Selection and characterization of nuclear mutations affecting mitochondria in Paramecium.

A screening method, based upon resistance to a tetrazolium salt (TTC), is described which permitted the isolation in Paramecium of 28 mutants resistant to TTC. These mutants displayed various defects in mitochondrial functions (cytochromic content cyanide insensitive respiration). Some mutations seemed to affect directly the respiration chain while others seemed to cause indirect modifications, possibly altering mitochondrial protein synthesis. Genetic analysis of four mutants showed in all that the resistance to TTC was of nuclear origin.

[Histochemical characteristics of the activity of succinate dehydrogenase and cytochrome oxidase in certain tissues and organs of patients with thyrotoxicosis and animals with experimental thyroidin toxicosis].

The activity of succinic dehydrogenase and cytochromoxidase was studied histochemically in the peripheral blood leukocytes, the thyroid gland tissue and the skeletal muscles of the patients suffering from thyrotoxicosis. The activity of these enzymes is also determined in the blood leukocytes, the cardiac muscle, the liver and the skeletal muscles of the experimental animals given thyroidin in a dose of 0.1 g per 100 g of body weight in the course of 10 days. A marked elevation of the activity of the oxidative enzymes in the patients with an increase of the severity of thyrotoxicosis and in th

Molecular Characterization of Mosquito Species and Confirmation of First Record of Aedes aegypti in Buraydah City, Kingdom of Saudi Arabia.

Mosquitoes are a source of concern because they transmit many infectious diseases, including Zika virus, chikungunya, and malaria. Accurate and rapid identification of mosquitoes is essential for disease surveillance and control. Genetic diversity in mosquito species, resulting from adaptation to different environments, leads to many differences in morphological characteristics. Traditional identification methods relying on morphology can be time-consuming and unreliable. The study aimed to identify medically and veterinary significant adult mosquito species throughout 2021 in Buraydah City, K

Colonisation history of a vector species of veterinary interest: phylogeography of Culicoides imicola (Diptera: Ceratopogonidae) in the Indian Ocean revealing low genetic structure.

Culicoides imicola (Diptera: Ceratopogonidae) is the main historical vector of livestock viruses such as bluetongue virus (BTV), Epizootic Haemorrhagic Disease virus (EHDV) and African Horse Sickness virus (AHSV), in the Afrotropical region. Despite its wide distribution across Africa, the Mediterranean basin and parts of Asia, the genetic structure and colonisation history of this species in the South-West Indian Ocean (SWIO) territories remain poorly understood. The aim of this study was to investigate how passive wind-driven dispersal and recent animal movements associated with human coloni

Molecular and morphological characterization of Oxyspirura conjunctivalis (Spirurida: Thelaziidae) in a captive gibbon with evidence of cockroach intermediate hosts.

Eyeworms of the genus Oxyspirura infect a wide range of avian hosts worldwide, with more than 50 species described. However, Oxyspirura conjunctivalis (syn. O. youngi) and the recently described O. tamarina have been reported exclusively from captive primates in zoological gardens in Europe and North America. In this study, we report the first record of Oxyspirura infection in a captive Bornean white-bearded gibbon (Hylobates albibarbis) in Japan. Five adult worms recovered from the conjunctival sac measured 7.8-11.1 mm in length and 0.29-0.35 mm in width and exhibited a bipartite buccal capsu

First record of the invasive American tapeworm Ophiotaenia saphena (Cestoda: Proteocephalidae) in the Tokyo daruma pond frog Pelophylax porosus porosus in Japan.

Adult proteocephalid tapeworms of the genus Ophiotaenia (Cestoda: Proteocephalidae) were recovered from the endemic Japanese amphibian species, Pelophylax porosus porosus (Amphibia: Ranidae) from Tochigi, Japan. As a result of molecular analyses based on the 28S ribosomal RNA and cytochrome c oxidase subunit I genes, these worms were identified as Ophiotaenia saphena, a species native to North America. Their morphological features were also consistent with those of O. saphena. This species is presumed to have been introduced into Japan with the American bullfrog, Lithobates catesbeianus (Ranid

Parachrisomon debonoae n. sp. (Digenea: Monorchiidae), a new species with significant intraspecific morphological variation infecting Mullidae (Mulliformes) from the eastern and western coasts of Australia.

We report trematodes conforming to the generic concept of Parachrisomon Madhavi, 2008, infecting multiple species of goatfishes (Mullidae) from Moreton Bay (south-eastern Queensland) and off Lizard Island (northern Great Barrier Reef) on the eastern coast of Australia, and from Ningaloo Reef on the western coast. Initial morphological analyses suggested the possible presence of multiple species; however, no morphological distinctions proved consistent. Molecular analyses demonstrate a distinction between western and eastern Australian samples, especially in mt cox1 sequence data, but the diffe

Amended diagnosis of the genus Provitellotrema (Digenea: Haplosplanchnidae) with description of a new species from vietnamese mullets.

Morphological and molecular data for a new representative of the genus Provitellotrema (Haplosplanchnidae) from the mullets of the coastal waters of Vietnam were provided in the present study. Morphologically, the trematodes described here are similar to representatives of Haplosplanchnus and Pseudohaplosplanchnus and demonstrate the most closeness to Haplosplanchnus purii by most of the metric parameters. Results of molecular phylogenetic analysis show that the trematodes we found were closely related to Provitellotrema crenimugilis, and H. purii was in the same clade. Genetic distance values

First record of Brachylaima asakawai (Trematoda: Brachylaimidae) in a carnivore host, the red fox (Vulpes vulpes schrencki).

Accumulating information on the relationship between hosts and parasites is important to understand their life cycle. Nine Brachylaima spp. species exist in Japan, but their life cycles are not fully understood, partly because of their low prevalence in their definitive hosts. In this study, 23 trematodes, thought to belong to the genus Brachylyma were found in the small intestine of the red fox (Vulpes vulpes schrencki) in Hokkaido, Japan. We analyzed these trematodes using morphological and molecular methods. Morphological analysis showed that the trematodes had the same features as Brachyla

Genetic structure and evolutionary dynamics of Babesia gibsoni populations: a cytochrome oxidase subunit I (COXI) gene-based study.

BACKGROUND: Babesia gibsoni is a tick-borne intraerythrocytic protozoan parasite causing piroplasmosis in dogs globally. A significant knowledge gap exists regarding the genetic characterization of B. gibsoni using the mitochondrial cytochrome c oxidase subunit I (COXI) gene. To address this, the current study aimed to investigate the genetic diversity and population genetics of B. gibsoni based on COXI gene sequences (≥ 649 bp) in the GenBank. RESULTS: Phylogenetic analysis revealed that all B. gibsoni isolates clustered into a single large monophyletic clade based on nucleotide and amino ac

First report and molecular characterisation of an adult liver fluke (Fasciola hepatica) in a brown bear (Ursus arctos) in Türkiye.

Fascioliasis, caused by the parasite Fasciola hepatica, is a worldwide zoonotic disease that can have serious consequences for livestock, certain wild animals and humans. This study was conducted to morphologically and molecularly characterise a F. hepatica isolate from a brown bear. After examination of the internal organs, a Fasciola sp. isolate was obtained from the bile ducts of the liver. The adult parasite was morphologically analysed under a stereomicroscope and identified as F. hepatica. Measurements of body length, body width and ventral sucker area were then recorded. After isolation

Is Type 2 Diabetes a Primary Mitochondrial Disorder?

Diabetes mellitus is the most common endocrine disturbance in inherited mitochondrial diseases. It is essential to increase awareness of the correct diagnosis and treatment of diabetes in these patients and screen for the condition in family members, as diabetes might appear with distinctive clinical features, complications and at different ages of onset. The severity of mitochondrial-related diabetes is likely to manifest on a large scale of phenotypes depending on the location of the mutation and whether the number of affected mitochondria copies (heteroplasmy) reaches a critical threshold.

Photosynthetic microbial fuel cells for methanol treatment using graphene electrodes.

Photosynthetic microbial fuel cells (pMFC) represent a promising approach for treating methanol (CH3OH) wastewater. However, their use is constrained by a lack of knowledge on the extracellular electron transfer capabilities of photosynthetic methylotrophs, especially when coupled with metal electrodes. This study assessed the CH3OH oxidation capabilities of Rhodobacter sphaeroides 2.4.1 in two-compartment pMFCs. A 3D nickel (Ni) foam modified with plasma-grown graphene (Gr) was used as an anode, nitrate mineral salts media (NMS) supplemented with 0.1% CH3OH as anolyte, carbon brush as cathode

Vibrio cholerae requires oxidative respiration through the bd-I and cbb3 oxidases for intestinal proliferation.

Vibrio cholerae respires both aerobically and anaerobically and, while oxygen may be available to it during infection, other terminal electron acceptors are proposed for population expansion during infection. Unlike gastrointestinal pathogens that stimulate significant inflammation leading to elevated levels of oxygen or alternative terminal electron acceptors, V. cholerae infections are not understood to induce a notable inflammatory response. To ascertain the respiration requirements of V. cholerae during infection, we used Multiplex Genome Editing by Natural Transformation (MuGENT) to creat

IAPV-Induced Paralytic Symptoms Associated with Tachypnea via Impaired Tracheal System Function.

Although it had been reported that Israeli acute paralysis virus (IAPV) can cause systemic infection in honey bees, little is known about how it establishes this infection and results in the typical symptoms, paralysis and trembling. Here, we used our previously constructed IAPV infectious clone to investigate viral loads in different tissues of honey bees and further identify the relation between tissue tropism and paralytic symptoms. Our results showed that tracheae showed a greater concentration of viral abundance than other tissues. The abundance of viral protein in the tracheae was positi
