 20 doses), administered as free drug or incorporated into a BZ-loaded multiparticulate drug delivery system (BZ-MDDS), with or without adjunctive CD73 inhibition (APCP), maintains therapeutic efficacy and improves tissue-level safety in an experimental model of infection. Smart thyroid-regulated drug delivery systems: a review of intelligent biosensor-integrated therapeutics for precision thyroxine replacement.

Levothyroxine (L-T4) replacement therapy has remained the standard treatment for post-thyroidectomy hypothyroidism since the mid-twentieth century. The drug works, but the delivery method has a basic flaw: it cannot adjust. A patient swallows the same dose every morning regardless of whether she is pregnant, fighting a fever, fasting, or sleeping. The healthy thyroid gland, by contrast, constantly tunes its output in response to signals from the hypothalamic-pituitary-thyroid (HPT) axis, adjusting on a timescale of minutes to hours. This mismatch between static pills and dynamic physiology lea
